| 1 | Numerical and experimental study of magnetic fluid seal with large sealing gap and multiple magnetic sources显示文摘To improve the pressure capability of magnetic fluid seal with more than a 0.25 mm single edge gap,a magnetic fluid sealing structure with multiple magnetic sources which has five permanent magnets was designed.Magnetic field distributions under the pole pieces of the magnetic fluid seal with single and multiple magnetic sources were simulated by finite element method and its sealing pressure difference could be calculated according to the theoretical formula of the magnetic fluid seal.The effects of sealing gap height and magnetic source amount on the sealing capability were investigated experimentally.The theoretical and experimental results were compared,analyzed and discussed.The results demonstrated that the magnetic fluid seal with multiple magnetic sources was an effective method to improve the sealing capability for the rotary shaft with large gaps.The theoretical results agreed well with the experimental results when the height of the single edge gap was equal to 0.4 mm.However,it was found that when the height of the single edge gap was larger than 0.4 mm,the difference between the theoretical values and experimental values increased with the gap height because part of magnetic source had less or even no effect on magnetic fluid sealing capability. | YANG XiaoLong ZHANG ZhiLi LI DeCai | 2013 | Science China(Technological Sciences)2013,56,11: | 10 |
| 2 | Community assemblage of free-living diazotrophs along the elevational gradient of Mount Gongga显示文摘Mountain systems are unique for studying the responses of species distribution and diversity to environmental changes along elevational gradients.It is well known that free-living diazotrophic microorganisms are important to nitrogen cycling in mountain systems.However,the elevational patterns of free-living diazotrophs and the underlying ecological processes in controlling their turnover along broader gradients are less well documented.Here,we investigated the pattern of diazotrophic diversity along the elevational gradient(1800 m-4100 m)in Mount Gongga of China.The results showed that the α-diversity of diazotrophs did not change with the elevation from 1800 m to 2800 m,but decreased at elevations above 3000 m.Such diversity pattern was driven mainly by soil total carbon,nitrogen,and plant richness.Various diazotrophic taxa showed differential abundance-elevation relationships.Ecological processes determining diazotrophic community assemblage shift along the elevations.Deterministic processes were relatively stronger at both low and high elevations,whereas stochastic processes were stronger at the middle elevation.This study also suggested a strong relationship among aboveground plants and diazotrophs,highlighting their potential interactions,even for free-living diazotrophs. | Yansu Wang Chaonan Li Zehao Shen Junpeng Rui Decai Jin Jiabao Li Xiangzhen Li | 2019 | Soil Ecology Letters2019,1,3: | 8 |

| 4 | Identification and characterization of tomato gibberellin 2-oxidases (GA2oxs) and effects of fruit-specific SlGA2ox1 overexpression on fruit and seed growth and development显示文摘Gibberellins(GAs)play a crucial role in growth and development of the tomato fruit.Previously published studies focusing on the effect of GAs on tomato fruits used chemical treatments,constitutive overexpression or silencing of GA biosynthetic and catabolic genes globally throughout the plant.Fruit-specific overexpression of GA catabolic enzyme genes GA2-oxidases(GA2oxs),however,may provide an alternative method to study the role of endogenous GAs on the fruit development.In this study,we have identified 11 SlGA2ox proteins in tomato that are classified into three subgroups.Motif analysis and multiple sequence alignments have demonstrated that all SlGA2oxs,except SlGA2ox10,have similar motif compositions and high-sequence conservation.Quantitative reverse transcription-PCR analysis has showed that SlGA2oxs exhibit differential expression patterns in tomato fruits at different developmental stages.When the fruit-specific promoter TFM7 was used to control the expression of SlGA2ox1,we observed no changes in growth and development of vegetative organs.However,fruit weight,seed number and germination rate were significantly affected.We also treated tomato fruits with GA biosynthesis inhibitor and observed phenotypes similar to those of the transgenic fruits.Furthermore,we have demonstrated that expression of cell expansion and GA responsive genes were downregulated in transgenic tomato fruits,supporting that overexpression of the SlGA2ox1 leads to reduction in endogenous GAs.This study provides additional evidence that endogenous GAs and the SlGA2ox1 gene play an important role in controlling on fruit weight,seed development and germination in tomato plant. | Shen Chen Xiaojing Wang Liying Zhang Shanshan Lin Decai Liu Quanzhi Wang Shanya Cai Rania El-Tanbouly Lijun Gan Han Wu Yi Li | 2016 | Horticulture Research2016,3,1: | 3 |

| 6 | Theoretical Analysis and Experimental Study on Breakaway Torque of Large-diameter Magnetic Liquid Seal at Low Temperature显示文摘The existing researches of the magnetic liquid rotation seal have been mainly oriented to the seal at normal temperature and the seal with the smaller shaft diameter less than 100 mm. However, the large-diameter magnetic liquid rotation seal at low temperature has not been reported both in theory and in application up to now. A key factor restricting the application of the large-diameter magnetic liquid rotation seal at low temperature is the high breakaway torque. In this paper, the factors that influence the breakaway torque including the number of seal stages, the injected quantity of magnetic liquid and the standing time at normal temperature are studied. Two kinds of magnetic liquid with variable content of large particles are prepared first, and a seal feedthrough with 140 mm shaft diameter is used in the experiments. All experiments are carried out in a low temperature chamber with a temperature range from 200℃ to -100℃. Different numbers of seal stages are tested under the same condition to study the relation between the breakaway torque and the number of seal stages. Variable quantity of magnetic liquid is injected in the seal gap to get the relation curve of the breakaway torque and the injecting quantity of magnetic liquid. In the experiment for studying the relation between the breakaway torque and the standing time at the normal temperature, the seal feedtrough is laid at normal temperature for different period of time before it is put in the low temperature chamber. The experimental results show that the breakaway torque is proportional to the number of seal stages, the injected quantity of magnetic liquid and the standing time at the normal temperature. Meanwhile, the experimental results are analyzed and the torque formula of magnetic liquid rotation seal at low temperature is deduced from the Navier-Stokes equation on the base of the model of magnetic liquid rotation seal. The presented research can make wider application of the magnetic liquid seal in general. | 7 | An efficient papaya leaf distortion mosaic potyvirus vector for virus-induced gene silencing in papaya显示文摘Papaya(Carica papaya L.)is regarded as an excellent model for genomic studies of tropical trees because of its short generation time and its small genome that has been sequenced.However,functional genomic studies in papaya depend on laborious genetic transformations because no rapid tools exist for this species.Here,we developed a highly efficient virus-induced gene silencing(VIGS)vector for use in papaya by modifying an artificially attenuated infectious clone of papaya leaf distortion mosaic virus(PLDMV;genus:Potyvirus),PLDMV-E,into a stable Nimble Cloning(NC)-based PLDMV vector,pPLDMV-NC,in Escherichia coli.The target fragments for gene silencing can easily be cloned into pPLDMV-NC without multiple digestion and ligation steps.Using this PLDMV VIGS system,we silenced and characterized five endogenous genes in papaya,including two common VIGS marker genes,namely,phytoene desaturase,Mg-chelatase H subunit,putative GIBBERELLIN(GA)-INSENSITIVE DWARF1A and 1B encoding GA receptors;and the cytochrome P450 gene CYP83B1,which encodes a key enzyme involved in benzylglucosinolate biosynthesis.The results demonstrate that our newly developed PLDMV VIGS vector is a rapid and convenient tool for functional genomic studies in papaya. | Decai Tuo Pu Yan Guangyuan Zhao Hongguang Cui Guopeng Zhu Yang Liu Xiukun Yang He Wang Xiaoying Li Wentao Shen Peng Zhou | 2021 | Horticulture Research2021,8,1: | 2 |

| 15 | Evolution and roles of cytokinin genes in angiosperms 1:Do ancient IPTs play housekeeping while non-ancient IPTs play regulatory roles?显示文摘Isopentenyltransferase(IPT)genes,including those encoding ATP/ADP-IPTs and tRNA-IPTs,control the rate-limiting steps of the biosynthesis of N6-(Δ2-isopentenyl)adenine(iP)-type and trans-zeatin(tZ)-type cytokinins and cis-zeatin(cZ)-type cytokinins,respectively.However,the evolution and roles of these IPTs in angiosperms are not well understood.Here,we report comprehensive analyses of the origins,evolution,expression patterns,and possible roles of ATP/ADP-IPTs and tRNA-IPTs in angiosperms.We found that Class I and II tRNA-IPTs likely coexisted in the last common ancestor of eukaryotes,while ATP/ADP-IPTs likely originated from a Class II tRNA-IPT before the divergence of angiosperms.tRNA-IPTs are conservatively retained as 2–3 copies,but ATP/ADP-IPTs exhibit considerable expansion and diversification.Additionally,tRNA-IPTs are constitutively expressed throughout the plant,whereas the expression of ATP/ADP-IPTs is tissue-specific and rapidly downregulated by abiotic stresses.Furthermore,previous studies and our present study indicate that ATP/ADP-IPTs and their products,iPs/tZs,may regulate responses to environmental stresses and organ development in angiosperms.We therefore hypothesize that tRNA-IPTs and the associated cZs play a housekeeping role,whereas ATP/ADP-IPTs and the associated iP/tZ-type cytokinins play regulatory roles in organ development and stress responses in angiosperms,which echoes the conclusions and hypothesis presented in the accompanying study by Wang,X.et al Evolution and roles of cytokinin genes in angiosperms 2:Do ancient CKXs play housekeeping roles while non-ancient CKXs play regulatory roles?Hortic Res http://gffzzd3cc09b8251d45dfsoqqwqw0vuffv60n5.ffgz.tsg.suse.edu.cn/10.1038/s41438-020-0246-z. | Xiaojing Wang Shanshan Lin Decai Liu Lijun Gan Richard McAvoy Jing Ding Yi Li | 2020 | Horticulture Research2020,7,1: | 1 |
| 16 | Evolution and roles of cytokinin genes in angiosperms 2:Do ancient CKXs play housekeeping roles while non-ancient CKXs play regulatory roles?显示文摘Cytokinin oxidase/dehydrogenase(CKX)is a key enzyme responsible for the degradation of endogenous cytokinins.However,the origins and roles of CKX genes in angiosperm evolution remain unclear.Based on comprehensive bioinformatic and transgenic plant analyses,we demonstrate that the CKXs of land plants most likely originated from an ancient chlamydial endosymbiont during primary endosymbiosis.We refer to the CKXs retaining evolutionarily ancient characteristics as“ancient CKXs”and those that have expanded and functionally diverged in angiosperms as“non-ancient CKXs”.We show that the expression of some non-ancient CKXs is rapidly inducible within 15 min upon the dehydration of Arabidopsis,while the ancient CKX(AtCKX7)is not drought responsive.Tobacco plants overexpressing a non-ancient CKX display improved oxidative and drought tolerance and root growth.Previous mutant studies have shown that non-ancient CKXs regulate organ development,particularly that of flowers.Furthermore,ancient CKXs preferentially degrade cis-zeatin(cZ)-type cytokinins,while non-ancient CKXs preferentially target N6-(Δ2-isopentenyl)adenines(iPs)and trans-zeatins(tZs).Based on the results of this work,an accompanying study(Wang et al.http://gffzzd3cc09b8251d45dfsoqqwqw0vuffv60n5.ffgz.tsg.suse.edu.cn/10.1038/s41438-019-0211-x)and previous studies,we hypothesize that non-ancient CKXs and their preferred substrates of iP/tZ-type cytokinins regulate angiosperm organ development and environmental stress responses,while ancient CKXs and their preferred substrates of cZs play a housekeeping role,which echoes the conclusions and hypothesis described in the accompanying report(Wang,X.et al.Evolution and roles of cytokinin genes in angiosperms 1:Doancient IPTs play housekeeping while non-ancient IPTs play regulatory roles?Hortic Res 7,(2020).http://gffzzd3cc09b8251d45dfsoqqwqw0vuffv60n5.ffgz.tsg.suse.edu.cn/10.1038/s41438-019-0211-x). | Xiaojing Wang Jing Ding Shanshan Lin Decai Liu Tingting Gu HanWu Robert N.Trigiano Richard McAvoy Jinling Huang Yi Li | 2020 | Horticulture Research2020,7,1: | 1 |
