1A novel method for purification of recombinant adeno-associated virus vectors on a large scale显示文摘A novel method for recombinant adeno-associ-ated virus (rAAV) purification on large scale is described. The method involves three steps, including chloroform treatment, PEG/NaCl precipitation and chloroform extraction. The whole procedure can be performed in four hours. Using this purification method, we can reproducibly obtain, from 4 × 109 of proviral cells cultured in roller bottles, purified rAAV-GFP stocks with titers of around 5×1013 particles/mL and purity greater than 95%. The infectious titers of the vector stocks were up to 2×1012 TU/mL, thus particle-to-infectivity rate was about 25. Under an electronic microscope, most rAAV particles appeared full and a few were in intermediate form. Empty particles were rarely seen. The purified rAAV-GFP stocks have been successfully used in in vitro and in vivo transfection experiments. 2Research progress of disinfection and disinfection by-products in China显示文摘Disinfection is an indispensable water treatment process for killing harmful pathogens and protecting human health. However, the disinfection has caused significant public concern due to the formation of toxic disinfection by-products(DBPs). Lots of studies on disinfection and DBPs have been performed in the world since 1974. Although related studies in China started in1980 s, a great progress has been achieved during the last three decades. Therefore, this review summarized the main achievements on disinfection and DPBs studies in China, which included:(1) the occurrence of DBPs in water of China,(2) the identification and detection methods of DBPs,(3) the formation mechanisms of DBPs during disinfection process,(4) the toxicological effects and epidemiological surveys of DBPs,(5) the control and management countermeasures of DBPs in water disinfection, and(6) the challenges and chances of DBPs studies in future. 3Driving and Braking Control of PM Synchronous Motor Based on Low-resolution Hall Sensor for Battery Electric Vehicle显示文摘Resolvers are normally employed for rotor positioning in motors for electric vehicles,but resolvers are expensive and vulnerable to vibrations.Hall sensors have the advantages of low cost and high reliability,but the positioning accuracy is low.Motors with Hall sensors are typically controlled by six-step commutation algorithm,which brings high torque ripple.This paper studies the high-performance driving and braking control of the in-wheel permanent magnetic synchronous motor(PMSM) based on low-resolution Hall sensors.Field oriented control(FOC) based on Hall-effect sensors is developed to reduce the torque ripple.The positioning accuracy of the Hall sensors is improved by interpolation between two consecutive Hall signals using the estimated motor speed.The position error from the misalignment of the Hall sensors is compensated by the precise calibration of Hall transition timing.The braking control algorithms based on six-step commutation and FOC are studied.Two variants of the six-step commutation braking control,namely,half-bridge commutation and full-bridge commutation,are discussed and compared,which shows that the full-bridge commutation could better explore the potential of the back electro-motive forces(EMF),thus can deliver higher efficiency and smaller current ripple.The FOC braking is analyzed with the phasor diagrams.At a given motor speed,the motor turns from the regenerative braking mode into the plug braking mode if the braking torque exceeds a certain limit,which is proportional to the motor speed.Tests in the dynamometer show that a smooth control could be realized by FOC driving control and the highest efficiency and the smallest current ripple could be achieved by FOC braking control,compared to six-step commutation braking control.Therefore,FOC braking is selected as the braking control algorithm for electric vehicles.The proposed research ensures a good motor control performance while maintaining low cost and high reliability.GU Jing OUYANG Minggao LI Jianqiu LU Dongbin FANG Chuan MA Yan 2013Chinese Journal of Mechanical Engineering2013,26,1:14
4Characterization of dissolved organic matter fractions and its relationship with the disinfection by-product formation显示文摘Dissolved organic matter(DOM) has been identified as precursor for disinfection by-products(DBPs) formation during chlorination. Recently,it has been demonstrated that the characteristics of DOM influence the DBPs formation mechanism. A study was,therefore,initiated to investigate the effects of DOM fractions on DBPs formation mechanism. In the chlorination process,organic acids are dominant precursors of total thihalomethanes(TTHM) because of the νC-O and unsaturated structures. Furthermore,the TTHM formation of organic acids was affected by pH more greatly. Based on the fluorescence spectroscopy analysis,DOM fractions contained several fluorescence substances. During chlorination,humic acid-like substances were found to exhibit high chlorine reactivity and hydrophobic organics decomposed to smaller molecules faster than hydrophilic organics even at lower chlorine dosages. 5Rule-based Fault Diagnosis of Hall Sensors and Fault-tolerant Control of PMSM显示文摘Hall sensor is widely used for estimating rotor phase of permanent magnet synchronous motor(PMSM). And rotor position is an essential parameter of PMSM control algorithm, hence it is very dangerous if Hall senor faults occur. But there is scarcely any research focusing on fault diagnosis and fault-tolerant control of Hall sensor used in PMSM. From this standpoint, the Hall sensor faults which may occur during the PMSM operating are theoretically analyzed. According to the analysis results, the fault diagnosis algorithm of Hall sensor, which is based on three rules, is proposed to classify the fault phenomena accurately. The rotor phase estimation algorithms, based on one or two Hall sensor(s), are initialized to engender the fault-tolerant control algorithm. The fault diagnosis algorithm can detect 60 Hall fault phenomena in total as well as all detections can be fulfilled in 1/138 rotor rotation period. The fault-tolerant control algorithm can achieve a smooth torque production which means the same control effect as normal control mode (with three Hall sensors). Finally, the PMSM bench test verifies the accuracy and rapidity of fault diagnosis and fault-tolerant control strategies. The fault diagnosis algorithm can detect all Hall sensor faults promptly and fault-tolerant control algorithm allows the PMSM to face failure conditions of one or two Hall sensor(s). In addition, the transitions between health-control and fault-tolerant control conditions are smooth without any additional noise and harshness. 6A biological safety evaluation on reclaimed water reused as scenic water using a bioassay battery显示文摘An assessment method based on three toxicity tests (algae growth inhibition,daphnia immobilization and larval fish toxicity) was used to screen the biological safety of reclaimed water which was reused as sole replenishment for scenic water system in a park (SOF Park) in northern China.A total of 24 water samples were collected from six sites of water system in the SOF Park in four different seasons.The results indicated that:(1) the reclaimed water directly discharged from a reclamation treatment plant near the SOF Park as influent of park had relatively low biological safety (all samples were ranked as C or D);(2) the biological safety of reclaimed water was improved greatly with the ecological reclamation treatment processes composing of artificial wetland system and followed oxidation pond system;(3) the biological safety of reclaimed water in the main lake of SOF Park kept at a health status during different seasons (all samples were ranked as A);(4) there was some certain correlation (R 2=0.5737) between the sum of toxicity scores and dissolved organic carbon for the studied water samples.It was concluded that the assessment method was reliable to screen the safety of reclaimed water reused as scenic water,and the reclaimed water with further ecological purification processes such as artificial wetland and oxidation pond system can be safely reused as scenic water in park.Dongbin Wei Zhuowei Tan Yuguo Du 2011Journal of Environmental Sciences2011,23,10:9
7A toxicity-based method for evaluating safety of reclaimed water for environmental reuses显示文摘A large quantity of toxic chemical pollutants possibly remains in reclaimed water due to the limited removal efficiency in traditional reclamation processes. It is not enough to guarantee the safety of reclaimed water using conventional water quality criteria. An integrated assessment method based on toxicity test is necessary to vividly depict the safety of reclaimed water for reuse. A toxicity test battery consisting of lethality, genotoxicity and endocrine disrupting effect was designed to screen the multiple biological effects of residual toxic chemicals in reclaimed water. The toxicity results of reclaimed water were converted into the equivalent concentrations of the corresponding positive reference substances(EQC). Simultaneously, the predicted no-effect concentration(PNEC) of each positive reference substance was obtained by analyzing the species sensitivity distribution(SSD) of toxicity data. An index 'toxicity score' was proposed and valued as 1, 2, 3, or 4 depending on the ratio of the corresponding EQC to PNEC. For vividly ranking the safety of reclaimed water, an integrated assessment index 'toxicity rank' was proposed, which was classified into A, B, C, or D rank with A being the safest. 8Sequential use of ultraviolet light and chlorine for reclaimed water disinfection显示文摘Several disinfection processes of ultraviolet (UV),chlorine or UV followed by chlorine were investigated in municipal wastewater according to the inactivation of Escherichia coli,Shigella dysenteriae and toxicity formation.The UV inactivation of the tested pathogenic bacteria was not affected by the quality of water.It was found that the inactivated bacteria were obviously reactivated after one day in dark.Fluorescent light irradiation increased the bacteria repair.The increase of UV dosage could cause more damage to bacteria to inhibit bacteria self-repair.No photoreactivation was detected when the UV dose was up to 80 mJ/cm2 for E.coli DH5α,and 23 mJ/cm2 for S.dysenteriae.Nevertheless,sequential use of 8 mJ/cm2 of UV and low concentration of chlorine (1.5 mg/L) could effectively inhibit the photoreactivation and inactivate E.coli below the detection limits within seven days.Compared to chlorination alone,the sequential disinfection decreased the genotoxicity of treated wastewater,especially for the sample with high NH 3-N concentration.Xiujuan Wang Xuexiang Hu Chun Hu Dongbin Wei 2011Journal of Environmental Sciences2011,23,10:8
9Toxicity-based assessment of the treatment performance of wastewater treatment and reclamation processes显示文摘The reclamation and reuse of wastewater is one of the possible ways to relieve the serious fresh water resource crisis in China. Efficient reclamation treatment technologies ensure the safe reuse of reclaimed water. In order to screen out and evaluate technologies appropriate for reclamation treatment, a great deal of efforts have been brought to bear. In the present study, a toxicity-based method including a Photobacterium phosphoreum test for acute toxicity and SOS/umu test for genotoxicity, accompanied by the traditional physicochemical parameters DOC (dissolved organic carbon) and UV 254 (absorbance at 254 nm), was used to measure the treatment performance of different reclamation processes, including the anaerobic-anoxic-oxic biological process (A2O) and subsequent physical/chemical reclamation processes (ultrafiltration, ozonation, chlorination). It was found that for the secondary effluent after the A2O process, both the toxicity and physicochemical indices had greatly decreased compared with those of the influent. However, chemical reclamation processes such as ozonation and chlorination could possibly raise toxicity levels again. Fortunately, the toxicity elevation could be avoided by optimizing the ozone dosage and using activated carbon after ozonation. It was noted that by increasing the ozone dosage to 10 mg/L and employing activated carbon with more than 10 min hydraulic retention time, toxicity elevation was controlled. Furthermore, it was shown that pre-ozonation before activated carbon and chlorination played an important role in removing organic compounds and reducing the toxicity formation potential. 10Leader-follower formation control without leader's velocity information显示文摘In this paper,we consider the mobile robots formation control problem without direct measurement of the leader robot’s linear velocity.Two decentralized nonlinear algorithms are proposed,respectively,based on adaptive dynamic feedback and immersion&invariance estimation based second order sliding mode control methodologies.The main idea is to solve formation problem by estimating the leader robots’s linear velocity,while maintaining the given predefined separation distance and bearing angle between the leader robot and the follower robot.The stability of the closed-loop system is proven by means of the Lyapunov method.The proposed controllers are smooth,continuous and robust against unknown bounded uncertainties such as sensor inaccuracy between the outputs of sensors and the true values in collision free environments.Simulation examples and physical vehicles experiments are presented to verify the effectiveness of the proposed design approaches,and the proposed designed methodologies are carefully compared to illustrate the pros and cons of the approaches.SHEN DongBin SUN ZhenDong SUN WeiJie 2014Science China(Information Sciences)2014,57,9:6
11Photolysis of chlortetracycline in aqueous solution:Kinetics,toxicity and products显示文摘The aqueous photodegradation of the widely used antibiotic chlortetracycline(CTC) was investigated under simulated sunlight.The quantum yield of photodegradation increased from 3.3 × 10-4 to 8.5 × 10-3 within the pH range of 6.0 to 9.0.The presence of Ca2+,Fe3+,and NO3-enhanced the photodegradation of CTC,whereas Mg2+,Mn2+,and Zn2+ inhibited the degradation with the order Mn2+ > Zn2+ > Mg2+ at pH 7.3.The monovalent cations(Na+ and K+) had negligible effect on the photolysis of CTC.Fulvic acid(FA) decreased the photodegradation of CTC due to light screening effect.Hydrogen peroxide(H2O2) was formed concurrently with direct photodegradation of CTC.The generation rate of H2O2 increased from 0.027 to 0.086 μmol/(L.min) when the pH ranged from 6.0 to 9.0.The CTC solution was about three-fold more toxic to the Photobacterium phosphoreum bacteria after irradiation,suggesting that the photoproducts and H2O2 formed in the CTC solution exhibited high risk on the bacteria.By LC-ESI(+)-MS,the photoproducts of CTC were identified.The direct photodegradation of CTC was involved in hydroxylation and N-demethyl/dedismethyl processes.The main photoproducts included the iso-CTC analog containing hydroxyl groups(m/z 511.4 and 495.4),and the N-demethyl/dedismethyl products of the photoproduct m/z 495.4(m/z 481.3 and 467.4).In addition,the photochemical dechlorination of CTC led to tetracycline(m/z 445.5).Yong Chen Hua Li Zongping Wang Tao Tao Dongbin Wei Chun Hu 2012Journal of Environmental Sciences2012,24,2:6
12Efficient Collocational Approach for Parametric Uncertainty Analysis显示文摘A numerical algorithm for effective incorporation of parametric uncertainty into mathematical models is presented.The uncertain parameters are modeled as random variables,and the governing equations are treated as stochastic.The solutions,or quantities of interests,are expressed as convergent series of orthogonal polynomial expansions in terms of the input random parameters.A high-order stochastic collocation method is employed to solve the solution statistics,and more importantly,to reconstruct the polynomial expansion.While retaining the high accuracy by polynomial expansion,the resulting“pseudo-spectral”type algorithm is straightforward to implement as it requires only repetitive deterministic simulations.An estimate on error bounded is presented,along with numerical examples for problems with relatively complicated forms of governing equations.Dongbin Xiu 2007Communications in Computational Physics2007,2,2:6
13Predicted no-effect concentrations for mercury species and ecological risk assessment for mercury pollution in aquatic environment显示文摘Mercury(Hg) exists in different chemical forms presenting varied toxic potentials. It is necessary to explore an ecological risk assessment method for different mercury species in aquatic environment. The predicted no-effect concentrations(PNECs) for Hg(Ⅱ) and methyl mercury(Me Hg) in the aqueous phase, calculated using the species sensitivity distribution method and the assessment factor method, were 0.39 and 6.5 × 10-3μg/L, respectively. The partition theory of Hg between sediment and aqueous phases was considered, along with PNECs for the aqueous phase to conduct an ecological risk assessment for Hg in the sediment phase. Two case studies, one in China and one in the Western Black Sea, were conducted using these PNECs. 14Acute toxicity formation potential of benzophenone-type UV filters in chlorination disinfection process显示文摘Benzophenones(BPs) are a class of widely used UV filters, which have been frequently detected within multiple environmental matrices. Disinfection is a necessary process in water treatment processes. The transformation behaviors and toxicity changes of 14 BP-type UV filters during chlorination disinfection treatment were investigated in this study. A new index, the acute toxicity formation potential, was proposed to evaluate the toxicity changes and potential risks of BP-type UV filters during chlorination treatment. It was found that 13 of 14 BP-type UV filters exhibited toxicity decreases in the chlorination disinfection process, more or less, while one showed a toxicity increase. The toxicity changes were dependent on substitution effects, such that 2,4-di-hydroxylated or 3-hydroxylated BPs exhibited significant toxicity decreases after chlorination treatment due to the ready cleavage of the aromatic ring. 15High-performance self-assembly MnCo2O4 nanosheets for asymmetric supercapacitors显示文摘In this study,MnCo2O4 nanosheets were proposed to be utilized as an electrode material for supercapacitors.A two-step hydrothermal method with post-annealing treatment was employed in preparation of the nanostructures.MnCo2O4 electrode delivered a high specific capacitance of 2000 F g^-1 at 0.5 A g^-1,remarkable high-rate capability of 1150 F g^-1 at 20 A g^-1,and an excellent cycling stability of 92.3%at 5 A g^-1 after 5000 cycles.It is found that a three-electrode supercapacitor based on MnCo2O4 exhibits a promising electrochemical performance,better than the other similar materials,benefited from the synergistic effects of MnCo2O4 nanosheets.In fact,the self-assembly of nanosheets structure with high specific surface area and mesoporous structure can potentially enhance the electrochemical performance of supercapacitors.Jianwei Li Dongbin Xiong Linzhe Wang Maleki Kheimeh Sari Hirbod Xifei Li 2019Journal of Energy Chemistry2019,28,10:4
16The potential risk assessment for different arsenic species in the aquatic environment显示文摘The different toxicity characteristics of arsenic species result in discrepant ecological risk.The predicted no-effect concentrations(PNECs) 43.65, 250.18, and 2.00 × 103μg/L were calculated for As(III), As(V), and dimethylarsinic acid in aqueous phase, respectively. With these PNECs, the ecological risk from arsenic species in Pearl River Delta in China and Kwabrafo stream in Ghana was evaluated. It was found that the risk from As(III) and As(V)in the samples from Pearl River Delta was low, while much high in Kwabrafo stream. 20Clique-based Cooperative Multiagent Reinforcement Learning Using Factor Graphs显示文摘In this paper,we propose a clique-based sparse reinforcement learning(RL) algorithm for solving cooperative tasks.The aim is to accelerate the learning speed of the original sparse RL algorithm and to make it applicable for tasks decomposed in a more general manner.First,a transition function is estimated and used to update the Q-value function,which greatly reduces the learning time.Second,it is more reasonable to divide agents into cliques,each of which is only responsible for a specific subtask.In this way,the global Q-value function is decomposed into the sum of several simpler local Q-value functions.Such decomposition is expressed by a factor graph and exploited by the general maxplus algorithm to obtain the greedy joint action.Experimental results show that the proposed approach outperforms others with better performance.Zhen Zhang Dongbin Zhao 2014IEEE/CAA Journal of Automatica Sinica2014,1,3:3
