1香港华人妇女产后抑郁危险因素的前瞻性研究显示文摘目的:产后抑郁是一种常见的精神障碍,对产妇、新生儿和其它家庭成员导致广泛的伤害。产后抑郁的病因学研究大多是基于西方样本,对其中的社会文化原因也缺乏实证研究。本研究从人口学、心理社会及种族文化因素等方面探讨中国妇女产后抑郁症状学的决定因素。方法:在大学附属综合医院产前门诊连续收集登记的中国孕妇959例,从她们第一次产前检查开始评估(基线),以后分别在孕期的最后3个月、产后即刻和产后3个月时评估。调查涉及的危险因素分6个方面:人口学和社会经济背景,躯体疾病及精神疾病病史, 产前抑郁情绪,围生期应激源,人际关系,和种族文化环境。采用等级回归进行分析。因变量是产后3个月时的抑郁评分。结果:决定产后抑郁症状学的因素有:目前的生活事件,缺乏社会支持,婚姻不满意,既往有故意自伤史,以及产前存在抑郁情绪。婆媳关系不好和没有陪月在产后阶段也与抑郁评分高有关联。结论:西方研究所确立的产后抑郁危险因素大体适用于中国妇女。产褥期的社会文化特点同样对产妇情绪有影响。Dominic TS Lee Alexander SK Yip Sandra SM Chan Fiona FY Lee Tony YS Leung Tony KH Chung 2005中国心理卫生杂志2005,19,9:50

3Boceprevir,GC-376,and calpain inhibitors Ⅱ,Ⅻ inhibit SARS-CoV-2 viral replication by targeting the viral main protease显示文摘A new coronavirus SARS-CoV-2,also called novel coronavirus 2019(2019-nCoV),started to circulate among humans around December 2019,and it is now widespread as a global pandemic.The disease caused by SARS-CoV-2 virus is called COVID-19,which is highly contagious and has an overall mortality rate of 6.35%as of May 26,2020.There is no vaccine or antiviral available for SARS-CoV-2.In this study,we report our discovery of inhibitors targeting the SARS-CoV-2 main protease(Mpro).Using the FRET-based enzymatic assay,several inhibitors including boceprevir,GC-376,and calpain inhibitorsⅡ,andⅫwere identified to have potent activity with single-digit to submicromolar ICs0 values in the enzymatic assay.The mechanism of action of the hits was further characterized using enzyme kinetic studies,thermal shift binding assays,and native mass spectrometry.Significantly,four compounds(boceprevir,GC-376,calpain inhibitorsⅡandⅫ)inhibit SARS-CoV-2 viral replication in cell culture with EC50 values ranging from 0.49 to 3.37μM.Notably,boceprevir,calpain inhibitorsⅡandⅫrepresent novel chemotypes that are distinct from known substrate-based peptidomimetic Mpro inhibitors.A complex crystal structure of SARS-CoV-2 Mpro with GC-376,determined at 2.15(A)resolution with three protomers per asymmetric unit,revealed two unique binding configurations,shedding light on the molecular interactions and protein conformational flexibility underlying substrate and inhibitor binding by Mpro.Overall,the compounds identified herein provide promising starting points for the further development of SARS-CoV-2 therapeutics.Chunlong Ma Michael Dominic Sacco Brett Hurst Julia Alma Townsend Yanmei Hu Tommy Szeto Xiujun Zhang Bart Tarbet Michael Thomas Marty Yu Chen Jun Wang 2020Cell Research2020,30,8:13
4人肿瘤细胞核苷酸切除修复蛋白表达与抗癌药耐药的相关性(英文)显示文摘背景与目的:核苷酸切除修复(Nucleotideexcisionrepair,NER)是真核细胞中的DNA修复多酶系统,它可能与人肿瘤细胞对抗癌药的耐药有关。本实验将探讨NER蛋白(XPA,XPB,XPD和ERCC1)的表达与人肿瘤细胞耐药的关系。方法:采用westernblot检测美国国家癌症研究所(NationalCancerInstitute,NCI)用于抗癌药筛选的60株人肿瘤细胞的ERCC1,XPA,XPBXPD表达,并与170种抗癌药物的细胞毒试验结果进行相关性分析。结果:ERCC1,XPB和XPD的蛋白表达与抗癌药物敏感性呈负相关,在170种抗癌药物中分别有13,17和32种的耐药性与上述蛋白水平相关性显著或非常显著(P<0.05)。而XPA的表达与药物敏感性无明显正/负相关。根据已确定的6种药物作用机理分析,肿瘤细胞XPD表达与其对烷化剂类抗癌药耐药相关性显著。结论:本实验结果肿瘤细胞的XPD蛋白表达与烷化剂类抗癌药的耐药相关,提示XPD在肿瘤细胞耐药过程中起重要作用。陈忠平 Areti MALAPETSA Anne MONKS Timothy G. 5Nano zinc, an alternative to conventional zinc as animal feed supplement:A review显示文摘The uniqueness of Zn is that, it is the second most abundant trace element in the animal body but can't be stored in the body, thus regular dietary intake is required. Zinc oxide(ZnO) nanoparticles(NP) particles are being extensively used in paints, skin lotions pigments, food, electronics appliances, biological and pharmaceutical applications and many more. Zinc oxide nanoparticles are the specially prepared mineral salt having particle size of 1 to 100 nm. It promotes growth can act as antibacterial agent,modulates the immunity and reproduction of the animals. Both in lower and higher doses of specifications it has exhibited a variety of effects on animal performances. Apart from being highly bioavailable, reports have already pointed out the growth promoting, antibacterial, immuno-modulatory and many more effects of nano zinc(nZn). These can be used at lower doses and can provide better result than the conventional Zn sources and indirectly prevents environmental contamination also. The toxicological studies provide mixed results in animal models. Studies been undertaken in diversified animal species and encouraging effects have been reported with nZn supplementation. However, there is a need to optimize the dose and duration of ZnO NP supplementation for human and livestock,depending on its biological effects. 6Dysregulation of innate immunity in ulcerative colitis patients who fail anti-tumor necrosis factor therapy显示文摘AIM To study the innate immune function in ulcerative colitis(UC) patients who fail to respond to anti-tumor necrosis factor(TNF) therapy.METHODS Effects of anti-TNF therapy, inflammation and medications on innate immune function were assessed by measuring peripheral blood mononuclear cell(PBMC) cytokine expression from 18 inflammatory bowel disease patients pre- and 3 mo post-anti-TNF therapy. Toll-like receptor(TLR) expression and cytokine production post TLR stimulation was assessed in UC 'responders'(n = 12) and 'non-responders'(n = 12) and compared to healthy controls(n = 12). Erythrocyte sedimentation rate(ESR) and C-reactive protein(CRP) levels were measured in blood to assess disease severity/activity and inflammation. Pro-inflammatory(TNF, IL-1β, IL-6), immuno-regulatory(IL-10), Th1(IL-12, IFNγ) and Th2(IL-9, IL-13, IL-17A) cytokine expression was measured with enzyme-linked immunosorbent assay while TLR cellular composition and intracellular signalling was assessed with FACS.RESULTS Prior to anti-TNF therapy, responders and nonresponders had similar level of disease severity and activity. PBMC's ability to respond to TLR stimulation was not affected by TNF therapy, patient's severity of the disease and inflammation or their medication use. At baseline, non-responders had elevated innate but not adaptive immune responses compared to responders(P < 0.05). Following TLR stimulation, nonresponders had consistently reduced innate cytokine responses to all TLRs compared to healthy controls(P < 0.01) and diminished TNF(P < 0.001) and IL-1β(P < 0.01) production compared to responders. This innate immune dysfunction was associated with reduced number of circulating plasmacytoid dendritic cells(p DCs)(P < 0.01) but increased number of CD4+ regulatory T cells(Tregs)(P = 0.03) as well as intracellular accumulation of IRAK4 in non-responders following TLR-2,-4 and-7 activation(P < 0.001). CONCLUSION Reduced innate immunity in non-responders may explain reduced efficacy to anti-TNF therapy. 7不同施肥方式对马铃薯农田土壤有机碳组分和碳库管理指数的影响显示文摘针对马铃薯生产实践中长期单施化肥或化肥过量施用等不合理施肥措施导致土壤碳库活性变差和土壤生物活性降低等问题。通过田间定位试验,分析不同施肥方式:不施肥(CK)、单施化肥(NPK)、单施有机肥(M)、有机无机肥配施(NPKM)对旱作马铃薯农田土壤有机碳组分和碳库管理指数的影响。结果显示,连续施肥5年后添加有机肥的M、NPKM处理较CK和NPK处理显著提高了土壤闭蓄态颗粒有机碳(OPOC)和土壤颗粒态有机碳(POC)含量,降低了土壤矿质结合态有机碳(MOC)含量。与CK和NPK处理相比,M、NPKM处理闭蓄态颗粒有机碳(OPOC)含量分别提高了32.52%、30.39%和27.09%、24.78%;土壤颗粒态有机碳(POC)含量分别提高了31.52%、29.65%和25.18%、23.15%。添加有机肥的M和NPKM处理显著提高了颗粒态有机碳(POC)的比例,降低了MOC的比例,与CK和NPK相比,M、MNPK处理中POC的比例分别增加了27.72%、25.23%和10.61%、7.54%,MOC的比例分别降低39.42%、33.16%和14.97%、9.81%。添加有机肥显著提高了土壤总有机碳(TOC)、微生物量碳(MBC)、易氧化有机碳(ROC)的含量,与CK处理相比,M和NPKM处理的TOC分别增加了15.28%和13.64%,MBC增加了21.82%和19.17%,ROC的含量分别增加了32.45%和31.35%;施用有机肥和有机无机配施均显著提高碳库管理指数(CPMI)较单施化肥分别提高28.8%和35.65%。综上所述,施用有机肥处理(M处理和MNPK处理)显著地提高了马铃薯农田土壤闭蓄态颗粒有机碳(OPOC)、颗粒态有机碳(POC)和总有机碳(TOC)及微生物量碳(MBC)、易氧化有机碳(ROC)含量,提高了土壤碳库管理指数;即马铃薯栽培施用有机肥有利于土壤活性有机碳的积累、能够改变土壤有机碳组分分布特征。郭亚军 邱慧珍 张玉娇 张建斌 王友玲 张春红 Anning Dominic Kwadwo 2021土壤通报2021,52,4:10

10Establishing the presence or absence of chronic kidney disease:Uses and limitations of formulas estimating the glomerular filtration rate显示文摘The development of formulas estimating glomerular filtration rate(eG FR) from serum creatinine and cystatin C and accounting for certain variables affecting the production rate of these biomarkers, including ethnicity, gender and age, has led to the current scheme of diagnosing and staging chronic kidney disease(CKD),which is based on e GFR values and albuminuria.This scheme has been applied extensively in various populations and has led to the current estimates of prevalence of CKD. In addition, this scheme is applied in clinical studies evaluating the risks of CKD and the efficacy of various interventions directed towards improving its course. Disagreements between creatinine-based and cystatin-based e GFR values and between e GFR values and measured GFR have been reported in various cohorts. These disagreements are the consequence of variations in the rate of production and in factors, other than GFR, affecting the rate of removal of creatinine and cystatin C. The disagreements create limitations for all e GFR formulas developed so far. The main limitations are low sensitivity in detecting early CKD in several subjects, e.g., those with hyperfiltration, and poor prediction of the course of CKD. 11针刺作为辅助手段治疗精神分裂症的探索性病例研究(英文)显示文摘目的:探索个体针刺疗法作为常规治疗的辅助手段在精神分裂症患者中的效果;采用探索性病例研究的方法,使用定量和定性评价工具,以便能够对针刺治疗此类患者的确切疗效作出科学评价,并对未来采用针刺治疗此类患者的试验设计提供方法学建议。方法:共纳入11例精神分裂症患者,在予以10周的针刺治疗前,治疗过程中及治疗后分别采用多种有效的定量和定性评价工具对患者进行多方面的评价。这些评价手段包括对针刺治疗、全科医生及精神科医生病历的整理回顾。分析定性资料以评价受试者向研究者及其临床医生和护理人员报告的内容的可靠性。结果:11名受试者中共有8名完成了1个疗程的针刺治疗,全部11名受试者都报告了针刺治疗的积极作用,包括精神分裂症症状的改善,缓解药物的副作用,对精力、内在动机、睡眠、成瘾症状及其他身体机能的促进作用。但是,受试者向研究者和针刺医生报告的信息在不同治疗阶段有一定差别,且往往不一致;在研究结束后,受试者向研究者报告了更多的信息。结论:本研究表明,在常规疗法之外辅以针刺治疗能够给精神分裂症病人带来一定益处。对数据的三角测量表明了受试者报告内容的某些不一致性,而这些可以通过多种研究方法的混合使用得以克服。通过对数据的比较,推荐在将来的研究设计中使用阳性与阴性症状量表、针刺临床试验干预措施报告标准以及在问卷中增加对药物副作用、运动、睡眠以及日常作息的调查。在将来的研究中还应充分考虑到,恢复期患者容易复发,应该让他们可以方便地获得帮助。Patricia Ronan Nicola Robinson Dominic Harbinson Douglas MacInnes 2011中西医结合学报2011,9,5:6

13Interplay between microstructural evolution and tribo-chemistry during dry sliding of metals显示文摘Understanding the microstructural and tribo-chemical processes during tribological loading is of utmost importance to further improve the tribological behavior of metals. In this study, the friction, wear and tribo-chemical behavior of Ni with different initial microstructures(nanocrystalline, bi-modal, coarse-grained) is investigated under dry sliding conditions. In particular, the interplay be-tween frictional response, microstructural evolution and tribo-oxidation is considered. Friction tests are carried out using ball-on-disk experiments with alumina balls as counter-bodies, varying the load between 1 and 5 N. The microstructural evolution as well as the chemical reactions beneath the samples’ surface is investigated by means of cross-sections. The samples with finer microstructures show a faster run-in and lower maximum values of the coefficient of friction(COF) which can be attributed to higher oxidation kinetics and a higher hardness. It is observed that with increasing sliding cycles, a stable oxide layer is formed. Furthermore, initially coarse-grained samples show grain refinement, whereas initially finer microstructures undergo grain coarsening converging towards the same superficial grain size after 2,000 sliding cycles. 16Antioxidant peptides encrypted in flaxseed proteome:An in silico assessment显示文摘Flaxseed proteins and antioxidant peptides(AP)encrypted in their sequences were analysed in silico with a range of bioinformatics tools to study their physicochemical properties,allergenicity,and toxicity.Nine proteases(digestive,plant and microbial sources)were assessed for their ability to release known APs from 23 mature flaxseed storage proteins using the BIOPEP database.The families of proteins identified were predominantly globulins,oleosins,and small amount of conlinin.Overall,253 APs were identified from these proteins.More peptides were released by enzymatic hydrolysis from the globulins than those from oleosins and conlinin.Compared with other enzymes studied,the plant proteases(papain,ficin,and bromelain)were found to be superior to releasing APs from the flaxseed proteins.Analysis of toxicity by ToxinPred showed that none of the peptides released was toxic.Most of the APs showed structural features that are important for antioxidation,including relatively low molecular weight(dipeptides and tripeptides only);amphipathic properties(hydrophobicity range of-0.5 to+0.5);relatively low Boman index(≤2);broad range of pI(3.7-10.8),and an abundance of antioxidant amino acid residues(e.g.glutamic acid and histidine).This study demonstrate the suitability of flaxseed proteins as a source of APs.Dawei Ji Chibuike C.Udenigwe Dominic Agyei 2019Food Science and Human Wellness2019,8,3:5

18Modeling of fiber bridging in fluid flow for well stimulation applications显示文摘Accurate acid placement constitutes a major concern in matrix stimulation because the acid tends to penetrate the zones of least resistance while leaving the low-permeability regions of the formation untreated.Degradable materials(fibers and solid particles)have recently shown a good capability as fluid diversion to overcome the issues related to matrix stimulation.Despite the success achieved in the recent acid stimulation jobs stemming from the use of some products that rely on fiber flocculation as the main diverting mechanism,it was observed that the volume of the base fluid and the loading of the particles are not optimized.The current industry lacks a scientific design guideline because the used methodology is based on experience or empirical studies in a particular area with a particular product.It is important then to understand the fundamentals of how acid diversion works in carbonates with different diverting mechanisms and diverters.Mathematical modeling and computer simulations are effective tools to develop this understanding and are efficiently applied to new product development,new applications of existing products or usage optimization.In this work,we develop a numerical model to study fiber dynamics in fluid flow.We employ a discrete element method in which the fibers are represented by multi-rigid-body systems of interconnected spheres.The discrete fiber model is coupled with a fluid flow solver to account for the inherent simultaneous interactions.The focus of the study is on the tendency for fibers to flocculate and bridge when interacting with suspending fluids and encountering restrictions that can be representative of fractures or wormholes in carbonates.The trends of the dynamic fiber behavior under various operating conditions including fiber loading,flow rate and fluid viscosity obtained from the numerical model show consistency with experimental observations.The present numerical investigation reveals that the bridging capability of the fiber–fluid system can be enhanced by increasing the fiber loading,selecting fibers with higher stiffness,reducing the injection flow rate,reducing the suspending fluid viscosity or increasing the attractive cohesive forces among fibers by using sticky fibers.Mehdi Ghommem Mustapha Abbad Gallyam Aidagulov Steve Dyer Dominic Brady 2020Petroleum Science2020,17,3:4
19A natural model of behavioral depression in postpartum adult female cynomolgus monkeys (Macaca fascicularis)显示文摘Within the postpartum period, both mothers and infants are susceptible; but because PPD typically occurs for short durations and has moderate symptoms, there exists challenges in exploring and addressing the underlying cause of the depression. This fact highlights the need for relevant animal models. In the present study, postpartum adult female cynomolgus monkeys(Macaca fascicularis) living in breeding groups were observed for typical depressive behavior. The huddle posture behavior was utilized as an indicator of behavioral depression postpartum(BDP) as it has been established as the core depressive-like behavior in primates. Monkeys were divided into two groups: A BDP group(n=6), which were found to spend more time huddling over the first two weeks postpartum than other individuals that formed a non-depression control group(n=4). The two groups were then further analyzed for locomotive activity, stressful events, hair cortisol levels and for maternal interactive behaviors. No differences were found between the BDP and control groups in locomotive activity, in the frequencies of stressful events experienced and in hair cortisol levels. These findings suggested that the postpartum depression witnessed in the monkeys was not related to external factors other than puerperium period. Interestingly, the BDP monkeys displayed an abnormal maternal relationship consisting of increased infant grooming. Taken together, these findings suggest that the adult female cynomolgus monkeys provide a natural model of behavioral postpartum depression that holds a number of advantages over commonly used rodent systems in PPD modeling. The cynomolgus monkeys have a highly-organized social hierarchy and reproductive characteristics without seasonal restriction—similar to humans—as well as much greater homology to humans than rodents. 20二甲双胍与吡格列酮两种2型糖尿病药物在亲水作用色谱上的保留机理研究(英文)显示文摘二甲双胍和吡格列酮是治疗2型糖尿病的常见药物,经常用于同一复方制剂。制药和临床领域常要求同时测定这两种药物。亲水作用色谱近来在医药分析领域展现出良好的前景。本文通过研究流动相的有机溶剂含量、pH值和缓冲盐浓度等参数对色谱保留的影响,从而尝试揭示两种药物在亲水性硅胶柱上的分离机理。流动相中甲醇的增加会导致二甲双胍的保留因子的增加。流动相pH值比较低时,二甲双胍的保留因子会随着pH值升高而增加,而当pH值>6.09,其反而会随着pH值的升高而下降。吡格列酮的保留因子几乎不受甲醇比例和pH值的影响。缓冲盐浓度对两种药物的保留因子几乎都没有影响。因此,对于二甲双胍,亲水作用是主要的保留机理,静电作用和氢键也起到了重要作用。对于吡格列酮,亲水作用是支配性的。经过优化的亲水作用色谱方法可成功地应用到分离测定两种药物在2型糖尿病复方药物中的含量。鲁肖飞 Caroline Martin 戚忠林 Pari Samani Zemen Berhe Dominic B Siaw Shyam K Pahari 左跃钢 2019分析科学学报2019,35,6:4
