| 1 | Nuclear magnetic resonance based metabolomics and liverdiseases:Recent advances and future clinical applications显示文摘Metabolomics is defined as the quantitative measurement of the dynamic multiparametric metabolic response of living systems to pathophysiological stimuli or genetic modification.It is an'omics'technique that is situated downstream of genomics,transcriptomics and proteomics.Metabolomics is recognized as a promising technique in the field of systems biology for the evaluation of global metabolic changes.During the last decade,metabolomics approaches have become widely used in the study of liver diseases for the detection of early biomarkers and altered metabolic pathways.It is a powerful technique to improve our pathophysiological knowledge of various liver diseases.It can be a useful tool to help clinicians in the diagnostic process especially to distinguish malignant and non-malignant liver disease as well as to determine the etiology or severity of the liver disease.It can also assess therapeutic response or predict drug induced liver injury.Nevertheless,the usefulness of metabolomics is often not understood by clinicians,especially the concept of metabolomics profiling or fingerprinting.In the present work,after a concise description of the different techniques and processes used in metabolomics,we will review the main research on this subject by focusing specifically on in vitro proton nuclear magnetic resonance spectroscopy based metabolomics approaches in human studies.We will first consider the clinical point of view enlighten physicians on this new approach and emphasis its future use in clinical'routine'. | Roland Amathieu Mohamed Nawfal Triba Corentine Goossens Nadia Bouchemal Pierre Nahon Philippe Savarin Laurence Le Moyec | 2016 | World Journal of Gastroenterology2016,22,1: | 6 |
| 2 | Retrospective evaluation of FOLFIRI3 alone or in combination with bevacizumab or aflibercept in metastatic colorectal cancer显示文摘BACKGROUND The treatment of metastatic colorectal cancer(mCRC) relies of chemotherapy. The efficacy of the standard FOLFIRI-therapy could be improved by a modification of the regimen by splitting the dose of irinotecan on day 1 and day 3 in the FOLFIRI3 regimen.AIM To determine safety and efficacy of FOLFIRI3 regimen.METHODS This is a monocentric retrospective study evaluating the efficacy and safety of the FOLFIRI3 regimen given alone or in combination with bevacizumab or aflibercept in patients with previously treated mCRC.RESULTS One hundred and fifty-three consecutive patients were included(18 treated with FOLFIRI3, 99 with FOLFIRI3 plus bevacizumab and 36 with FOLFIRI3 plus aflibercept). The overall response rate(ORR) and disease control rate were 51%and 62%, respectively. Similar ORRs were observed in all 3 cohorts. Median progression-free survival(PFS) and overall survival(OS) were 3.9 mo(95%CI:3.2-4.9) and 9.4 mo(95%CI: 6.6-12), respectively. Median PFS and OS values were improved in the FOLFIRI3 plus aflibercept group. The most common grade 3-4 adverse events were diarrhoea(21.6%) and neutropenia(11.8%), and these toxicities were more frequent in the FOLFIRI3 plus aflibercept group. According to the multivariate Cox proportional model, previous surgery of metastasis andaflibercept were associated with outcomes.CONCLUSION The modification of the FOLFIRI regimen impacted treatment response of mCRC patients. | 5 | Adding body load modifies the vibratory sensation of the foot sole and affects the postural control显示文摘Background: Heavy backpacks are often used by soldiers and firefighters. Weight carrying could reduce the speed and efficiency in task completion by altering the foot sole sensitivity and postural control.Methods: In fifteen healthy subjects, we measured the changes in sensitivity to vibrations applied to the foot sole when standing upright or walking after load carrying(30% body weight). The participants were asked to judge different vibration amplitudes applied on the 2 nd or 5 th metatarsal head and the heel at two frequencies(25 and 150 Hz) to determine the vibration threshold and the global perceptual representation(Ψ)of the vibration amplitude(Φ)given by the Stevens power function(Ψ=k×Φ~n). Any increase in negative k value indicated a reduction in sensitivity to the lowest loads. Pedobarographic measurements, with computation of the center of pressure(COP) and its deviations, were performed during weight carrying.Results: The 25-Hz vibration threshold significantly increased after weight carrying when standing upright or walking.After standing with the added loads, the absolute negative k value increased for the 25 Hz frequency. After walking with the added loads, the k coefficient increased for the two vibration frequencies. | 9 | EGFR amplification induces sensitivity to antiEGFR therapy in pancreatic acinar cell carcinoma显示文摘Pancreatic acinar cell carcinoma(PACC) is a rare cancer. When the tumor is metastatic, few therapeutic options are available. Precision medicine using next-generation sequencing is defined by the administration of drugs based on the tumor genetic mutations. The usage of precision medicine for finding new therapeutic options for rare cancers is an emerging field. We have reported here the case of a patient bearing a multitreated metastatic PACC. This patient underwent somatic and constitutional exome analyses. The analyses revealed in the liver metastasis an amplification of the EGFR gene. Accordingly, the patient was treated with off-label usage of panitumumab. We observed rapid response with necrosis of the liver metastasis, while no efficacy was observed in the primary tumor. An exome analysis of the primary tumor revealed amplification of HER2 and MET with EGFR amplification. Such amplifications are known as a resistance mechanism to anti EGFR therapy. Our results suggest that exome analysis may be helpful to highlight targets in rare cancers, such as PACC. | 10 | New early Eocene vertebrate assemblage from western India reveals a mixed fauna of European and Gondwana affinities显示文摘The Ypresian Cambay Shale Formation at Vastan and Mangrol lignite mines in Gujarat, western India, has yielded a rich vertebrate fauna with numerous taxa of European affinities. Here we report a new,approximately contemporary vertebrate assemblage from two fossiliferous layers in the nearby mine of Tadkeshwar. These layers have yielded a similar mammal fauna with the co-occurrence of the perissodactyl-like cambaytheriid Cambaytherium thewissi, the adapoid primates Marcgodinotius indicus and cf. Asiadapis cambayensis, and the hyaenodontid Indohyaenodon raoi. The presence of these species in both Vastan and Tadkeshwar mines and at different levels suggests that the deposits between the two major lignite seams represent a single land mammal age. Apart from the aforementioned species there is a new, smaller species of Cambaytherium, and a new genus and species of esthonychid tillodont. This fauna also contains the first large early Eocene vertebrates from India, including an unidentified Coryphodon-like pantodont, a dyrosaurid crocodyliform and a new giant madtsoiid snake. Among the Tadkeshwar vertebrates several taxa are of Gondwana affinities, such as Pelomedusoides turtles, dyrosaurids, and large madtsoiids, attesting that the early Eocene was a crucial period in India during which Laurasian taxa of European affinities co-existed with relict taxa from Gondwana before the India-Asia collision. Our results suggest that terrestrial faunas could have dispersed to or from Europe during episodes of contact between the Indian subcontinent and different island blocks along the northern margin of the Neotethys, such as the Kohistane Ladakh island-arc system. Gondwana taxa might represent remnants of ghost lineages shared with Madagascar, which reached the Indian subcontinent during the late Cretaceous; alternatively they might have come from North Africa and passed along the southern margin of the Neotethys to reach the Indian subcontinent. | 16 | Influence of'protective'symbionts throughout the different steps of an aphid-parasitoid interaction显示文摘Microbial associates are widespread in insects,some conferring a protection to their hosts against natural enemies like parasitoids.These protective symbionts may affect the infection success of the parasitoid by modifying behavioral defenses of their hosts,the development success of the parasitoid by conferring a resistance against it or by altering life-history traits of the emerging parasitoids.Here,we assessed the effects of different protective bacterial symbionts on the entire sequence of the host-parasitoid interaction(i.e.,from parasitoid attack to offspring emergence)between the pea aphid,Acyrthosiphon pisum,and its main parasitoid,Aphidius ervi and their impacts on the life-history traits of the emerging parasitoids.To test whether symbiont-mediated phenotypes were general or specific to particular aphid-symbiont associations,we considered several aphid lineages,each harboring a different strain of either Hamiltonella defensa or Regiella insecticola,two protective symbionts commonly found in aphids.We found that symbiont species and strains had a weak effect on the ability of aphids to defend themselves against the parasitic wasps during the attack and a strong effect on aphid resistance against parasitoid development.While parasitism resistance was mainly determined by symbionts,their effects on host defensive behaviors varied from one aphid-symbiont association to another.Also,the symbiotic status of the aphid individuals had no impact on the attack rate of the parasitic wasps,the parasitoid emergence rate from parasitized aphids nor the life-history traits of the emerging parasitoids.Overall,no correlations between symbiont effects on the different stages of the host-parasitoid interaction was observed,suggesting no trade-offs or positive associations between symbiont-mediated phenotypes.Our study highlights the need to consider various sequences of the host-parasitoid interaction to better assess the outcomes of protective symbioses and understand the ecological and evolutionary dynamics of insect-symbiont associations. | Corentin Sochard Laura Bellec Jean-Christophe Simon Yannick Outreman | 2021 | Current Zoology2021,67,4: | 0 |

| 18 | Reassessment of historical sections from the Paleogene marine margin of the Congo Basin reveals an almost complete absence of Danian deposits显示文摘The early Paleogene is critical for understanding global biodiversity patterns in modern ecosystems. During this interval, Southern Hemisphere continents were largely characterized by isolation and faunal endemism following the breakup of Gondwana. Africa has been proposed as an important source area for the origin of several marine vertebrate groups but its Paleogene record is poorly sampled, especially from sub-Saharan Africa. To document the early Paleogene marine ecosystems of Central Africa, we revised the stratigraphic context of sedimentary deposits from three fossil-rich vertebrate localities: the Landana section in the Cabinda exclave(Angola), and the Manzadi and Bololo localities in western Democratic Republic of Congo.We provide more refined age constraints for these three localities based on invertebrate and vertebrate faunas, foraminiferal and dinoflagellate cyst assemblages, and carbon isotope records. We find an almost complete absence of Danian-aged rocks in the Landana section, contrary to prevailing interpretations over the last half a century(only the layer 1, at the base of the section, seems to be Danian). Refining the age of these Paleocene layers is crucial for analyzing fish evolution in a global framework, with implications for the early appearance of Scombridae(tunas and mackerels) and Tetraodontiformes(puffer fishes). The combination of vertebrate fossil records from Manzadi and Landana sections suggests important environmental changes around the K/Pg transition characterized by an important modification of the ichthyofauna. A small faunal shift may have occurred during the Selandian. More dramatic is the distinct decrease in overall richness that lasts from the Selandian to the Ypresian. The Lutetian of West Central Africa is characterized by the first appearance of numerous cartilaginous and bony fishes. Our analysis of the ichthyofauna moreover indicates two periods of faunal exchanges: one during the Paleocene, where Central Africa appears to have been a source for the European marine fauna, and another during the Eocene when Europe was the source of the Central Africa fauna.
