| 1 | 己酮可可碱对肺结节病患者肺泡巨噬细胞释放细胞因子的作用显示文摘目的 研究己酮可可碱 (POF)对肺结节病患者肺泡巨噬细胞 (AM)产生细胞因子的作用 ,并与地塞米松 (DEX)的作用相比较。方法 收集 1 4例活动期肺结节病患者的AM ,以 1 0 %RPMI为培养液 (含有 1 0 %热灭活胎牛血清、2mmol/LL 谷氨酰胺、2 0 0kU/L青霉素及 2 0 0mg/L链霉素 ) ;或1 0 %RPMI加内毒素 (LPS ,1 0 0 μg/L) ;或分别加入浓度为 0 0 1mmol/L、0 1mmol/L和1mmol/L的POF ;或加入 0 1mmol/LDEX进行AM培养 2 4h。用酶联免疫吸附 (ELISA)法测定培养上清液中细胞因子含量。结果 POF对结节病患者AM自发释放的肿瘤坏死因子α(TNF α)有剂量依赖性抑制作用 (P <0 0 0 1 ) ,而对其他自发释放的细胞因子无影响。 0 1mmol/LDEX抑制自发释放的TNF α、可溶性肿瘤坏死因子受体 (sTNFR 2 )、白细胞介素 (IL) 1 β和IL 1 0 (P <0 0 0 1或 <0 0 5 或 <0 0 1 )。除sTNFR 1外 ,POF亦抑制这些由LPS刺激的AM释放的细胞因子 (P <0 0 5或 <0 0 0 1 )。与POF相似 ,0 1mmol/LDEX同样抑制这些LPS刺激的细胞因子释放 (P <0 0 5或 <0 0 0 1 ) ,但对sTNFR 1和IL 1 β无影响。 结论 与DEX相比 ,POF有更宽的治疗窗。用在结节病治疗上可以减少皮质激素用量或可将其替代。 | 童朝辉 代华平 陈宝敏 王辰 Josune Guzman Ulrich Costabel | 2003 | 中华结核和呼吸杂志2003，26，7： | 9 |

| 3 | IL-12I、L-18和TNF-α在外源性过敏性肺泡炎发病中的作用显示文摘目的从临床的角度出发、评价肺泡巨噬细胞(alveolar macrophages,AM)的产物白介素-12(IL-12)、白介素-18(IL-18)和肿瘤坏死因子-α(TNF-α)在外源性过敏性肺泡炎(extrinsic allergic alveolitis,EAA)炎症形成及发病中的作用。方法收集11例EAA患者和10例正常对照的AM,以10%RPMI(含有10%热灭活胎牛血清、2 mmol/LL-谷氨酰胺、200 kU/L青霉素及200mg/L链霉素)为培养液,加或不加内毒素(LPS,100μg/L)进行AM培养24 h。用ELISA方法测定培养上清液中细胞因子含量。结果与对照组相比,无论有无内毒素刺激,IL-18和TNF-α的水平在EAA患者中均明显增加(P<0.05或P<0.01)。EAA患者自发释放的IL-12的水平很低,内毒素刺激后明显升高(P<0.01)。结论IL-12、IL-18和TNF-α可能参与EAA炎症和肉芽肿形成过程,在其发病中起重要作用。 | 童朝辉 陈宝敏 王辰 Guzman Josune Costabel Ulrich | 2006 | 首都医科大学学报2006，27，1： | 3 |
| 4 | 己酮可可碱对外源性过敏性肺泡炎治疗作用的初步研究显示文摘目的 通过研究己酮可可碱 (POF)对外源性过敏性肺泡炎 (EAA)患者肺泡巨噬细胞(AM)产生的细胞因子的作用 ,探讨其治疗EAA的可能性 ,并与地塞米松 (DEX)的作用进行比较。方法 入选EAA患者 9例 ,通过支气管肺泡灌洗收AM ,并以 10 %RPMI为培养液或 10 %RPMI加内毒素 (LPS ,10 0 μg/L) ;或分别加入浓度为 0 0 1mmol/L、0 1mmol/L、1mmol/L的POF ;或加入 0 1mmol/LDEX进行AM培养 2 4h。用ELISA方法测定培养上清液中细胞因子含量。结果 POF可抑制EAA患者AM自发释放的TNFα和IL 10 ,此作用有剂量依赖关系 (P <0 0 0 1和P <0 0 5 )。POF对其他自发释放的细胞因子则无影响。 0 1mmol/LDEX只抑制自发释放的TNFα(P <0 0 5 )。除IL 1β和可溶性肿瘤坏死因子受体外 ,POF和 0 1mmol/LDEX均抑制LPS刺激的其他细胞因子的释放 (P <0 0 0 1或P <0 0 1或P <0 0 5 )。结论 POF对EAA的炎症有一定的抑制作用 ,然而POF治疗EAA及其他肺部疾病的临床价值 ,需要进一步的临床试验来评价。 | 童朝辉 陈宝敏 代华平 王辰 Josune Guzman Ulrich Costabel | 2004 | 中华医学杂志2004，84，6： | 2 |
