1Toward targeting inflammasomes: insights into their regulation and activation显示文摘Inflammasomes are multi-component signaling complexes critical to the initiation of pyroptotic cell death in response to invading pathogens and cellular damage.A number of innate immune receptors have been reported to serve as inflammasome sensors.Activation of these sensors leads to the proteolytic activation of caspase-1,a proinflammatory caspase responsible for the cleavage of proinflammatory cytokines interleukin-1β and interleukin-18 and the effector of pyroptotic cell death, gasdermin D.Though crucial to the innate immune response to infection, dysregulation of inflammasome activation can lead to the development of inflammatory diseases, neurodegeneration, and cancer. Therefore, clinical interest in the modulation of inflammasome activation is swiftly growing. As such,it is imperative to develop a mechanistic understanding of the regulation of these complexes. In this review,we divide the regulation of inflammasome activation into three parts. We discuss the transcriptional regulation of inflammasome components and related proteins, the post-translational mechanisms of inflammasome activation, and advances in the understanding of the structural basis of inflammasome activation.Shelbi Christgen David E.Place Thirumala-Devi Kanneganti 2020Cell Research2020,30,4:17
2Metabolic regulation of pyroptotic cell death expands the therapeutic landscape for treating inflammatory disease显示文摘A recent study in Science by Fiachra Humphries et al.1 reveals an important role for the metabolic intermediate fumarate in reducing gasdermin D-mediated pyroptosis.Their work adds a new inhibitor to the growing toolkit with the potential for treating pyroptosis-driven inflammatory diseases.Pyroptosis is an inflammatory programmed cell death pathway initiated by direct sensing of pathogen-derived ligands or indirect sensing of changes to the normal cell state through sensors that can activate the inflammasome.David E.Place Thirumala-Devi Kanneganti 2021Signal Transduction and Targeted Therapy2021,6,2:1
3Intestinal tete-a-tete: helminths blunt immunity against flaviviruses显示文摘Intestinal helminth infections are known to alter host immune responses, but their impact on neurotropic flaviviruses is poorly understood.A recent study in Cell by Desai et al.finds that helminth infection promotes a tuft cell immune response, via IL-4,that increases susceptibility to subsequent challenge by the mosquito-transmitted flavivirus,West Nile virus.Helminth infection continues to affect〜1.5 billion people globally with significant consequences on human health. These infections cause intestinal disease, malnutrition, impaired growth and development, and altered susceptibility to other infections.David E.Place Thirumala-Devi Kanneganti 2021Cell Research2021,31,7:0
