| 3 | Invasion by exogenous RNA:cellular defense strategies and implications for RNA inference显示文摘Exogenous RNA poses a continuous threat to genome stability and integrity across various organisms.Accumulating evidence reveals complex mechanisms underlying the cellular response to exogenous RNA,including endo-lysosomal degradation,RNA-dependent repression and innate immune clearance.Across a variety of mechanisms,the natural anti-sense RNAdependent defensive strategy has been utilized both as a powerful gene manipulation tool and gene therapy strategy named RNA-interference(RNAi).To optimize the efficiency of RNAi silencing,a comprehensive understanding of the whole life cycle of exogenous RNA,from cellular entry to its decay,is vital.In this paper,we review recent progress in comprehending the recognition and elimination of foreign RNA by cells,focusing on cellular entrance,intracellular transportation,and immune-inflammatory responses.By leveraging these insights,we highlight the potential implications of these insights for advancing RNA interference efficiency,underscore the need for future studies to elucidate the pathways and fates of various exogenous RNA forms,and provide foundational information for more efficient RNA delivery methods in both genetic manipulation and therapy in different organisms. | Danxu Tang Yan Liu Chundi Wang Lifang Li Saleh AAl‑Farraj Xiao Chen Ying Yan | 2023 | Marine Life Science & Technology2023,5,4: | 1 |
| 4 | SOME BASIC PROBLEMS IN NUMERICALLY SIMULATING EFFECTIVE PROPERTIFS AND LOCAL FIELDS OF COMPOSITE MATERIALS显示文摘It is pointed out that to numerically estimate the effective properties and local fields ofmatrix-inclusion composites,a commonly adopted method is accompanied with some serious draw-backs.We call this method the nominal loading scheme(NLS),which considers the actual inclusiondistribution inside a finite domain,Ω say,treats the external domain of Ω to be of the pure matrix ma-terial,and imposes the actural traction,σ~∞ say on the remote boundary.It thus gives rise to the fol-lowing basic problems:(i)Can NLS be improved remarkably just by adjusting σ~∞?(it)What isthe relationship between the size of Ω and the,scale of inclusions?(iii)Which choice isbetter in calculating the effective properties,the whole domain Ω or an appropriatelyselected sub-domain of Ω? | 5 | Pro-osteogenesis and in vivo tracking investigation of a dental implantation system comprising novel mTi implant and HYH-Fe particles显示文摘Insufficient early osteogenesis seriously affects the later stage osteogenic quality and osseointegration of dental implants.To promote early osteogenesis,we first designed a Ti dental implant with a built-in magnet(mTi)to produce a local static magnetic field(SMF).Then,a dental implantation system comprising the mTi implant and the superparamagnetic hydroxyapatite(HA:Yb/Ho-Fe,named HYH-Fe)particles was implanted into the alveolar bone of beagles.The results showed that the mTi+HYH-Fe group displayed better early osteogenesis and later stage osseointegration than the Ti+HA and mTi+HA groups.A combination of the local SMF(mTi)and superparamagnetic HYH-Fe particles had a positive effect on the pro-osteogenesis of Ti implants.The results also indicated that week 10 could be adopted as the key time point to evaluate the early osteogenic effect of the mTi+HYH-Fe implantation system,which would be a promising prospect for promotion of osteogenesis,in vivo tracking investigation of material-bone relationships,and clinical applications. | Xiyu Li Juan Wu Danxue Li Qin Zou Yi Man Ling Zou Wei Li | 2021 | Bioactive Materials2021,6,9: | 0 |
| 6 | Hematopoietic Stem Cell Transplantation for Patients with Chronic Myeloid Leukemia显示文摘OBJECTIVE To evaluate the effects of autologous and allogeneic hematopoietic stem cell transplantation (HSCT) for patients with chronic myeloid leukemia(CML).METHODS Fifty-seven patients with CML were treated by HSCT, including 8 cases treated with autologous transplantation purged in vivo and in vitro of minimal residual disease (MRD), 39 cases with related donor allogeneic HSCT (allo-HSCT) and 10 cases with unrelated donor alIo-HSCT. The conditioning regimen was a TBI (total-body irradiation) +CY (cyclophosphamide, CTX) protocol in 32 patients, a modified BuCY (hydroxyurea, busulfan, Ara-C, CTX) protocal in 24 patients, and a MACC ( Melphalan, Ara-C, CTX and chlorethyl cyclohexyl nitrosourea ) protocol in one patient. Cyclosporine (CsA) and methotrexate (MTX) were used in patients with related-donor allo-HSCT, and CsA and MTX were added to mycophenolate mofetil (MMF) and antithymocyte globulin (ATG) in unrelated donor allo-HSCT for graft versus host disease (GVHD) prophylaxis. Otherwise, CsA was only used for GVHD prophylaxis in patients with accelerated phase (AP) and blast crisis (BC). The Kaplan-Meier survival analysis model was used to estimate the overall survival (OS) and the disease-free survival (DSF) at 5 years after transplantation.RESULTS Eight patients with autologous transplantation, except for 1 case who died of transplantation-related complications, obtained cytogenetic part or complete remission (CR) within 3 months after transplantation. One patient, who was in BC and obtained CR in hematology before transplantation, had been in molecular CR for 92 months after autologous transplantation. Among the 49 patients treated with alIo-HSCT, all obtained CR, except for one patient who died of hepatic veno-occlusive disease (VOD) and one who had not obtained CR. The incidence of infection and VOD was 33.3% and 7.0%, respectively, during transplantation. After transplantation the incidence of hemorrhagic cystitis (HC) and cytomegalovirus (CMV) interstitial pneumonia (IP) was 22.8% and 8.8%, respectively. VOD, HC and CMV IP did not occur in patients with autologous transplantation. The incidence of acute GVHD and the frequency of chronic GVHD was 41.0% and 48.6%, respectively, in patients with related and unrelated transplantation. The rate of relapse in patients with autologous and allogeneic transplantation was 57.1% and 12.8%, respectively. The DFS at 5 years after transplantation was 25.0% and 61.7%, respectively, in patients with autologous and related donor transplantation. The DFS at 5 years was 70.7% and 34.1%, respectively, in patients with CP (chronic phase) or AP and BC before transplantation.CONCLUSION AIIo-HSCT may have a higher clinical cure rate for CML patients with CP. | 7 | Induction of the Sulfolobus shibatae virus SSV1 DNA replication by mitomycin C显示文摘The temperate virus SSV1 from the hyper-thermophilic archaeon Sulfolobus shibatae provides a useful model system for the study of archaeal DNA replication. Southern hybridization showed that SSV1 existed primarily as a provirus in its host that was grown without shaking. Upon UV or mitomycin C induction, the cellular level of free SSV1 DNA increased drastically whereas that of integrated viral DNA remained unchanged. The results of mitomycin C induction were more reproducible than those of UV induction. We found that, when the cells that had been grown without shaking were shaken, the replication of SSV1 DNA was also induced. Based on our results, we developed a method for the induction of SSY1 DNA replication by mitomycin C. When the S. shibatae virus production was induced using this method, the cellular level of free SSV1 DNA started to increase 10 h after induction, and peaked after 12-15 h.
