1Modified constraint-induced movement therapy alters synaptic plasticity of rat contralateral hippocampus following middle cerebral artery occlusion显示文摘Modified constraint-induced movement therapy is an effective treatment for neurological and motor impairments in patients with stroke by increasing the use of their affected limb and limiting the contralateral limb.However,the molecular mechanism underlying its efficacy remains unclear.In this study,a middle cerebral artery occlusion(MCAO)rat model was produced by the suture method.Rats received modified constraint-induced movement therapy 1 hour a day for 14 consecutive days,starting from the 7^th day after middle cerebral artery occlusion.Day 1 of treatment lasted for 10 minutes at 2r/min,day 2 for 20 minutes at 2 r/min,and from day 3 onward for 20 minutes at 4 r/min.CatWalk gait analysis,adhesive removal test,and Y-maze test were used to investigate motor function,sensory function as well as cognitive function in rodent animals from the 1st day before MCAO to the 21^st day after MCAO.On the 21^st day after MCAO,the neurotransmitter receptor-related genes from both contralateral and ipsilateral hippocampi were tested by micro-array and then verified by western blot assay.The glutamate related receptor was shown by transmission electron microscopy and the glutamate content was determined by high-performance liquid chromatography.The results of behavior tests showed that modified constraint-induced movement therapy promoted motor and sensory functional recovery in the middle cerebral artery-occluded rats,but had no effect on cognitive function.The modified constraint-induced movement therapy upregulated the expression of glutamate ionotropic receptor AMPA type subunit 3(Gria3)in the hippocampus and downregulated the expression of the beta3-adrenergic receptor gene Adrb3 and arginine vasopressin receptor 1 A,Avprla in the middle cerebral artery-occluded rats.In the ipsilateral hippocampus,only Adra2 a was downregulated,and there was no significant change in Gria3.Transmission electron microscopy revealed a denser distribution the more distribution of postsynaptic glutamate receptor 2/3,which is an a-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid receptor,within 240 nm of the postsynaptic density in the contralateral cornu ammonis 3 region.The size and distribution of the synaptic vesicles within 100 nm of the presynaptic active zone were unchanged.Western blot analysis showed that modified constraint-induced movement therapy also increased the expression of glutamate receptor 2/3 and brain-derived neurotrophic factor in the hippocampus of rats with middle cerebral artery occlusion,but had no effect on Synapsin I levels.Besides,we also found modified constraint-induced movement therapy effectively reduced glutamate content in the contralateral hippocampus.This study demonstrated that modified constraint-induced movement therapy is an effective rehabilitation therapy in middle cerebral artery-occluded rats,and suggests that these positive effects occur via the upregulation of the postsynaptic membrane a-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid receptor expression.This study was approved by the Institutional Animal Care and Use Committee of Fudan University,China(approval No.201802173 S)on March 3,2018.Bei-Yao Gao Dong-Sheng Xu Pei-Le Liu Ce Li Liang Du Yan Hua Jian Hu Jia-Yun Hou Yu-Long Bai 2020Neural Regeneration Research2020,15,6:19
2Osteoclast-derived apoptotic bodies couple bone resorption and formation in bone remodeling显示文摘Bone remodeling is precisely coordinated by bone resorption and formation.Apoptotic osteoclasts generate large amounts of apoptotic bodies(ABs)marking the end of the bone resorption phase,whereas the functions of osteoclast-derived ABs remain largely unknown.Here,we identified the molecular profile of ABs derived from osteoclasts at distinct differentiation stages and investigated their corresponding functions.ABs were isolated from apoptotic bone marrow macrophages,preosteoclasts,and mature osteoclasts induced by staurosporine.Proteomic signature analysis with liquid chromatography-tandem mass spectrometry suggested marked protein cargo differences among the different ABs.Further bioinformatic analysis showed that the proteomic signatures of the ABs were highly similar to those of their parental cells.Functionally,pOC-ABs induced endothelial progenitor cell differentiation and increased CD31hiEmcnhi endothelial cell formation in a murine bone defect model via their PDGF-BB cargo.mOC-ABs induced osteogenic differentiation of mesenchymal stem cells and facilitated osteogenesis via RANKL reverse signaling.In summary,we mapped the detailed proteomic landscapes of ABs derived from osteoclasts and showed that their potential biological roles are important in coupling bone formation with resorption during bone remodeling.Qinyu Ma Mengmeng Liang Yutong Wu Fei Luo Zaisong Ma Shiwu Dong Jianzhong Xu Ce Dou 2021Bone Research2021,9,1:8
3Size,morphology,and composition of lunar samples returned by Chang'E-5 mission显示文摘This study focuses on the physical and chemical properties of surficial lunar regolith(LR)samples returned from the Moon by the Chang’E-5(CE-5)mission.Insights regarding the effect of a new sampling geological site on the surficial lunar sample CE5 C0400 were illustrated using nondestructive techniques such as laser diffractometry coupled with image analysis,X-ray computed tomography,and field emission scanning electron microscopy equipped with energy dispersive spectroscopy,and Xray diffraction combined with Rietveld refinement.From the characterization analyses,the CE-5 sampling site in the northeastern Oceanus Procellarum on the Moon yields a unique collection of relatively regular-shaped and fine basalt-dominated particles.The median grain size D_(50) is(55.24±0.96)μm,falling within the relatively low end of the range of the Apollo lunar returned samples.The coefficient of uniformity C_(u)of 15.1 and the coefficient of curvature C_(c)of 1.7 could classify CE5 C0400 to be well-graded.The minerals in CE5 C0400 comprise approximately 44.5%pyroxene,30.4%plagioclase,3.6%olivine,and6.0%ilmenite.There is a relatively low content of approximately 15.5%glass phase in the CE-5 lunar sample.From the results,we deduce that the CE-5 LR structure could have mainly resulted from micrometeoroid impacts to achieve such a high level of maturity.Hui Zhang Xian Zhang Guang Zhang Keqi Dong Xiangjin Deng Xiaosong Gao Yaodong Yang Yuan Xiao Xiao Bai Kaixin Liang Yiwei Liu Wenbin Ma Shaofan Zhao Ce Zhang Xiaojing Zhang Jian Song Wei Yao Hong Chen Weihua Wang Zhigang Zou Mengfei Yang 2022Science China(Physics,Mechanics & Astronomy)2022,65,2:4

6Redefinition and Formation Age of the Tanjianshan Group in Xitieshan Region, Qinghai显示文摘The Tanjianshan Group, which was previously divided into a, b, c and d formations, has been controversial for a long time. It mainly distributes in the northern margin of Qaidam Basin and is an important early Paleozoic greenschist facies metamorphic volcanic sedimentary rock formation. Detailed field investigation and zircon LA-ICPMS U-Pb dating of the key strata suggest that the original lower part of a Formation(a-1) versus the original middle upper of d Formation(d-3 and d-4), the original upper part of a Formation(a-2) and b Formation versus the original lower part of d Formation(d-1 and d-2) of Tanjianshan Group are contemporaneous heterotopic facies volcanicclasolite deposit, respectively. The former formations formed during the middle-late Ordovician(463–458 Ma), while the latter ones formed in the late Ordovician(about 445 Ma). The original c formation of Tanjianshan Group, which formed after 430 Ma, is similar to the Maoniushan Formation of Kunlun Mountains and north Qaidam Basin. According to the rules of stratigraphic division and naming, new stratum formations of Tanjianshan Group are re-built and divided into Duancenggou(O1-2td), Zhongjiangou(O2-3tz) and Xitieshan(O3tx) formations. The original c Formation is separated from Tanjianshan Group and is renamed as the Wuminggou Formation(S3-D1w), which shows a discordant contact with underlying Tanjianshan Group and overlying Amunike Formation(D3a). 7Multiple active components synergistically driven heteroatom-doped porous carbon as high-performance counter electrode in dye-sensitized solar cells显示文摘A facile template-free in situ self-activation approach for the multiple active components synergistically driven porous carbon was presented via a feasible annealing process.The biomass-derived carbon without additional activation reagents was fabricated using K-rich pomelo peel(PP)as the carbon source,which possesses a high electric conductivity where abundant functional hetero-metal atoms are doped into the carbon framework that playing the role of catalytic graphitization.The K^+that exists within the biomass can induce self-activation during pyrolysis apart from the activating gases during the pyrolysis process.The resulting electrocatalyst of PP-850(PP was pyrolyzed at 850°C in an N_2atmosphere)with abundant heteroatoms possesses a higher power conversion efficiency(PCE)of 7.81%as the counter electrode(CE)of dye-sensitized solar cells(DSCs)compared with the CEs calcinated at other temperatures and a similar PCE with Pt counterpart(8.24%)based on the liquid I_3^-/I^-electrolyte.The better electrocatalytic performance is attributed to the synergistic effect between self-activation and the co-doping of nitrogen,sulfur and phosphorus all together in a carbon matrix.Due to the feasibility of large-scale production,rich heteroatom doping,the PP-derived carbon,which simplifies the procedure and decreases the cost,has a potential application for an alternative electrocatalyst for high-performance photovoltaic devices.Hongyu Jing Danyang Wu Suxia Liang Xuedan Song Yonglin An Ce Hao Yantao Shi 2019Journal of Energy Chemistry2019,28,4:2

15Scattering of seismic waves by three-dimensional large-scale hill topography simulated by a fast parallel IBEM显示文摘To solve seismic wave scattering by a large-scale three-dimensional(3-D) hill topography, a fast parallel indirect boundary element method(IBEM) is developed by proposing a new construction method for the wave field, modifying the generalized minimum residual(GMRES) algorithm and constructing an Open MP plus MPI parallel model. The validations of accuracy and efficiency show that this method can solve 3-D seismic response of a large-scale hill topography for broadband waves, and overcome the weakness of large storage and low efficiency of the traditional IBEM. Based on this new algorithm architecture, taking the broadband scattering of plane SV waves by a large-scale Gaussian-shaped hill of thousands-meters height as an example, the influence of several important parameters is investigated, including the incident frequency, the incident angle and the height-width and length-width ratio of the hill. The numerical results illustrate that the amplification effect on the ground motion by a near-hemispherical hill is more significant than the narrow hill. For low-frequency waves, the scattering effect of the higher hill is more pronounced, and there is only a single peak near the top of the hill.
