1Respiratory Control by Phox2b-expressing Neurons in a Locus Coeruleus–pre B?tzinger Complex Circuit显示文摘The locus coeruleus(LC) has been implicated in the control of breathing.Congenital central hypoventilation syndrome results from mutation of the paired-like homeobox 2 b(Phox2 b) gene that is expressed in LC neurons.The present study was designed to address whether stimulation of Phox2 b-expressing LC(Phox2 b~(LC)) neurons affects breathing and to reveal the putative circuit mechanism.A Cre-dependent viral vector encoding a Gqcoupled human M3 muscarinic receptor(hM3 Dq) was delivered into the LC of Phox2 b-Cre mice.The hM3 Dqtransduced neurons were pharmacologically activated while respiratory function was measured by plethysmography.We demonstrated that selective stimulation of Phox2 b~(LC) neurons significantly increased basal ventilation in conscious mice.Genetic ablation of these neurons markedly impaired hypercapnic ventilatory responses.Moreover,stimulation of Phox2 b~(LC) neurons enhanced the activity of preBotzinger complex neurons.Finally,axons of Phox2 b~(LC) neurons projected to the preBotzinger complex.Collectively,Phox2 b~(LC) neurons contribute to the control of breathing most likely via an LC-preBotzinger complex circuit.Na Liu Congrui Fu Hongxiao Yu Yakun Wang Luo Shi Yinchao Hao Fang Yuan Xiangjian Zhang Sheng Wang 2021Neuroscience Bulletin2021,37,1:3
2Response of PM_(2.5)-bound elemental species to emission variations and associated health risk assessment during the COVID-19 pandemic in a coastal megacity显示文摘The coronavirus(COVID-19)pandemic is disrupting the world from many aspects.In this study,the impact of emission variations on PM_(2.5)-bound elemental species and health risks associated to inhalation exposure has been analyzed based on real-time measurements at a remote coastal site in Shanghai during the pandemic.Most trace elemental species decreased significantly and displayed almost no diel peaks during the lockdown.After the lockdown,they rebounded rapidly,of which V and Ni even exceeded the levels before the lockdown,suggesting the recovery of both inland and shipping activities.Five sources were identified based on receptor modeling.Coal combustion accounted for more than 70%of the measured elemental concentrations before and during the lockdown.Shipping emissions,fugitive/mineral dust,and waste incineration all showed elevated contributions after the lockdown.The total non-carcinogenic risk(HQ)for the target elements exceeded the risk threshold for both children and adults with chloride as the predominant species contributing to HQ.Whereas,the total carcinogenic risk(TR)for adults was above the acceptable level and much higher than that for children.Waste incineration was the largest contributor to HQ,while manufacture processing and coal combustion were the main sources of TR.Lockdown control measures were beneficial for lowering the carcinogenic risk while unexpectedly increased the non-carcinogenic risk.From the perspective of health effects,priorities of control measures should be given to waste incineration,manufacture processing,and coal combustion.A balanced way should be reached between both lowering the levels of air pollutants and their health risks.Guochen Wang Kan Huang Qingyan Fu Jia Chen Juntao Huo Qianbiao Zhao Yusen Duan Yanfen Lin Fan Yang Wenjie Zhang Hao Li Jian Xu Xiaofei Qin Na Zhao Congrui Deng 2022Journal of Environmental Sciences2022,34,12:2
3A Neural Circuit Mechanism Controlling Breathing by Leptin in the Nucleus Tractus Solitarii显示文摘Leptin,an adipocyte-derived peptide hormone,has been shown to facilitate breathing.However,the central sites and circuit mechanisms underlying the respiratory effects of leptin remain incompletely understood.The present study aimed to address whether neurons expressing leptin receptor b(LepRb)in the nucleus tractus solitarii(NTS)contribute to respiratory control.Both chemogenetic and optogenetic stimulation of LepRb-ex-pressing NTS(NTS^(LepRb))neurons notably activated breathing.Moreover,stimulation of NTS^(LepRb) neurons projecting to the lateral parabrachial nucleus(LPBN)not only remarkably increased basal ventilation to a level similar to that of the stimulation of all NTS^(LepRb) neurons,but also activated LPBN neurons projecting to the preBotzinger complex(preBotC).By contrast,ablation of NTS^pRb neurons projecting to the LPBN notably eliminated the enhanced respiratory effect induced by NTSLepRb neuron stimulation.In brainstem slices,bath application of leptin rapidly depolarized the membrane potential,increased the spontaneous firing rate,and accelerated the Ca2+transients in most NTSLepRb neurons.Therefore,leptin potentiates breathing in the NTS most likely via an NTS-LPBN-preBdtC circuit.Hongxiao Yu Luo Shi Jinting Chen Shirui Jun Yinchao Hao Shuang Wang Congrui Fu Xiang Zhang Haiyan Lu Sheng Wang Fang Yuan 2022Neuroscience Bulletin2022,38,2:1
