| 2 | Neuroprotection of aucubin in primary diabetic encephalopathy显示文摘Hippocampal neuronal apoptosis accompanied by impairment of cognitive function occurs in primary diabetic encephalopathy. In this study, we investigated the neuroprotective mechanism of the iridoid glycoside, aucubin, using rats (n=8). Diabetes mellitus was induced in the rats by intraperitoneal (i.p.) injection of streptozotocin (60 mg/kg body weight). After 65 d, half of the DM rats were administered aucubin (5 mg/kg; i.p.) for 15 d, yielding treatment DM+A. A third group of rats received no strepto- zotocin or aucibin, and served as controls (CON). Encephalopathy was assessed using Y-maze be- havioral testing. Rats were euthanized on Day 87, and hippocampi were excised for visual (light and transmission electron microscopic) and immunochemical (Western blot; immunohistochemical) as- sessments of the CA1 subfield for apoptosis and expression of regulatory proteins Bcl-2 and Bax. Treatment responses to all the parameters examined (body weight, plasma glucose, Y-maze error rates, pyramidal cell ultrastructure, proportions of apoptotic cells, levels of expression of Bcl-2 and Bax, and survivability of neuronal cells) were identical: there were highly significant differences between DM and CON groups (P<0.001), but the effects were significantly moderated (P<0.01) in DM+A compared with DM. These findings confirm the association of apoptosis with the encephalopathic effects of diabetes mellitus, and suggest a major role of the expression levels of Bcl-2 and Bax in the regulation of apop- totic cell death. | 3 | Selenium and the Thioredoxin and Glutaredoxin Systems显示文摘Thioredoxin (Trx) is a small ubiquitous dithiol protein which together with the FADcontaining enzyme thioredoxin reductase (TR) and NADPH (the Trx system) is a hydrogen donor for ribonucleotide reductase essential for DNA synthesis and a general protein disulfide reductase involved in redox regulation. Selenite, selenodiglutathione (GS-Se-SG) and selenocystine are efficiently reduced by thioredoxins and also directly by NADPH and mammalian TR but not by the E. coli enzyme. Incubation of selenite or GS-Se-SG with the Trx system or with mammalian TR results in a rapid formation of selenide, which by redox cycling with oxygen may cause a large non-stoichiometric oxidation of NADPH. Selenocystine is efficiently reduced into two molecules of the selenol amino acid selenocysteine by mammalian TR with a Km-value (6μmol·L-1 ) and a high turnover number (kcat, 3200 min-1) almost identical to the natural substrate Trx-S2. TR also directly reduces lipid hydroperoxides and this peroxidase reaction is strongly stimulated by the presence of catalytic amounts of free selenocysteine. Glutaredoxin (Grx) which catalyzes GSH dependent disulfide reduction also via a redox-active disulfide and Trx are both efficient electron donors to the hut-nan plasrna glutathione peroxidase providing a mechanism by which human plasma glutathione peroxidase may reduce hydroperoxides in an environment almost free from glutathione. Selenate is reduced by Grx and Trx in the presence of GSH. The DNA-binding of the transcription factor AP-1 is strongly inhibited by GS-Se-SG and selenite. Furtherrnore, selenide formed by TR-mediated reduction of selenite and GS-Se-SG inhibits lipoxygenase and changes the electron spin resonance spectrum of the active site iron. Mammalian TR with two subunits of 57 kDa has recently been cloned and shown to be homologous to glutathione reductase. The rat enzyme contains a selenocysteine residue in a unique Cterminal position and a conserved SEClS sequence directing insertion of the selenocysteine. | 4 | Structure comparisons of Aedes albopictus densovirus with other parvoviruses显示文摘Parvoviridae is a family of the smallest viruses known with a wide variety of hosts. The capsid structure of the Aedes albopictus C6/36 cell densovirus (C6/36 DNV) at 1.2-nm resolution was obtained by elec-tron cryomicroscopy (cryoEM) and three-dimensional (3D) image reconstruction. Structure compari-sons between the C6/36 DNV and other parvoviruses reveal that the degree of structural similarity be-tween C6/36 DNV and the human parvovirus B19 is higher than that between C6/36 DNV and other in-sect parvoviruses. The amino acid sequence comparisons of structural and non-structural proteins also reveal higher levels of similarity between C6/36 DNV and parvovirus B19 than those between C6/36 DNV and other parvoviruses. | 5 | Study on the Resistance Indexes of Four Kinds of Desert Plants in Qinghai Lake Area显示文摘[Objective] The research aimed to study the resistance of four kinds of desert plants in Qinhai Lake area.[Method] By contrasting the resistance indexes of four kinds of plants which included Ephedra intermedia,Stellera chamaejasme,Achnatherum splendens and Xanthopappus subacaulis,the resistance of four kinds of plants in Qinghai Lake area was analyzed.The resistance indexes included the soluble protein,MDA,free Pro content and the activities of SOD,POD,CAT.Moreover,the resistance of four kinds of plants was evaluated comprehensively by using Fuzzy membership function method.[Result] Under the low-temperature and arid adversity in Qinghai Lake area,SOD activity presented as Stellera chamaejasme>Xanthopappus subacaulis>Ephedra intermedia>Achnatherum splendens,and CAT activity presented as Stellera chamaejasme>Xanthopappus subacaulis>Achnatherum splendens>Ephedra intermedia.Under the same high-salt environment,CAT activity of Stellera chamaejasme was eight times higher than that of Ephedra intermedia.It illustrated that the salt resistance of Stellera chamaejasme was stronger than that of Ephedra intermedia.The free Pro content showed as Xanthopappus subacaulis>Ephedra intermedia>Stellera chamaejasme>Achnatherum splendens.Under the same adversity,Pro content of Ephedra intermedia was 2.83 times of Stellera chamaejasme.It was because that MDA content in Stellera chamaejasme was the lowest,and the membranous peroxide harm was the smallest.It caused that Pro content (stress penetration material) was low.It also illustrated that the resistance of Stellera chamaejasme was stronger than that of Ephedra intermedia.The comprehensive evaluation of Fuzzy membership function showed that the resistance presented as Xanthopappus subacaulis>Stellera chamaejasme>Ephedra intermedia>Achnatherum splendens.[Conclusion] The research provided the theory basis for the breeding of drought-resistance new variety and the development of good germ plasm resource in Qinghai Lake area. | SHI Hui-lan Department of Biosciences,Qinghai University,Xining 810016,China | 2011 | Meteorological and Environmental Research2011,2,5: | 1 |

| 10 | A PCA-based method for ancestral informative markers selection in structured populations显示文摘Identification of population structure can help trace population histories and identify disease genes. Structured association (SA) is a commonly used approach for population structure identification and association mapping. A major issue with SA is that its performance greatly depends on the informa-tiveness and the numbers of ancestral informative markers (AIMs). Present major AIM selection meth-ods mostly require prior individual ancestry information, which is usually not available or uncertain in practice. To address this potential weakness, we herein develop a novel approach for AIM selection based on principle component analysis (PCA), which does not require prior ancestry information of study subjects.
