| 1 | Immune Responses in Wild-type Mice Against Prion Proteins Induced Using a DNA Prime-Protein Boost Strategy显示文摘Objective To break immune tolerance to prion (PrP) proteins using DNA vaccines.Methods Four different human prion DNA vaccine candidates were constructed based on the pcDNA3.1 vector:PrP‐WT expressing wild‐type PrP,Ubiq‐PrP expressing PrP fused to ubiquitin,PrP‐LII expressing PrP fused to the lysosomal integral membrane protein type II lysosome‐targeting signal,and PrP‐ER expressing PrP locating the ER.Using a prime‐boost strategy,three‐doses of DNA vaccine were injected intramuscularly into Balb/c mice,followed by two doses of PrP protein.Two weeks after the last immunization,sera and spleens were collected and PrP‐specific humoral and cellular immune responses evaluated by ELISA and ELISPOT tests.Results Higher levels of serum PrP antibodies were detected in mice vaccinated using the strategy of DNA priming followed by protein boosting.Of these,WT‐PrP,Ubiq‐PrP,and PrP‐LII induced significantly higher humoral responses.ELISPOT tests showed markedly increased numbers of IFN‐γ‐secreting T cells in mice vaccinated using the strategy of DNA priming followed by protein boosting after stimulation with recombinant PrP23‐90 and PrP23‐231.PrP‐ER inducedthe strongest T‐cell response.Conclusion Prion vaccines can break tolerance to PrP proteins and induce PrP‐specific humoral and cellular immune responses. | HAN YanLing LI Yuan SONG Juan WANG Ying SHI Qi CHEN Cao ZHANG BaoYun GUO Yan LI ChaoPing HAN Jun DONG XiaoPing | 2011 | Biomedical and Environmental Sciences2011,24,5: | 2 |

| 3 | Foam-like boron-doped g-C_(3)N_(4) as acid-base cooperative catalyst for efficient nucleophilic addition in water显示文摘Intramolecular acid-base cooperative catalysis is a powerful strategy to promote the nucleophilic additions in synthetic chemistry,since both substrates and nucleophiles can be activated simultaneously,yielding a lower energy reaction pathway.However,the systems often suffer from catalyst quenching due to the mutual neutralization of acid/base sites.Herein,we doped the boron(B)atoms in the tri-s-triazine ring of graphite-like carbon nitride(g-C_(3)N_(4))framework,to develop the acid-base cooperative catalyst for nucleophilic addition.Characterization results demonstrated a foam-like hierarchical porous structure,as well as the coexistence of Lewis acid and base sites in the as-synthesized samples.As a result,the B-doped g-C_(3)N_(4) acted as the efficient acidbase cooperative catalysts,significantly promoting the Knoevenagel condensation whose key step involves in nucleophilic addition.A wide range of α,β-unsaturated carbonyl compounds with remarkably high yields(>90%)was achieved even within 1 h in water,whereas the acid-free or base-free counterpart was far less efficient.In addition,the bifunctional catalysts exhibited excellent reusability with simple treatment.The novel B-doped g-C_(3)N_(4) catalytic system offers several advantages,such as operational simplicity,mild reaction conditions,high efficiency,and facile reuse of catalyst.All these advantages make it a benign protocol from the sustainability point of view,and inspired us to design the other bifunctional cooperative catalysts for a wide range of sustainable industrial applications. | Jiajun Wang Zewei Liu Shiye Li Chaoping Li Su Liu Jingwen Feng Rong Tan | 2021 | Green Chemical Engineering2021,2,2: | 1 |

| 8 | The difference of clinical characteristics of patients with COVID-19 in Fourth-tier city and Wuhan显示文摘Objective:This study mainly analyzed the clinical characteristics of patients with COVID-19 in Suining,one of China’s fourth-tier cities,and Wuhan and compared data between the 2 cities.Methods:A retrospective analysis of the clinical data of 68 patients with COVID-19 diagnosed at Wuhan Red Cross Hospital and 17 patients diagnosed at Suining Central Hospital,was conducted.Results:Among the 68 patients with COVID-19 in Wuhan,30(44.1%)had hypertension,and 25(36.8%)had diabetes.Three out of the 17 patients in Suining(17.6%)had hypertension,and 2 patients(11.8%)had diabetes.In the clinical classification,there were 1(5.9%)and 23(33.8%)patients with severe COVID-19 in Suining and Wuhan,respectively.Chest CT showed that lung consolidation occurred in 2(11.8%)and 26(38.2%)patients with COVID-19 in Suining and Wuhan,respectively.The proportion of patients with diabetes or hypertension,severe COVID-19,lung consolidation in Wuhan was significantly higher than that in Suining.The laboratory tests suggested that percentage of elevated C-reactive protein(CRP)(58.8%),ALT(33.8%),blood glucose(45.6%),creatine kinase(CK)(33.8%)or D-dimer(47.1%)of patients in Wuhan were significantly increased than those in Suining(29.4%,5.9%,17.6%,5.9%,and 17.7%,respectively).Conclusion:The incidence of COVID-19 in fourth-tier city and the disease severity were significantly lower than that in Wuhan. | Xiaojuan Wu Chaoping Wang Gaoyan He Baolin Jia Li Luo Xiaobin Luo | 2021 | Infectious Diseases Research2021,2,3: | 0 |
| 9 | Identification of crystal orientation for turbine blades with anisotropy materials显示文摘A novel approach to identify the crystal orientation of turbine blades with anisotropy materials is proposed. Based on enhanced mode basis, with the main advantages of its efficiency,accuracy and general applicability, the blade vibration mode of each order is linearly constructed by several specified mode shapes, which are obtained from the considered turbine blade with specified crystal orientations correspondingly. Then, a surrogate model based on Kriging method is introduced for constructing the condensed perturbed matrix of stiffness in order to improve the efficiency even further. The constructed surrogate model allows to perform the modal analysis of turbine blades with arbitrary crystal orientations in higher efficiency, due to the fact that the elements of condensed perturbed matrix of stiffness are considered in construction of the surrogate model rather than concerning the perturbation of all the elements of the initial stiffness matrix for the blade. Genetic algorithm is finally employed to optimize the defined fitness functions in order to identify the crystal orientation angles of turbine blades.
