2液相色谱质谱联用测定多种兽药残留方法中标准品稳定性的挑战显示文摘采用等时测量法对可用于食品和饲料链中兽药检测的标准品混合溶液和中间混合液进行了短期和长期稳定性研究。其中短期稳定性实验考察了混合标准曲线在–20(作为基线)、4和23℃(避光和不避光保存)条件下存储1、2、4和7 d的稳定性,而长期稳定性实验考察了中间混合液在–20、4、23℃(避光和不避光保存)和–80℃储存条件下2、4、8和12周的稳定性。结果表明,使用不含酸溶剂配制的混合标准曲线在4℃下建议储存1~2 d,中性储存条件即使在室温也可短期储存1~2 d。对于含有ß-内酰胺和头孢菌素类标准品的中间混合液在4℃和室温下储存超过1个月会损失近90%。对于含有青霉素V和G的混和标准品中间储备液,在–20℃不含酸的条件下可储存2周。其他类别的兽药标准品对长期储存条件要求较少。整体而言,兽药标准品中间混合液长期储存的最佳储存条件为–20℃。Lidija Kenjeric Michael Sulyok Alexandra Malachova David Steiner Rudolf Krska Brian Quinn Brett Greer Christopher T.Elliott 2021粮油食品科技2021,29,6:0
3The Challenge of Standard Stability in LC-MS Based Multi-Analyte Approaches for Veterinary Drugs显示文摘The short-and long-term stability of multi-component mixtures and intermediate mixtures of analytical standards of veterinary drugs,which can potentially occur in food and feed chains,was examined by an isochronous measurement approach.Short-term stability testing of calibrants included storage for 1,2,4,and 7 days at-20℃(as a baseline)4℃,and 23℃(with and without exposure to sunlight),respectively.Long-term stability testing conditions of intermediate mixes were-20℃,4℃,23℃(with and without exposure to sunlight),and control temperature at-80℃while the testing period was 2,4,8,and 12 weeks,respectively.Results indicated that calibration standards should ideally be stored at 4℃for only 1~2 days,without the presence of acid.Neutral storage conditions were acceptable even at room temperature.Storage of intermediate mixtures containingß-lactams and cephalosporins for longer than 1 month under 4℃ and room temperature resulted in a loss of almost 90%.When it comes to the intermediate mixtures with penicillin V and G,acceptable storage conditions were 2 weeks at-20℃,without the presence of acid.Other classes of veterinary drugs were less critical as considers long-term stability.Overall,storage conditions at-20℃were considered optimal for long-term storage of intermediate mixes of veterinary drug standards.Lidija Kenjeric Michael Sulyok Alexandra Malachova David Steiner Rudolf Krska Brian Quinn Brett Greer Christopher T.Elliott 2021粮油食品科技2021,29,6:0
4Rapid metabolic fingerprinting with the aid of chemometric models to identify authenticity of natural medicines: Turmeric, Ocimum, and Withania somnifera study显示文摘Herbal medicines are popular natural medicines that have been used for decades.The use of alternative medicines continues to expand rapidly across the world.The World Health Organization suggests that quality assessment of natural medicines is essential for any therapeutic or health care applications,as their therapeutic potential varies between different geographic origins,plant species,and varieties.Classification of herbal medicines based on a limited number of secondary metabolites is not an ideal approach.Their quality should be considered based on a complete metabolic profile,as their pharmacological activity is not due to a few specific secondary metabolites but rather a larger group of bioactive compounds.A holistic and integrative approach using rapid and nondestructive analytical strategies for the screening of herbal medicines is required for robust characterization.In this study,a rapid and effective quality assessment system for geographical traceability,species,and variety-specific authenticity of the widely used natural medicines turmeric,Ocimum,and Withania somnifera was investigated using Fourier transform near-infrared(FT-NIR)spectroscopy-based metabolic fingerprinting.Four different geographical origins of turmeric,five different Ocimum species,and three different varieties of roots and leaves of Withania somnifera were studied with the aid of machine learning approaches.Extremely good discrimination(R^(2)>0.98,Q^(2)>0.97,and accuracy=1.0)with sensitivity and specificity of 100%was achieved using this metabolic fingerprinting strategy.Our study demonstrated that FT-NIR-based rapid metabolic fingerprinting can be used as a robust analytical method to authenticate several important medicinal herbs.Samreen Khan Abhishek Kumar Rai Anjali Singh Saudan Singh Basant Kumar Dubey Raj Kishori Lal Arvind Singh Negi Nicholas Birse Prabodh Kumar Trivedi Christopher T.Elliott Ratnasekhar Ch 2023Journal of Pharmaceutical Analysis2023,13,9:0
