| 1 | Physiopathology of splanchnic vasodilation in portal hypertension显示文摘In liver cirrhosis, the circulatory hemodynamic alterations of portal hypertension signifi cantly contribute to many of the clinical manifestations of the disease. In the physiopathology of this vascular alteration, mesen- teric splanchnic vasodilation plays an essential role by initiating the hemodynamic process. Numerous studies performed in cirrhotic patients and animal models have shown that this splanchnic vasodilation is the result of an important increase in local and systemic vasodilators and the presence of a splanchnic vascular hyporesponsiveness to vasoconstrictors. Among the molecules and factors known to be potentially involved in this arterial vasodilation, nitric oxide seems to have a crucial role in the physiopathology of this vascular alteration. However, none of the wide variety of mediators can be described as solely responsible, since this phenomenon is multifactorial in origin. Moreover, angiogenesis and vascular remodeling processes alsoseem to play a role. Finally, the sympathetic nervous system is thought to be involved in the pathogenesis of the hyperdynamic circulation associated with portal hypertension, although the nature and extent of its role is not completely understood. | 2 | Single-lung transplantation in emphysema:Retrospective study analyzing survival and waiting list mortality显示文摘AIM: To performed remains a subject of debate and is the principal aim of the study. METHODS: This retrospective analysis included 73 patients with emphysema(2000-2012). The outcomes of patients undergoing single-lung transplantation(SL)(n = 40) or double-lung transplant(DL)(n = 33) were compared in a Cox multivariate analysis to study the impact of the technique, postoperative complications and acute and chronic rejection on survival rates. Patients were selected for inclusion in the waiting list according to the International Society of Heart Lung Transplantation criteria. Pre and postoperative rehabilitation and prophylaxis, surgical technique and immunosuppressive treatment were similar in every patients. Lung transplantation waiting list information on a national level and retrospective data on emphysema patient survival transplanted in Spain during the study period, was obtained from the lung transplantation registry managed by the National Transplant Organization(ONT). RESULTS: Both groups were comparable in terms of gender and clinical characteristics. We found significant differences in the mean age between the groups, the DL patients being younger as expected from the inclusion criteria. Perioperative complications occurred in 27.6% SL vs 54% DL(P = 0.032). Excluding perioperative mortality, median survival was 65.3 mo for SL and 59.4 mo for DL(P = 0.96). Bronchiolitis obliterans and overall 5-year survival were similar in both groups. Bacterialrespiratory infection, cytomegalovirus and fungal infection rates were higher but not significant in SL. No differences were found between type of transplant and survival(P = 0.48). To support our results, national data on all patients with emphysema in waiting list were obtained(n = 1001). Mortality on the waiting list was 2.4% for SL vs 6.2% for DL. | 3 | Transcriptomic and metabolomic analyses of cucumber fruit peels reveal a developmental increase in terpenoid glycosides associated with age-related resistance to Phytophthora capsici显示文摘The oomycete,Phytophthora capsici,infects cucumber(Cucumis sativus L.)fruit.An age-related resistance(ARR)to this pathogen was previously observed in fruit of cultivar‘Vlaspik’and shown to be associated with the peel.Young fruits are highly susceptible,but develop resistance at~10–12 days post pollination(dpp).Peels from resistant(16 dpp)versus susceptible(8 dpp)age fruit are enriched with genes associated with defense,and methanolic extracts from resistant age peels inhibit pathogen growth.Here we compared developing fruits from‘Vlaspik’with those of‘Gy14’,a line that does not exhibit ARR.Transcriptomic analysis of peels of the two lines at 8 and 16 dpp identified 80 genes that were developmentally upregulated in resistant‘Vlaspik’16 dpp versus 8 dpp,but not in susceptible‘Gy14’at 16 dpp.A large number of these genes are annotated to be associated with defense and/or specialized metabolism,including four putative resistance(R)genes,and numerous genes involved in flavonoid and terpenoid synthesis and decoration.Untargeted metabolomic analysis was performed on extracts from 8 and 16 dpp‘Vlaspik’and‘Gy14’fruit peels using Ultra-Performance Liquid Chromatography and Quadrupole Time-of-Flight Mass Spectrometry.Multivariate analysis of the metabolomes identified 113 ions uniquely abundant in resistant‘Vlaspik’16 dpp peel extracts.The most abundant compounds in this group had relative mass defects consistent with terpenoid glycosides.Two of the three most abundant ions were annotated as glycosylated nor-terpenoid esters.Together,these analyses reveal potential mechanisms by which ARR to P.capsici may be conferred. | Ben N Mansfeld Marivi Colle Yunyan Kang A Daniel Jones Rebecca Grumet | 2017 | Horticulture Research2017，4，1： | 3 |
