2Biliary fistula after treatment for hydatid disease of the liver:When to intervene显示文摘AIM:To determine the outcome of patients with biliary fistula(BF)after treatment for hydatid disease of the liver. METHODS:Between January 2000 and December 2010,out of 301 patients with a diagnosis of hydatid cyst of the liver,282 patients who underwent treatment [either surgery or puncture,aspiration,injection and reaspiration(PAIR)procedure]were analysed.Patients were grouped according to the presence or absence of postoperative biliary fistula(PBF)(PBF vs no-PBF groups,respectively).Preoperative clinical,radiological and laboratory characteristics,operative characteristics including type of surgery,peroperative detection of BF,postoperative drain output,morbidity,mortality and length of hospital stays of patients were compared amongst groups.Multivariate analysis was performed to detect factors predictive of PBF.Receiver operative characteristics(ROC)curve analysis were used to determine ideal cutoff values for those variables found to be significant.A comparison was also made between patients whose fistula closed spontaneously(CS)and those with intervention in order to find predictive fac-tors associated with spontaneous closure. RESULTS:Among 282 patients[median(range)age, 23(16-78)years;77.0%male];210(74.5%)were treated with conservative surgery,33(11.7%)radical surgery and 39(13.8%)underwent percutaneous drainage with PAIR procedure A PBF developed in 46(16.3%) patients,all within 5 d after operation.The maximum cyst diameter and preoperative alkaline phosphatase levels(U/L)were significantly higher in the PBF group than in the no-PBF group[10.5±3.7 U/L vs 8.4±3.5 U/L(P<0.001)and 40.0±235.1 U/Lvs 190.0±167.3 U/L(P=0.02),respectively].Hospitalization time was also significantly longer in the PBF group than in the no-PBF group[37.4±18.0 d vs 22.4±17.9 d(P< 0.001)].A preoperative high alanine aminotransferase level(>40 U/L)and a peroperative attempt for fistula closure were significant predictors of PBF development (P=0.02,95%CI:-0.03-0.5 and P=0.001,95%CI:0.1-0.4),respectively.Comparison of patients whose PBF CS or with biliary intervention(BI)revealed that the mean diameter of the cyst was not significantly different between CS and BI groups however maximum drain output was significantly higher in the BI group(81.6± 118.1 cm vs 423.9±298.4 cm,P<0.001).Time for fistula closure was significantly higher in the BI group(10.1 ±3.7 d vs 30.7±15.1 d,P<0.001).The ROC curve analysis revealed cut-off values of a maximum bilious drainage<102 mL and a waiting period of 5.5 postoperative days for spontaneous closure with the sensitivity and specificity values of(83.3%-91.1%,AUC:0.90)and (97%-91%,AUC:0.95),respectively.The multivariate analysis demonstrated a PBF drainage volume<102 mL to be the only statistically significant predictor of spontaneous closure(P<0.001,95%CI:0.5-1.0). 4Epigenetics of proteasome inhibition in the liver of rats fed ethanol chronically显示文摘AIM:To examine the effects of ethanol-induced proteasome inhibition,and the effects of proteasome inhibition in the regulation of epigenetic mechanisms. METHODS:Rats were fed ethanol for 1 mo using the Tsukamoto-French model and were compared to rats given the proteasome inhibitor PS-341(Bortezomib, Velcade?)by intraperitoneal injection.Microarray analysis and real time PCR were performed and proteasome activity assays and Western blot analysis were performed using isolated nuclei. RESULTS:Chronic ethanol feeding caused a significant inhibition of the ubiquitin proteasome pathway in the nucleus,which led to changes in the turnover of transcriptional factors,histone-modifying enzymes, and,therefore,affected epigenetic mechanisms. Chronic ethanol feeding was related to an increase in histone acetylation,and it is hypothesized that the proteasome proteolytic activity regulated histone modifications by controlling the stability of histone modifying enzymes,and,therefore,regulated the chromatin structure,allowing easy access to chromatin by RNA polymerase,and,thus,proper gene expression.Proteasome inhibition by PS-341 increased histone acetylation similar to chronic ethanol feeding. In addition,proteasome inhibition caused dramatic changes in hepatic remethylation reactions as there was a significant decrease in the enzymes responsible for the regeneration of S-adenosylmethionine,and, in particular,a significant decrease in the betaine- homocysteine methyltransferase enzyme.This suggested that hypomethylation was associated with proteasome inhibition,as indicated by the decrease in histone methylation. 5Mathematical, Mathematics Educational, and Educational Values in Mathematical Modeling Tasks显示文摘Purpose:The purpose of this study is to explore the mathematical values,mathematics educational values,and educational values invoIved in mathematical modeling tasks based on different mathematical modeling perspectives.Design/Approach/Methods:In this con text,the prese nt study is a qualitative research based on document analysis.The data were analyzed usingsemantic content analysis,and the selected modeling tasks based on different mathematical modeling perspectives were examined at the sentence level.Findings:Control,mystery,and openness mathematical values appeared in all mathematical modeling tasks,and rationalism and objectivism mathematical values appeared in realistic/applied and socio-critical modeli ng perspectives.Product,exploration,creating,releva nee,pleasure,and application mathematics educational values also emerged in all modeling tasks.Educational values of social justice,equity,social welfare,humanity,and altruism were more important in the socio critical modeling,while the value of individualism was more emphasized in the model-eliciting approach.Originality/Value:By determining mathematical,mathematics educational,and educational values involved in mathematical modeling tasks based on different mathematical modeling perspectives,an effective and more value-balaneed mathematical modeling instruction can be provided.Yiiksel Dede Veysel Akcakin Gürcan Kaya 2021ECNU Review of Education2021,4,2:4
6Non-contact tensile viscoelastic characterization of microscale biological materials显示文摘Many structures and materials in nature and physiology have important 'meso-scale' structures at the micron lengthscale whose tensile responses have proven difficult to characterize mechanically. Although techniques such as atomic force microscopy and micro-and nano-identation are mature for compression and indentation testing at the nano-scale, and standard uniaxial and shear rheometry techniques exist for the macroscale, few techniques are applicable for tensile-testing at the micrometre-scale, leaving a gap in our understanding of hierarchical biomaterials. Here, we present a novel magnetic mechanical testing(MMT) system that enables viscoelastic tensile testing at this critical length scale. The MMT system applies non-contact loading, avoiding gripping and surface interaction effects. 7Is hemodialysis a reason for unresponsiveness to hepatitis B vaccine? Hepatitis B virus and dialysis therapy显示文摘Impaired renal function is associated with a high risk of chronicity of hepatitis B virus(HBV) infection.Patients on hemodialysis(HD) or peritoneal dialysis are at an increased risk of viral transmission due to frequent necessity of blood product transfer as well as use of contaminated dialysate or dialysis materials.Additionally,health professionals may cause viral spread via contaminated hands and carelessness against hygiene rules.The frequency of chronic HBV infection may be as high as 80% in patients on renal replacement therapies.This is because HBV vaccination is essential to eliminate chronic HBV infection.However,response rates of HD patients to HBV vaccination vary between 10%-50%.Dialysis adequacy and early vaccination before the onset of dialysis therapy seem to be major determinants of high seroconversion rates.Older age,male gender,duration of dialysis therapy and nutritional status are other well-known factors associated with seroconversion rate.There are controversial reports regarding the role of the presence of diabetes mellitus,HCV positivity,erythropoietin resistance,hyperparathyroidism,and vitamin D inadequacy.The role of genetic alteration in the functions or production of cytokines still needs to be elucidated.Dede Sit Bennur Esen Ahmet Engin Atay Hasan Kayabasi 2015World Journal of Hepatology2015,7,5:3
8Rapid and user-friendly open-source CRISPR/Cas9 system for single-or multi-site editing of tomato genome显示文摘CRISPR/Cas9-induced genome editing is a powerful tool for studying gene function in a variety of organisms,including plants.Using multi-sgRNAs to target one or more genes is helpful to improve the efficacy of gene editing and facilitate multi-gene editing.Here,we describe a CRISPR/Cas9 system which can be conveniently developed as a CRISPR kit.SgRNA expression cassettes can be rapidly generated by one-step PCR using our CRISPR kit.In our kit,there are two binary vectors pHNCas9 and pHNCas9HT.The binary vector pHNCas9 was constructed to allow to assemble up to eight sgRNA expression cassettes by one-step Golden Gate cloning.Another binary vector pHNCas9HT can be used to generate a large number of single target constructs by directly transforming ligation reactions products into A.tumefaciens without several procedures,such as PCR and plasmid extraction.The two binary vectors are designed according to the principles of standard BioBrick assembly to be used as an open-source tool.For example,we used BioBrick as a visual T-DNA tag.We also developed a primer design aid to complement the system.With this primer design aid,researchers can rapidly obtain primers and GC content,and sgRNA sequence of target site.Our CRISPR/Cas9 system can perform single-and multi-site editing and multiple gene editing to produce various types of mutations in tomato.This rapid and user-friendly CRISPR/Cas9 system for tomato can be potentially used for mutagenesis of important crop species for genetic improvement and is suitable for research into the function of genes.Nan Hu Zhiqiang Xian Ning Li Yudong Liu Wei Huang Fang Yan Deding Su Jingxuan Chen Zhengguo Li 2019Horticulture Research2019,6,1:2

13SlGRAS4 accelerates fruit ripening by regulating ethylene biosynthesis genes and SlMADS1 in tomato显示文摘GRAS proteins are plant-specific transcription factors that play crucial roles in plant development and stress responses.However,their involvement in the ripening of economically important fruits and their transcriptional regulatory mechanisms remain largely unclear.Here,we demonstrated that SlGRAS4,encoding a transcription factor of the GRAS family,was induced by the tomato ripening process and regulated by ethylene.Overexpression of SlGRAS4 accelerated fruit ripening,increased the total carotenoid content and increased PSY1 expression in SlGRAS4-OE fruit compared to wild-type fruit.The expression levels of key ethylene biosynthesis genes(SlACS2,SlACS4,SlACO1,and SlACO3)and crucial ripening regulators(RIN and NOR)were increased in SlGRAS4-OE fruit.The negative regulator of tomato fruit ripening,SlMADS1,was repressed in OE fruit.Exogenous ethylene and 1-MCP treatment revealed that more endogenous ethylene was derived in SlGRAS4-OE fruit.More obvious phenotypes were observed in OE seedlings after ACC treatment.Yeast one-hybrid and dual-luciferase assays confirmed that SlGRAS4 can directly bind SlACO1 and SlACO3 promoters to activate their transcription,and SlGRAS4 can also directly repress SlMADS1 expression.Our study identified that SlGRAS4 acts as a new regulator of fruit ripening by regulating ethylene biosynthesis genes in a direct manner.This provides new knowledge of GRAS transcription factors involved in regulating fruit ripening.Yudong Liu Yuan Shi Deding Su Wang Lu Zhengguo Li 2021Horticulture Research2021,8,1:2
