3小麦农杆菌介导转基因植株的稳定获得和检测显示文摘以预培养 4 d的小麦幼胚愈伤组织为受体 ,L B培养基上激活培养 3 d或 AB培养基上激活培养 4 d,并在WCC培养基中重悬的 C5 8c1农杆菌菌系为供体 ,将含有目的基因、标记基因的 p PTN2 4 9、p PTN 2 5 4、p PTN2 70、p SIS-GFP等重组 DNA质粒转入了小麦 ,经在含 10~ 5 0 mg/ L geneticin( G4 18)、5 0 mg/ L cefotaxime、5 0 m g/ L vancomycin、5 0 m g/ L ticillicin选择培养基上多次选择 ,移栽前利用 npt EL ISA检测 ,移栽后利用叶片离体退绿、PCR和 South-ern blot 4种方法综合检测 ,共获得了 2 9株转基因植株 ,转化频率平均为 0 .82 %。在所获得的转基因植株中 ,单拷贝整合植株占 65 .5 2 %。研究结果还表明 ,不同基因型的转化效率显著不同 ,除了 Bobwhite外 ,笔者也从扬麦 10号获得了转基因植株 ;EL ISA测定值的高低与外源 DNA整合的拷贝数有一定关系 ;初步建立了农杆菌介导稳定转化小麦的技术体系。叶兴国 Shirley Sato 徐惠君 杜丽璞 Tom Clemente 2001中国农业科学2001,34,5:66
4Intestinal microbiota in health and disease: Role of bifidobacteria in gut homeostasis显示文摘The pool of microbes inhabiting our body is known as 'microbiota' and their collective genomes as 'microbiome'. The colon is the most densely populated organ in the human body, although other parts, such as the skin, vaginal mucosa, or respiratory tract, also harbour specific microbiota. This microbial community regulates some important metabolic and physiological functions of the host, and drives the maturation of the immune system in early life, contributing to its homeostasis during life. Alterations of the intestinal microbiota can occur by changes in composition(dysbiosis), function, or microbiota-host interactions and they can be directly correlated with several diseases. The only disease in which a clear causal role of a dysbiotic microbiota has been demonstrated is the case of Clostridium difficile infections. Nonetheless, alterations in composition and function of the microbiota have been associated with several gastrointestinal diseases(inflammatory bowel disease, colorectal cancer, or irritable bowel syndrome), as well as extra-intestinal pathologies, such as those affecting the liver, or the respiratory tract(e.g., allergy, bronchial asthma, and cystic fibrosis), among others. Species of Bifidobacterium genus are the normal inhabitants of a healthy human gut and alterations in number and composition of their populations is one of the most frequent features present in these diseases. The use of probiotics, including bifidobacteria strains, in preventive medicine to maintain a healthy intestinal function is well documented. Probiotics are also proposed as therapeutic agents for gastrointestinal disorders and other pathologies. The World Gastroenterology Organization recently published potential clinical applications for several probiotic formulations, in which species of lactobacilli are predominant. This review is focused on probiotic preparations containing Bifidobacterium strains, alone or in combination with other bacteria, which have been tested in human clinical studies. In spite of extensive literature on and research into this topic, the degree of scientific evidence of the effectiveness of probiotics is still insufficient in most cases. 5Pediatric non-alcoholic fatty liver disease: Recent solutions, unresolved issues, and future research directions显示文摘Non-alcoholic fatty liver disease(NAFLD) in children is becoming a major health concern. A 'multiple-hit' pathogenetic model has been suggested to explain the progressive liver damage that occurs among children with NAFLD. In addition to the accumulation of fat in the liver, insulin resistance(IR) and oxidative stress due to genetic/epigenetic background, unfavorable lifestyles, gut microbiota and gut-liver axis dysfunction, and perturbations of trace element homeostasis have been shown to be critical for disease progression and the development of more severe inflammatory and fibrotic stages [non-alcoholic steatohepatitis(NASH)]. Simple clinical and laboratory parameters, such as age, history, anthropometrical data(BMI and waist circumference percentiles), blood pressure, surrogate clinical markers of IR(acanthosis nigricans), abdominal ultrasounds, and serum transaminases, lipids and glucose/insulin profiles, allow a clinician to identify children with obesity and obesity-related conditions, including NAFLD and cardiovascular and metabolic risks. A liver biopsy(the 'imperfect' gold standard) is required for a definitive NAFLD/NASH diagnosis, particularly to exclude other treatable conditions or when advanced liver disease is expected on clinical and laboratory grounds and preferably prior to any controlled trial of pharmacological/surgical treatments. However, a biopsy clearly cannot represent a screening procedure. Advancements in diagnostic serum and imaging tools, especially for the non-invasive differentiation between NAFLD and NASH, have shown promising results, e.g., magnetic resonance elastography. Weight loss and physical activity should be the first option of intervention.Effective pharmacological treatments are still under development; however, drugs targeting IR, oxidative stress, proinflammatory pathways, dyslipidemia, gut microbiota and gut liver axis dysfunction are an option for patients who are unable to comply with the recommended lifestyle changes. 13A time series for monitoring vegetation activity and phenology at 10-daily time steps covering large parts of South America显示文摘It is widely accepted that natural resources should only be sustainably exploited and utilized to effectively preserve our planet for future generations.To better manage the natural resources,and to better understand the closely linked Earth systems,the concept of Digital Earth has been strongly promoted since US Vice President Al Gore’s speech in 1998.One core element of Digital Earth is the use and integration of remote sensing data.Only satellite imagery can cover the entire globe repeatedly at a sufficient high-spatial resolution to map changes in land cover and land use,but also to detect more subtle changes related for instance to climate change.To uncover global change effects on vegetation activity and phenology,it is important to establish high quality time series characterizing the past situation against which the current state can be compared.With the present study we describe a time series of vegetation activity at 10-daily time steps between 1998 and 2008 covering large parts of South America at 1 km spatial resolution.Particular emphasis was put on noise removal.Only carefully filtered time series of vegetation indices can be used as a benchmark and for studying vegetation dynamics at a continental scale.Without temporal smoothing,subtle spatio-temporal patterns in vegetation composition,density and phenology would be hidden by atmospheric noise and undetected clouds.Such noise is immanent in data that have undergone solely a maximum value compositing.Within the present study,the Whittaker smoother(WS)was applied to a SPOT VGT time series.The WS balances the fidelity to the observations with the roughness of the smoothed curve.The algorithm is extremely fast,gives continuous control over smoothness with only one parameter,and interpolates automatically.The filtering efficiently removed the negatively biased noise present in the original data,while preserving the overall shape of the curves showing vegetation growth and development.Geostatistical variogram analysis revealed a significantly increased signal-to-noise ratio compared to the raw data.Analysis of the data also revealed spatially consistent key phenological markers.Extracted seasonality parameters followed a clear meridional trend.Compared to the unfiltered data,the filtered time series increased the separability of various land cover classes.It is thus expected that the data set holds great potential for environmental and vegetation related studies within the frame of Digital Earth.Clement Atzbergera Paul HCEilers 2011International Journal of Digital Earth2011,4,5:14

15国内外36个苜蓿品种的发芽特性综合评价显示文摘苜蓿品种的优劣影响苜蓿草地建植与生产性能,其中发芽特性是首要指标。为筛选出优良的品种用于试验研究和生产,收集了我国育成苜蓿品种14个和国外22个品种的种子(其中包衣种子8个),测定了发芽率、硬实率、苗长等12个指标,采用隶属函数平均值法综合评价了品种优劣。结果表明,供试品种的千粒重为1.56~4.87g,发芽势为49.33%~97.33%(平均为83.52%),发芽率为49.33%~97.33%(平均为86.85%),发芽指数为5.83~17.04(平均为10.90),活力指数为0.18~0.60(平均为0.390),25个品种有硬实种子,其中,新疆大叶硬实率为49.33%显著高于其他品种,将硬实种子划破种皮继续培养,发芽率均有提升。综合指标评价最高的5个品种为甘农6号、中苜3号、甘农9号、耐盐之星、前景,综合指标评价最低的5个品种为公农5号、WL 168HQ、WL 354HQ、太阳神、新疆大叶。我国品种综合指标较高,平均为0.51,国外品种平均为0.44,国内品种表现优于国外品种。马婷燕 罗飞 Clement Nzabanita 李彦忠 2019种子2019,38,1:13

18边疆深度贫困地区社会资本、生计策略与精准扶贫显示文摘社会资本和人力资本建设作为边疆深度贫困地区的社会无形资产,对边疆地区精准扶贫实现'两个回流'及边民生计可持续具有重要作用。本文利用2016年7月新疆克州的田野调查数据,采用logistic模型估计边疆深度贫困地区社会资本对于边民生计策略以及脱贫的影响效应,并运用定性与定量相结合的研究方法对研究结果进行解读。研究表明,地区社会资本有助于边民减贫,这种正向效应主要通过两个路径实现:一是边民借助社会网络提高对正规金融的可得性进而实现养殖扩大再生产;二是边民通过集体公共活动付酬增加转移支付收入。但社会资本通过生计策略多样化机制提升边民生计能力的减贫路径尚未形成。在边疆深度贫困地区脱贫攻坚的关键期,既要充分发挥社会资本对边民收入增收的促进作用,同时也需配套其他措施破解社会资本引导边民建立多样化生计的主要障碍,强化社会资本的能力扶持路径,实现精准脱贫的可持续。刘红 Clement Tisdell 李文静 2019商业研究2019,,5:12
19单髁与全膝关节置换治疗内侧间室膝骨关节炎:5年假体在位率比较显示文摘背景:单髁置换越来越多的被应用于治疗单纯内侧胫股间室型膝关节骨关节炎。目的:比较膝关节单髁置换与全膝关节置换对膝关节单纯内侧间室骨关节炎的修复效果。方法:纳入单纯内侧间室型膝关节骨关节炎患者109例,根据修复方式分为两组,单髁置换组41例,全膝关节置换组68例。比较两组患者的平均住院时间、置换后并发症发生率、身体健康总评分、精神健康总评分、牛津膝关节评分以及置换后5年假体在位率。结果与结论:单髁置换组患者平均住院时间(4.21±2.44)d,较全膝关节置换组(6.11±2.23)d短(P=0.001)。两组患者置换后并发症发生率、身体健康总评分、精神健康总评分、牛津膝关节评分等方面差异均无显著性意义(P>0.05)。置换后5年,单髁置换组假体在位率(90%)较全膝关节置换组(100%)低(P<0.05)。提示对于单纯内侧胫股间室型膝关节骨关节炎患者,单髁置换与全膝关节置换具有相似的修复效果,但单髁置换后5年假体在位率稍低。梁宁 Samuel Yih Shyan Wong Nick Clement Deborah MacDonald Daniel E Porter 2015中国组织工程研究2015,19,53:12
20Hepatocellular adenoma: An unsolved diagnostic enigma显示文摘Hepatocellular adenoma (HCA) is a rare benign liver tumour associated with the use of oral contraceptives or other steroid medications which occurs predominantly in young and middle-aged women. Unlike other benign liver tumours, an HCA may be complicated by bleeding and malignant transformation. HCAs have been divided into four subtypes based on molecular and pathological features: hepatocyte nuclear factor 1α-mutated HCA, inflammatory HCA,β-catenin-mutated HCA, and unclassified HCA.β-cateninmutated HCA has the highest risk of haemorrhage or malignant transformation. In the latest upgrade of the guidelines regarding the management of benign liver tumours published in 2016 by the European Association for the Study of the Liver, magnetic resonance imaging (MRI) was recognized to be superior to all other imaging modalities in detecting HCAs and in being able to subtype HCAs up to 80%, with positive identification of 1α-mutated HCA or inflammatory HCA achievable with > 90% specificity.
