1Betulinic acid induces apoptosis of gallbladder cancer cells via repressing SCD1显示文摘Gallbladder cancer(GBC)is the most common and aggressive malignancy of the biliary tract.Betulinic acid(BetA)has been reported to have anti-inflammatory and antitumor effects;however,the effect of BetA on GBC is still unknown.In this study,we investigated the effect of BetA on five GBC cell lines and found that BetA significantly inhibited the proliferation of NOZ cells but had lttle inhibitory effect on other GBC cells.BetA disturbed mitochondrial membrane potential and induced apoptosis in NOZ cells.Real-time polymerase chain reaction analysis revealed that stearoyl-coenzyme A desaturase 1(SCD1)was highly expressed in NOZ cells but low expressed in other GBC cells.BetA inhibited SCD1 expression in a concentration-dependent manner in NOZ cells.Downregulation of SCD1 expression by RNA interference inhibited the proliferation of NOZ cells and induced cell apoptosis.Moreover,BetA inhibited the growth of xenografted tumors and suppressed SCD1 expression in nude mice.Thus,our results showed that BetA induced apoptosis through repressing SCD1 expression in GBC,suggesting that BetA might be an effective agent for the treatment of patients with GBC that highly expresses SCD1.Hongfei Wang Fangxiao Dong Ye Wang Xu'an Wang Defei Hong Yingbin Liu Jian Zhou 2020Acta Biochimica et Biophysica Sinica2020,52,2:6
2Regulation of BMP2K in AP2M1-mediated EGFR internalization during the development of gallbladder cancer显示文摘Dear Editor,Gallbladder cancer(GBC),the most common malignant tumor of the biliary tract,is a highly invasive form of cancer.Surgical resection is currently the first line approach to effectively treat GBC;however,very few patients have the opportunity to receive radical surgical treatment due to lack of obvious symptoms.1 The median survival of patients with GBC is only 12 months and 5-year survival rate is<5%,indicating GBC is extremely poor prognosis.Therefore,it is urgent to identify novel key molecules that can potentially serve as early diagnostic biomarkers and/or therapeutic targets.The current study focused on the potential role of bone morphogenetic protein 2 inducible kinase(BMP2K),a serine/threonine kinase,which was recently identified as clathrin-coated vesicle-associated protein in the development of GBC;the outcome may hold diagnostic and therapeutic promising for clinical practice.Xiaoling Song Maolan Li Wenguang Wu Wei Dang Yuan Gao Rui Bian Runfa Bao Yunping Hu Defei Hong Jun Gu Yingbin Liu 2020Signal Transduction and Targeted Therapy2020,5,1:1
