| 1 | Effects of salvianolic acid B on proliferation, neurite outgrowth and differentiation of neural stem cells derived from the cerebral cortex of embryonic mice显示文摘Salvianolic acid B is isolated from Salvia miltiorrhiza,the root of which is widely used as a traditional Chinese medicine to treat stroke.However,little is known about how salvianolic acid B influences growth characteristics of neural stem cells (NSCs).The purpose of the present study was to evaluate the effects of salvianolic acid B on proliferation,neurite outgrowth and differentiation of NSCs derived from the cerebral cortex of embryonic mice using MTT,flow cytometry,immunofluorescence and RT-PCR.It was found that 20 μg mL·1 and 40 μg mL·1 salvianolic acid B had similar effects on proliferation of NSCs,and a suitable concentration of salvianolic acid B increased the number of NSCs and their derivative neurospheres.The growth-promoting activity of salvianolic acid B was dependent on and associated with an accumulation in the G2/S-phase cell population.Salvianolic acid B also promoted the neurite outgrowth of NSCs and their differentiation into neurons.The mRNA for tau,GFAP and nestin were present in differentiating neurospheres induced by salvianolic acid B.However,high-level expression of tau mRNA and low-level expression of GFAP mRNA was detected in differentiated cells,in contrast to the control conditions.This collective evidence indicates that exogenous salvianolic acid B is capable of promoting proliferation of neurospheres and differentiation towards the neuronal lineage in vitro and may act in the proliferation of NSCs and may promote NSC differentiation into neuronal cells. | GUO GuoQing1,LI Bin1,WANG YuanYuan1,SHAN AiJing1,SHEN WeiZai1,YUAN Lin2 & ZHONG ShiZhen2 1Department of Anatomy,Medical College of Jinan University,Guangzhou 510630,China 2Institute of Clinical Anatomy,Key Laboratory of Tissue Construction and Detection of Guangdong Province,Southern Medical University,Guangzhou 510150,China | 2010 | Science China(Life Sciences)2010,53,6: | 21 |
| 2 | 启东肝癌早诊早治示范基地项目工作进展显示文摘[目的]在肝癌高发现场开展肝癌的筛查以发现早期肝癌病例、落实及时的、适宜的治疗措施。[方法]肝癌的筛查经历了不同的过程,从筛查自然人群到选择特定的目标人群,筛查乙肝病毒表面抗原(HBsAg)阳性者作为肝癌的高危险人群,从现场摸索到系统地实践并加以研究,目前按照中国癌症基金会对'早诊早治示范基地'制订并提出的方案和流程,采用甲胎蛋白(AFP)加B超检测的方法对肝癌实行一年两次的筛查;对AFP异常者实施临床随访复查,直至交临床确诊并治疗。[结果]20世纪70年代之初,在启东自然人群中检测AFP200多万人次,其中普查近180万人次,检出肝癌1000多例,其中早期(Ⅰ期)病例达到35%。80年代末到90年代初,在目标人群中筛查到5581例HBsAg阳性者,确定了肝癌高危险人群筛检模式和可行方案。2007年以来,AFP筛查启东6个乡镇29233人,查出HBsAg阳性者2878人(9.85%),每年接受两次AFP检测、B超检查(复查)。三年筛查共发现64例肝癌新病例,肝癌发病率为1882.53/10万人年。其中早期肝癌为45例,早期诊断率为70.31%。64例筛查病例中,52例(81.25%)落实了及时的适宜治疗:有12例(18.75%)因经济或思想原因而放弃复诊或放弃治疗,其中6例为早期肝癌。其中在2009年上半年的筛查中,筛查高危人群9926人,查出HBsAg阳性者840人(8.46%),其中670人参加了B超复查(79.76),发现肝癌12例;对原有的HBsAg阳性者进行复查,也发现12例肝癌。此24例中,19例为早期肝癌病例(79.17%),17例及时落实了治疗(87.57%)。[结论]AFP筛查一直是现场肝癌早诊中最常用的检测方法,现场高危人群是筛查的最佳目标人群,AFP检测加B超检查已被证明是肝癌最有效的早期发现和早期诊断手段。有效的组织管理、完善的技术方案和健全的技术队伍可以保证筛查的质量、提高早期病人的发现率,并落实适宜的早期治疗措施。早期病人在落实治疗过程中遇到的经济原因和思想问题,是卫生体制改革、新农合保障机制、二级预防和防癌宣传方面值得继续探讨的课题。 | Group of Early Detection and Treatment,Research Institute of Liver Cancer of Qidong City,Jiangsu Province,Qidong 226200,China | 2009 | 中国肿瘤2009,18,9: | 7 |

| 4 | Synthesis of GoldMag particles with assembled structure and their applications in immunoassay显示文摘Micrometer-sized Fe3O4 particles and nano-sized gold particles were first synthesized by methods of self-aggregation of surface-chemically modified Fe3O4 nanoparticles and citrate reduction of the Au3+ to Au0, respectively. Interaction between these two types of particles resulted in the assembly of nano-sized gold particles on the surface of the micrometer-sized Fe3O4 particles, forming an assembled structure with the Fe3O4 core particles around which are attached nano-sized gold parti- cles. The Fe3O4/Au structure is named GoldMag particles with assembled structure. The synthetic process, structure, and magnetic property of the GoldMag particles were analyzed. GoldMag particles with assembled structure have an irregular shape, rough surface with a diameter of 2―3 μm. These particles exhibit the superparamagnetic property with saturated magnetization of 41 A·m2/kg. In a single step, antibodies could be readily immobilized onto the surface of the particles with a high binding capacity. The GoldMag particles can be used as a novel carrier in immunoassays. The maximum quantity of human IgG immobilized onto GoldMag particles was 330 μg/mg. In order to validate the quality of the GoldMag particles as immunoassay carriers, an immunoassay system was used. The relative amount of immobilized human IgG was measured by HRP-labeled anti human IgG. The coefficient of variation within parallel samples of each group was below 6% and the coefficient of variation of means between five groups carried out separately was below 7%. Based on the sandwich method, the Hepatitis B surface antigen (HBsAg) and interleukin-8 (IL-8) were also analyzed by qualitative and quantitative detection, respectively.
