| 1 | MicroRNA-143 suppresses gastric cancer cell growth and induces apoptosis by targeting COX-2显示文摘AIM:To investigate the function of microRNA-143(miR-143)in gastric cancer and explore the target genes of miR-143.METHODS:A quantitative real-time reverse transcription-polymerase chain reaction(RT-PCR)analysis was performed to evaluate miR-143 expression in gastric cancer cell lines.After transfecting gastric cancer cells with miR-143-5p and miR-143-3p precursors,Alamar blue and apoptosis assays were used to measure the respective proliferation and apoptosis rates.Cyclooxygenase-2(COX-2)expression was determined by realtime RT-PCR and Western blot assays after miR-143transfection.Reporter plasmids were constructed,and a luciferase reporter assay was used to identify the miR-143 binding site on COX-2.RESULTS:Both miR-143-5p and miR-143-3p were significantly downregulated in multiple gastric cancer cell lines.Forced miR-143-5p and miR-143-3p expression in gastric cancer cells produced a profound cytotoxic effect.MiR-145-5p transfection into gastric cancer cells resulted in a greater growth inhibitory effect(61.23%±3.16%vs 46.58%±4.28%,P<0.05 in the MKN-1cell line)and a higher apoptosis rate(28.74%±1.93%vs 22.13%±3.31%,P<0.05 in the MKN-1 cell line)than miR-143-3p transfection.Further analysis indicated that COX-2 expression was potently suppressed by miR-143-5p but not by miR-143-3p.The activity of a luciferase reporter construct that contained the 3’-untranslated region(UTR)of COX-2 was downregulated by miR-143-5p(43.6%±4.86%,P<0.01)but not by miR-143-3p.A mutation in the miR-145-5p binding site completely ablated the regulatory effect on luciferase activity,which suggests that there is a direct miR-145-5p binding site in the 3’-UTR of COX-2.CONCLUSION:Both miR-143-5p and miR-143-3p function as anti-oncomirs in gastric cancer.However,miR-143-5p alone directly targets COX-2,and it exhibits a stronger tumor suppressive effect than miR-143-3p. | Xiao-Li Wu Bin Cheng Pei-Yuan Li Huan-Jun Huang Qiu Zhao Zi-Li Dan Dean Tian Peng Zhang | 2013 | World Journal of Gastroenterology2013,19,43: | 15 |

| 3 | Regioselective oxidation of tetracycline by permanganate through alternating susceptible moiety and increasing electron donating ability显示文摘Permanganate has attracted much attention in wide range of chemistry and particularly in degradation of environmental pollutants.However,few studies have discussed the feature of regioselective reactivity of permanganate with specific moiety of the target compound.Herein,we studied the reaction between permanganate and tetracycline that is an emerging micropollutant with different species containing several electron-rich groups.The second-order rate constants increased from 6.0 to 9.0 and could be quantitatively modeled by considering the speciation of both reactants,yielding kTC0=11.7(mol/L)^-1 sec^-1,kTC-=35.7(mol/L)^-1 sec^-1,kTC2-=43.1(mol/L)^-1 sec^-1 for individual reaction channels.Degradation products were then identified as the hydroxylated and demethylated compounds.The result suggested a rate-limiting step of simple hydroxylation at the phenolic and/or alkene moieties,while the demethylation should be caused by the unavoidably formed manganese oxide via single electron oxidation.This is supported by the DFT calculation,indicating the primary oxidation of phenolic group of TC0 with activation barrier of 44.5 kcal/mol and of alkene group of TC-and TC2-with activation barriers of 44.0 and 43.4 kcal/mol,respectively.This is in agreement with the experimental results,implying the alternation of regioselectivity associated with the deprotonation process.The result was further supported by performing the Fukui function and electrostatic potential analysis,reflecting the more probable site and better electron donating tendency beneficial to the permanganate oxidation. | Xiaohua Jiang William AJefferson Dean Song Hanyang Cheng Fengmin Li Zhimin Qiang Aiqian Zhang Huijuan Liu Jiuhui Qu | 2020 | Journal of Environmental Sciences2020,32,1: | 1 |

| 14 | Evaluation of hydro-mechano-chemical behaviour of bentonite-sand mixtures显示文摘Bentonite-sand mixtures are widely used in engineering barrier of deep geological disposal of high-level radioactive nuclear waste and anti-seepage barrier of civil geotechnical engineering.Under the action of groundwater solution infiltration and external stress,the hydro-mechanical(HM)behaviour of bentonitesand mixtures,i.e.the swelling characteristics and permeability,will change.Once the anti-seepage and filtration effect is weakened or lost,the pollutants will spread to the biosphere.Therefore,it is necessary to study the swelling characteristics and permeability of bentonite-sand mixtures under coupled mechanochemical(MC)effect and to establish corresponding prediction model.For this reason,swelling tests under salt solution with different concentrations are conducted on pure bentonite and its mixtures with 30%,70%and 90%sand contents,the compression tests are carried out on saturated samples,and the saturated permeability coefficient k of the sample under each load is calculated by Terzaghi’s one-dimensional consolidation theory.The concepts of true effective stress pe,montmorillonite void ratio em and critical sand content as are introduced to determine the em-pe relationship and finally the k-em relationship of bentonite-sand mixtures.It is found that when the sand content aas,the em-pe relationship of the mixture is linear and independent of the salt solution concentration,and when a>as,the em-pe relationship of bentonite-sand mixture is bi-linear with the true effective deviatoric stress pesa as the intersection.In addition,the em-k relationship also shows the linear trend when aas,and the slope of the line increases with the increase of the salt solution concentration.When a>as,the k-em relationship will deviate from the linear relationship.Moreover,the larger the sand content is,the farther the deviation is.On the basis of summing the regularity,a model for predicting the HM behaviour of bentonite-sand mixture under the coupled MC effect is proposed.By comparing the swelling and permeability test results with model prediction results of different types of bentonite and its sand mixtures,the predictive model is verified.The study on the HM behaviour of bentonite-sand mixtures under salt solution infiltration and the model establishment can provide experimental and theoretical basis for the design and construction of anti-seepage engineering by bentonite-sand mixtures. | Wenjing Sun Cheng Liu Diansen Yang Dean Sun | 2022 | Journal of Rock Mechanics and Geotechnical Engineering2022,14,2: | 1 |

| 19 | Control Effect of UAV Low Volume Spray Technology on Thrips and Tomato Spotted Wilt Virus in Tobacco Field显示文摘[Objectives]The paper was to explore the influence of unmanned aerial vehicle(UAV)low volume spray technology on the control effect of viruliferous thrips and tomato spotted wilt virus(TSWV)under different pesticide concentrations,and to explore the effective control nodes and methods of thrips and TSWV.[Methods]According to the occurrence characteristics of thrips and TSWV,the field control effects of 4 pesticides on thrips and TSWV were tested by UAV with the thrips number and disease index as indicators,following the principle of regional and periodic control trials.[Results]In the groups of UAV and artificial prevention and control,4 agents were used alternatively,including 1%emamectin benzoate ME,1.7%abamectin imidacloprid ME,5%emamectin benzoate WDG,and 30%pyriproxyfen dinotefuran SL.There was no significant difference in the control effect of thrips and TSWV among treatment 1(recommended dosage of UAV agent),treatment 2(reduction of recommended dosage of UAV agent by 25%)and treatment 4(recommended dosage of artificial control agent),and the control effects on thrips were more than 83.16%in the 3 treatments.The disease index of TSWV in the 3 treatments decreased from 8.64±1.37 in the blank control group to less than 3,which reached the prevention and control threshold.But treatment 3(reduction of recommended dosage of UAV agent by 50%)did not reach the prevention and control threshold.The disease index of TSWV in the blank control area increased with the increase of the number of live nymphs of thrips,and there was a significant positive correlation,with good fitness.[Conclusions]UAV has a significant impact on the control effect of thrips and TSWV under different pesticide concentrations.In the actual flight control application,it is recommended that the amount of the pesticides is about 3/4 and not less than 1/2 of that of the artificial application.The control area should be extended to 100 m of the ridge of the tobacco field,and UAV is used periodically to control thrips and TSWV for 5 times from pre-transplanting stage to pre-squaring stage of flue-cured tobacco.The control effect is significantly different from the traditional artificial application. | Zefan LIU Dean XIE Jiabao HAN Yanfen XIE Li MA Siyuan GAO Ruiqi PENG Yuting ZHAO Xiangze FU Wengang CHENG Yonghong FENG Yan ZHANG Huaping ZHANG Ying HUANG Leijie DAI Lanfeng WANG | 2023 | Plant Diseases and Pests2023,14,4: | 0 |
