| 2 | Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12 |
| 3 | Stimulation impact of electric currents on heterotrophic denitrifying microbial viability and denitrification performance in high concentration nitrate-contaminated wastewater显示文摘Electric current stimulation has been shown to have a positive influence on heterotrophic denitrifying microbial viability and has the potential to improve wastewater denitrification performance. This study investigated the effects of varying current densities on microbial activity and NO_3^- removal efficiency under heterotrophic conditions.NO_3^-removal rate was highest at an applied current density of 400 mA/m^2. However, the optimum removal efficiency of total inorganic nitrogen(TIN; 99%) was achieved when the current density was fixed at 200 m A/m^2. Accumulation of NH_4^+-N and NO_2^--N byproducts were also minimized at this current density. The activity of heterotrophic denitrifying microorganisms was much higher at both 200 and 400 mA/m^2. Moreover, the average adenosine-5′-triphosphate(ATP)content(an indicator of cell metabolism) at a current density of 1600 mA/m^2 was lower than that under no current, indicating heterotrophic denitrifying microbial activity can be inhibited at high current densities. Hence, direct electrical stimulation on the activity of heterotrophic denitrifying microorganisms in the developed system should be lower than 1600 mA/m^2. | 4 | Removal of trimethoprim and sulfamethoxazole in artificial composite soil treatment systems and diversity of microbial communities显示文摘Four artificial composite soil treatment systems(ACSTs)fed with reclaimed water containing trimethoprim(TMP)and sulfamethoxazole(SMX)were constructed to investigate SMX and TMP biodegradation efficiency,ammonia and nitrite removal conditions and the microbial community within ACST layers.Results showed SMX and TMP removal rates could reach 80% and 95%,respectively,and removal rates of ammonia and nitrite could reach 80% and 90%,respectively,in ACSTs.The MiSeq sequencing results showed that microbial community structures of the ACSTs were similar.The dominant microbial community in the adsorption and biodegradation layers of the ACSTs contained Proteobacteria,Chloroflexi,Acidobacteria,Firmicutes,Actinobacteria and Nitrospirae.Firmicutes and Proteobacteria were considerably dominant in the ACST biodegradation layers.The entire experimental results indicated that Nitrosomonadaceae_uncultured,Nitrospira and Bacillus were associated with nitrification processes,while Bacillus and Lactococcus were associated with SMX and TMP removal processes.The findings suggest that ACSTs are appropriate for engineering applications. | Qinqin Liu Miao Li Rui Liu Quan Zhang Di Wu Danni Zhu Xuhui Shen Chuanping Feng Fawang Zhang Xiang Liu | 2019 | Frontiers of Environmental Science & Engineering2019,13,2: | 2 |
