1Osteoblasts are inherently programmed to repel sensory innervation显示文摘Tissue innervation is a complex process controlled by the expression profile of signaling molecules secreted by tissue-resident cells that dictate the growth and guidance of axons.Sensory innervation is part of the neuronal network of the bone tissue with a defined spatiotemporal occurrence during bone development.Yet,the current understanding of the mechanisms regulating the map of sensory innervation in the bone tissue is still limited.Here,we demonstrated that differentiation of human mesenchymal stem cells to osteoblasts leads to a marked impairment of their ability to promote axonal growth,evidenced under sensory neurons and osteoblastic-lineage cells crosstalk.The mechanisms by which osteoblast lineage cells provide this nonpermissive environment for axons include paracrine-induced repulsion and loss of neurotrophic factors expression.We identified a drastic reduction of NGF and BDNF production and stimulation of Sema3A,Wnt4;and Shh expression culminating at late stage of OB differentiation.We noted a correlation between Shh expression profile,OB differentiation stages,and OB-mediated axonal repulsion.Blockade of Shh activity and signaling reversed the repulsive action of osteoblasts on sensory axons.Finally,to strengthen our model,we localized the expression of Shh by osteoblasts in bone tissue.Overall,our findings provide evidence that the signaling profile associated with osteoblast phenotype differentiating program can regulate the patterning of sensory innervation,and highlight osteoblast-derived Shh as an essential player in this cue-induced regulation.Luis Leitao Estrela Neto Francisco Conceicao Ana Monteiro Marina Couto Cecilia JAlves Daniela MSousa Meriem Lamghari 2020Bone Research2020,8,3:4
2Coating independent cytotoxicity of citrate-and PEG-coated silver nanoparticles on a human hepatoma cell line显示文摘The antibacterial potential of silver nanoparticles(AgNPs) resulted in their increasing incorporation into consumer,industrial and biomedical products.Therefore,human and environmental exposure to AgNPs(either as an engineered product or a contaminant)supports the emergent research on the features conferring them different toxicity profiles.In this study,30 ran AgNPs coated with citrate or poly(ethylene glycol)(PEG) were used to assess the influence of coating on the effects produced on a human hepatoma cell line(HepG2),namely in terms of viability,apoptosis,apoptotic related genes,cell cycle and cyclins gene expression.Both types of coated AgNPs decreased cell proliferation and viability with a similar toxicity profile.At the concentrations used(11 and 5 μg/mL corresponding to IC50 and-IC10 levels,respectively) the amount of cells undergoing apoptosis was not significant and the apoptotic related genes BCL2(anti-apoptotic gene)and BAX(pro-apoptotic gene) were both downregulated.Moreover,both AgNPs affected HepG2 cell cycle progression at the higher concentration(11 μg/mL) by increasing the percentage of cells in S(synthesis phase) and G2(Gap 2 phase) phases.Considering the cell-cycle related genes,the expression of cyclin B1 and cyclin E1 genes were decreased.Thus,this work has shown that citrate- and PEG-coated AgNPs impact on HepG2 apoptotic gene expression,cell cycle dynamics and cyclin regulation in a similar way.More research is needed to determine the properties that confer AgNPs at lower toxicity,since their use has proved helpful in several industrial and biomedical contexts.Veronica Bastos Jose M.P.Ferreira-de-Oliveira Joana Carrola Ana L.Daniel-da-Silva Iola F.Duarte Conceicao Santos Helena Oliveira 2017Journal of Environmental Sciences2017,29,1:4
3Phosphomonoester Phosphoethanolamine Induces Apoptosis in Human Chronic Myeloid Leukemia Cells显示文摘Background:Leukemia is a type of cancer that starts in the blood or blood-forming tissues.It results from the clonal proliferation of hematopoietic cells in the bone marrow and/or lymphoid tissues,which subsequently reach the peripheral circulation and can infiltrate other systems.There are many different kinds of leukemia,and treatments are different for each one.Chronic leukemia is with a slower growing than acute leukemia but could be just as life-threatening.Phospholipids are antitumor analogs,such as synthetic phosphoethanolamine,which is a phosphorylated compound capable of controlling cellular proliferation and inducing apoptosis in several types of tumor cells.Methods:K562 and K562-Lucena(MDR+)human chronic myeloid leukemia cells were treated with synthetic phosphoethanolamine(Pho-s).The viability was evaluated by sulforhodamine B(SRB)assay and cell cycle phases,apoptosis,markers expression,and mitochondrial potential were assessed by flow cytometry.Results:Tumor cells formed clusters in suspension and decreased significantly viability.The concentrations for IC50%were obtained.Pho-s treated were 43.1 mM(K562)and 145.9 mM(K562-Lucena MDR+)in a period of 24 hours.Pho-s induced changes in the distribution of cell population phases of cell cycle which showed an increase in fragmented DNA and increased markers expression envolved apoptosis pathways a decrease in the G1/G0 phase.Discussion:Treatment of K562 and K562-Lucena(MDR+)chronic myeloid leukemia cells with Pho-s showed dose and time dependent cytotoxic effects.This cytotoxicity induced a decrease in proliferative capacity,mitochondrial electrical potential,and consequently release of cytochrome C;inhibition of Bcl-2 family protein expression,increase in pro-apoptotic family members Bad and Bax,dependent on p53 expression.Conclusion:This study presented a significant therapeutic potential of Phos-s in this type of leukemia through the apoptotic effects on tumor cells independently of the molecular resistance profile(MDR+).Thais de Oliveira Conceicao Manuela Garcia Laveli da Silva Durvanei Augusto Maria 2019Journal of Pharmacy and Pharmacology2019,7,7:3
