| 4 | Characterizati0n,development and exploitation of EST-derived microsatellites in Gossypium raimondii Ulbrich显示文摘Microsatellite DNA or simple sequence repeats (SSRs) can be derived from expressed se- quence tags (ESTs). These markers are important for gene mapping as well as marker-assisted selection (MAS). To develop EST-SSRs for cotton gene map- ping, we selected and characterized functional markers in Gossypium raimondii, which consisted of 58906 non-redundant EST sequences from NCBI. Among them there were 2620 microsatellite se- quences containing 2818 EST-SSRs, which amoun- ted to 4.45% of the non-redundant starting sequence population. This incidence was equivalent to one EST-SSR in every 14.8 kb of G. raimondii genetic material. Among the different motifs ranging from 1 to 6 bp, trinucleotide repeats were most abundant (38.31%), followed by dinucleotide repeats (24.09%) and mononucleotide repeats (23.35%). Among all identified motif types, A/T had the highest frequency (18.67%), followed by AT/TA (14.83%). Among the compound motifs, tandem trinucleotides occurred with the highest frequency (48.65%). In all, we identi- fied 1554 EST-SSRs primer pair sequences. 300 of them were randomly selected to screen the poly- morphisms between the mapping parents G· hirsutum acc. TM-1 and G· barbadense cv. Hai7124, to con- struct linkage groups in cultivated allotetraploid cot- ton. Among them, 129 (43%) primer pairs were found to have polymorphisms. | 6 | Association analysis of fiber quality traits and exploration of elite alleles in Upland cotton cultivars/accessions(Gossypium hirsutum L.)显示文摘Exploring the elite alleles and germplasm accessions related to fiber quality traits will accelerate the breeding of cotton for fiber quality improvement. In this study, 99 Gossypium hirsutum L. accessions with diverse origins were used to perform association analysis of fiber quality traits using 97 polymorphic microsatellite marker primer pairs. A total of 107 significant marker-trait associations were detected for three fiber quality traits under three different environments, with 70 detected in two or three environments and 37 detected in only one environment. Among the 70 significant marker-trait associations, 52.86% were reported previously, implying that these are stable loci for target traits. Furthermore, we detected a large number of elite alleles associated simultaneously with two or three traits. These elite alleles were mainly from accessions collected in China, introduced to China from the United States, or rare alleles with a frequency of less than5%. No one cultivar contained more than half of the elite alleles, but 10 accessions were collected from China and the two introduced from the United States did contain more than half of these alleles. | 7 | Fluxes of CO_2,CH_4 and N_2O from alpine grassland in the Tibetan Plateau显示文摘Using stat ic chamber technique, fluxes of CO 2 , CH 4 and N 2 O were measured in the alpine grassland area from July 2000 to July 2001, d eterminations of mean fluxes showed that CO 2 and N 2 O were gene rally released from the soil, while the alpine grassland accounted for a weak CH 4 sink. Fluxes of CO 2 , CH 4 and N 2 O ranged widely. The highest CO 2 emission occurred in August, whereas a lmost 90% of the whole year emission occurred in the growing season. But the variations of CH 4 and N 2 O fluxes did not show any clear patterns over the one-year-experim ent. During a daily variation, the maximum CO 2 emission occurred at 16:00, and then decreased to the minimum emi ssion in the early morning. Daily pattern analyses indicated that the variation in CO 2 fluxes was positively related to air temperatures (R 2 =0.73) and soil temperatures at a depth of 5 cm (R 2 =0.86), whereas daily variations in CH 4 and N 2 O fluxes were poorly explained by soil temperatures and climatic va riables. | 10 | Fine mapping of the red plant gene R_1 in upland cotton (Gossypium hirsutum)显示文摘Sub 16 is a substitution line with G. hirsutum cv. TM-1 genetic background except that the 16th chro-mosome (Chr. 16) is replaced by the corresponding homozygous chromosome of G. barbadense cv. 3-79, and T586 is a G. hirsutum multiple gene marker line with 8 dominant mutation genes. The R1 gene for anthocyanin pigmentation was tagged in Chr. 16 in T586. The objective of this research was to screen SSR markers tightly linked with R1 by using the F2 segregating population containing 1259 plants derived from the cross of Sub 16 and T586 and the backbone genetic linkage map from G. hir-sutum×G. barbadense BC1 newly updated by our laboratory. Genetic analysis suggested that the seg-regation ratio of red plants in the F2 population fit Mendelian 1:2:1 inheritance, confirming that the red plant trait was controlled by an incomplete dominance gene. Preliminary mapping of R1 was conducted using 237 randomLy selected F2 individuals and JoinMap v3.0 software. Then, a fine map of R1 was constructed using the F2 segregating population containing 1259 plants, and R1 was located between NAU4956 and NAU6752, with only 0.49 cM to the nearest maker loci (NAU6752). | 11 | Shuanghuanglian injection downregulates nuclear factor-kappa B expression in mice with viral encephalitis显示文摘A mouse model of viral encephalitis was induced by intracranial injection of a Coxsackie virus B3 suspension. Quantitative real-time reverse transcription-PCR and western blot assay were applied to detect mRNA and protein expression of intelectin-2 and nuclear factor-kappa B in the viral encephalitis and control groups. Nuclear factor-kappa B and intelectin-2 mRNA and protein expression were significantly increased in mice with viral encephalitis. After intraperitoneal injection of Shuanghuanglian at a dose of 1.5 mg/kg for 5 successive days, intelectin-2 and nuclear factor-kappa B protein and mRNA expression were significantly decreased. To elucidate the relationship between intelectin-2 and nuclear factor-kappa B, mice with viral encephalitis were administered an intracerebral injection of 107 pfu recombinant lentivirus expressing intelectin shRNA. Both protein and mRNA levels of intelectin and nuclear factor-kappa B in brain tissue of mice were significantly decreased. | 12 | Adsorption behavior and mechanism of D113 resin for lanthanum显示文摘The sorption properties of macroporous weak acid resin (D113) for La3+ ion were studied by chemical analysis and IR spectra. Experimental results indicate that the D113 resin has a good adsorption ability for La3+ at pH = 6.0 in the HAc-NaAc medium. The statically saturated adsorption capacity is 273.3 mg/g. Separation coefficients of βLa3+/ Ce3+, βLa3+/ Gd3+, βLa3+/ Er3+, and βLa3+/ Y3+ are 2.29, 3.64, 4.27, and 0.627, respectively. The apparent activation energy of adsorption, Ea is 18.4 kJ/mol, the thermodynamics parameters H, S, and G of sorption are 4.53 kJ/mol, 61.8 J/(molK),-13.9 kJ/mol, respectively. The adsorption behavior of D113 for La3+ obeys the Freundlich isotherm. | 14 | Adsorption behavior of Pd(Ⅱ) from aqueous solutions by D201 resin显示文摘The adsorption and desorption behaviors of Pd(Ⅱ) on D201 resin were investigated. The effects of parameters including HCl concentrations, initial metal ion concentrations, contact time, and temperature on the adsorption of Pd(Ⅱ) were studied by batch method. The HCl concentration for studying the effects was set from 0.01 to 2 mol·L-1 and the optimal adsorption condition is in 0.01 mol·L-1 HCl solution. The adsorption isotherm of Pd(Ⅱ) on D201 resin was determined and the equilibrium data fitted the Langmuir model better than the Freundlich model. The maximum uptake capacity of Pd(Ⅱ) was 253 mg·g-1 at 298 K. The adsorption could be best described by the liquid film diffusion model. The enthalpy variation (ΔH) and entropy change (ΔS) of adsorption were equal to 2.49 kJ·mol-1 and 74.8 J·(mol·K)-1 respectively, indicating that the adsorption was endothermic in nature. Furthermore, the negative value of ΔG showed that movement of adsorbate from the solution to adsorbent surface was a spontaneous process. Pd(Ⅱ) could be eluted with 0.5 mol·L-1 HNO 3 solution. | 16 | The Achyranthes bidentata polypeptide k fraction enhances neuronal growth in vitro and promotes peripheral nerve regeneration after crush injury in vivo显示文摘We have previously shown that Achyranthes bidentata polypeptides(ABPP), isolated from Achyranthes bidentata Blume(a medicinal herb), exhibit neurotrophic and neuroprotective effects on the nervous system. To identify the major active component of ABPP, and thus optimize the use of ABPP, we used reverse-phase high performance liquid chromatography to separate ABPP. We obtained 12 fractions, among which the fraction of ABPPk demonstrated the strongest neuroactivity. Immunocytochemistry and western blot analysis showed that ABPPk promoted neurite growth in cultured dorsal root ganglion explant and dorsal root ganglion neurons, which might be associated with activation of Erk1/2. A combination of behavioral tests, electrophysiological assessment, and histomorphometric analysis indicated that ABPPk enhanced nerve regeneration and function restoration in a mouse model of crushed sciatic nerve. | 17 | Mining,characterization,and exploitation of EST-derived microsatellites in Gossypium barbadense显示文摘Simple sequence repeats(SSRs) have been widely applied as molecular markers in genetic studies.However,the number of expressed sequence tags(ESTs) and SSR markers from Gossypium barbadense is fewer than those from other cotton species.In this study,EST-SSR distribution from G.barbadense was characterized and new G.barbadense-derived EST-SSR markers were determined on the basis of the ESTs obtained by randomly sequencing 2 cDNA libraries associated with fiber development in G.barbadense.By mining 9697 non-redundant ESTs,a total of 638 SSR loci derived from 595 ESTs were observed.In G.barbadense,the frequency of ESTs containing SSRs was 6.13%,with an average of 1 SSR in every 10.4 kb of EST sequence.Furthermore,trinucleotide was found to be the most abundant repeat type among 2-6-nucleotide repeat types.It accounted for 26.6% of the total,followed by the hexanucleotide(26.0%) and pentanucleotide repeats(25.9%).Among all the repeat motifs,(AAG)n accounted for the highest proportion.EST-SSR primer pairs were developed using the Primer3 program,and the redundant primers were removed using the virtual PCR approach.As a result,380 non-redundant EST-SSR primer pairs were developed and used to detect polymorphisms between the mapping parents G.hirsutum 'TM-1' and G.barbadense 'Hai7124' for constructing linkage groups in cultivated allotetraploid cotton.Out of these,98(25.8%) primer pairs detected polymorphisms.Finally,95 polymorphic loci from 82 primer pairs were integrated into the backbone genetic map;of these,42 were mapped into the A subgenome and 53 into the D subgenome.The present work provided the foundation for constructing saturated genetic maps and conducting comparative genomic studies on different cotton species. | LU YuanDa CAI CaiPing WANG Lei LIN ShaoYan ZHAO Liang TIAN LiangLiang LU JunHong ZHANG TianZhen GUO WangZhen | 2010 | Chinese Science Bulletin2010,55,18: | 5 |

| 19 | Adaptive Spectr um Decision Method for Heterogeneous Cognitive Radio Networks显示文摘To improve spectrum utilization and minimize interference to Primary User (PU), an adaptive spectrum decision method is proposed for Secondary User (SU), while taking traffic load balancing and spectrum heterogeneity into consideration. Long-term statistics and current sensing results are integrated into the proposed decision method of spectrum access. Two decision methods, namely probability based and sensing based, are presented, compared and followed by performance analysis in terms of delay. For probability based spectrum decision, Short-Time-Job-First (STJF) priority queuing discipline is employed to minimize average residual time and theoretical conclusion is derived in a novel way. For sensing based decision we treat the interrupted service of SU as newly incoming and re-decision process is initialized to find available spectrum in a First-Available-First-Access (FAFA) fashion. Effect of sensing error in PHY layer is also analyzed in terms of extended average residual time. Simulation results show that, for relatively low arriving rate of SU traffic, the proposed spectrum decision method yields at least a delay reduction of 39.5% compared with non-adaptive method.
