1Characterization of toxin from Verocytotoxigenic Ecscherichia coli(VTEC) strains isolated from neonatal calves in India显示文摘Objective:The present study has characterized dialyzed toxin from non-0157 VTEC E.coli isolates by vero cell toxicity assay and pathogenecity in mice model.Methods:Toxins from non-0157 verocytotoxic Escherichia coli isolated from neonatal calves were characterized.Dialyzed toxin from E.coli 026,0111 and 0103 serotypes were prepared and characterized by verocell toxicity assay and pathogenicity in mice model.E.coli 0157:H7 considered as positive control for this study.Results:Cytopathic effects in vero cell line first rounding of vero cells,followed by clumping of cells and finally disintegrated,blackened,shriveled cell line within 16 to 72 hrs.Phenotypic markers such as hind limb paralysis and reddening of tail were prominent in all the toxicated mice.Extensive histopathological study was conducted for multiple organ involvement.Conclusion: Several methods for toxin assay were developed based on biological,immunological and detection of virulence genes related to toxin production but each test has draw back.Therefore,it is likely that future effort will be focused on the development of assay,which is fast,reliable,specific and sensitive methods based on mice model.Diganta Pan Ashok Kumar Bhatia Bhilegaonkar KN 2009Asian Pacific Journal of Tropical Medicine2009,2,2:1
2Molecular differentiation of cryptic stage of Echinococcus granulosus and Taenia species from faecal and environmental samples显示文摘Objective:To differentiate cryptic stage of Echinococcus granulosus(E.granulosus) and Taenia by PCR-RFLP and sequence information of amplicon.Methods:DNA were isolated from metacestodes stage of Taenia and E.granulosus using DNA isolation kit(Q-BIOgene kit,USA), the amplified and purified DNA product was then cloned and sent for sequencing.The generating sequence information was used for amplicons identification.Results:Out of 112 faecal and environmental samples,16 exhibited positive result.The product size of amplicon positive for E.granulosus was 310 bp;whereas,for Taenia spp.sizes varied from 379 to 388 bp.Restriction pr of ile of actinⅡwith Csp61 also differed Taenia spp.and E.granulosus.Conclusions:The result of the study indicated that,the primers were useful to differentiate cryptic stage of the two genera which is yet to be reported earlier.Diganta Pan Sumanta De Asit Kumar Bera Subhashis Bandyopadhyay Subrata Kumar Das Debasis Bhattacharya 2010Asian Pacific Journal of Tropical Medicine2010,3,4:0
3Enterohaemolysin production and verotoxin genes in Escherichia coli strains isolated from neonatal calves in India显示文摘Objective:To screened all haemolytic strains of Escherichia coli(E.coli) for the presence of verotoxin genes and speculate the association between enterohaemolysin production and presence of verotoxin genes.Methods: A total 176 of E.coli strains of 64 serogroups isolated from neonatal calves were selected and screened for alpha -haemolysin and enterohaemolysin production on sheep blood agar and 5%washed sheep blood agar.Two types of haemolytic strains were further characterized by PCR for presence of Verotoxin gene(VT1 and VT2) and confirmed by verocell cytotoxicity assay.Results:Among 27 enterohaemolytic positive strains,19(70. 37%) strains were found positive for presence of verotoxins(VTs) gene and verocell cytotoxicity assay. Whereas 34 alpha-haemolysin strains were found negative for VTs.Eight strains of E.coli were found VTs negative but Ehx positive.Conclusion:The close association between enterohaemolysin production and presence of verotoxin genes makes it useful epidemiological marker for rapid screening of Verotoxic E.coli but this phenotypic marker for screening of verotoxigenic Escherichia coli(VTEC) alone may raise a question in animal.Diganta Pan Ashok Kumar Bhatia K.N.Bhilegaonkar 2009Asian Pacific Journal of Tropical Medicine2009,2,4:0
