| 1 | Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual. | Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang | 2020 | Protein & Cell2020,11,4: | 44 |

| 4 | N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. | 6 | CXCR7/CXCL12 axis is involved in lymph node and liver metastasis of gastric carcinoma显示文摘AIM To investigate the role of CXC chemokine receptor(CXCR)-7 and CXCL12 in lymph node and liver metastasis of gastric carcinoma.METHODS In 160 cases of gastric cancer, the expression of CXCR7 and CXCL12 in tumor and matched tumoradjacent non-cancer tissues, in the lymph nodes around the stomach and in the liver was detected using immunohistochemistry to analyze the relationship between CXCR7/CXCL12 expression and clinicopathological features and to determine whether CXCR7 and CXCL12 constitute a biological axis to promote lymph node and liver metastasis of gastric cancer. Furthermore, the CXCR7 gene was silenced and overexpressed in human gastric cancer SGC-7901 cells, and cell proliferation, migration and invasiveness were measured by the MTT, wound healing and Transwell assays, respectively. RESULTS CXCR7 expression was up-regulated in gastric cancer tissues(P = 0.011). CXCR7/CXCL12 expression was significantly related to poor tumor differentiation, high tumor stage and lymph node(r = 0.338, P = 0.000) and liver metastasis(r = 0.629, P = 0.000). The expression of CXCL12 in lymph node and liver metastasis was higher than that in primary gastric cancer tissues(χ2 = 6.669, P = 0.010; χ2 = 25379, P = 0.000), and the expression of CXCL12 in lymph node and liver metastasis of gastric cancer was consistent with the positive expression of CXCR7 in primary gastric cancer(r = 0.338, P = 0.000; r = 0.629, P = 0.000). Overexpression of the CXCR7 gene promoted cell proliferation, migration and invasion. Silencing of the CXCR7 gene suppressed SGC-7901 cell proliferation, migration and invasion. Human gastric cancer cell lines expressed CXCR7 and showed vigorous proliferation and migratory responses to CXCL12.CONCLUSION The CXCR7/CXCL12 axis is involved in lymph node and liver metastasis of gastric cancer. | 9 | Study of HIV-1 Drug Resistance in Patients Receiving Free Antiretroviral Therapy in China显示文摘To investigate the prevalence of drug-resistance mutations,resistance to antiretroviral drugs,and the subsequent virological response to therapy in treatment-naive and antiretroviral-treated patients infected with HIV/AIDS in Henan,China,a total of 431 plasma samples were collected in Queshan county between 2003 and 2004,from patients undergoing the antiretroviral regimen Zidovudine + Didanosine + Nevirapine(Azt+Ddi+Nvp).Personal information was collected by face to face interview.Viral load and genotypic drug resistance were tested.Drug resistance mutation data were obtained by analyzing patient-derived sequences through the HIVdb Program(http://gffzz38c50ed9119344cfhc65fpuxxpqqf6f5b.ffgz.tsg.suse.edu.cn).Overall,38.5% of treatment-naive patients had undetectable plasma viral load(VL),the rate significantly increased to 61.9% in 0 to 6 months treatment patients(mean 3 months)(P<0.005)but again significantly decrease to 38.6% in 6 to 12 months treatment patients(mean 9 months)(P<0.001)and 40.0% in patients receiving more than 12 months treatment(mean 16 months)(P<0.005).The prevalence of drug resistance in patients who had a detectable VL and available sequences were 7.0%,48.6%,70.8%,72.3% in treatment-na?ve,0 to 6 months treatment,6 to 12 months treatment,and treatment for greater than 12 months patients,respectively.No mutation associated with resistance to Protease inhibitor(PI)was detected in this study.Nucleoside RT inhibitor(NRTI)mutations always emerged after non-nucleoside RT inhibitor(NNRTI)mutations,and were only found in patients treated for more than 6 months,with a frequency less than 5%,with the exception of mutation T215Y(12.8%,6/47)which occurred in patients treated for more than 12 months.NNRTI mutations emerged quickly after therapy begun,and increased significantly in patients treated for more than 6 months(P<0.005),and the most frequent mutations were K103N,V106A,Y181C,G190A.There had been optimal viral suppression in patients undergoing treatment for less than 6 months in Queshan,Henan.The drug resistance strains were highly prevalent in antiretroviral-treated patients,and increased with the continuation of therapy,with many patients encountering virological failure after 6 months therapy. | Xin-ping LIP Hui XING Zhe WANG Xue-feng SI Lian-en WANG Hua CHENG Wei-guo CUI Shu-lin JIANG Ling-jie LIAO Hai-wei ZHOU Jiang-hong HUANG Hong PENG Peng-fei MA Yi-ming SHAO | 2007 | 中国病毒学2007,22,3: | 9 |

| 12 | Colonic stenting vs emergent surgery for acute left-sided malignant colonic obstruction:A systematic review and meta-analysis显示文摘AIM:To investigate the effects of emergent preopera-tive self-expandable metallic stent (SEMS) vs emer-gent surgery for acute left-sided malignant colonic obstruction. METHODS:Two investigators independently searched the MEDLINE, EMBASE and Cochrane Central Register of Controlled Trials, as well as references of included studies to identify randomized controlled trials (RCTs) that compared two or more surgical approaches for acute colonic obstruction. Summary risk ratios (RR) and 95% CI for colonic stenting and emergent surgery were calculated. RESULTS:Eight studies met the selection criteria, involving 444 patients, of whom 219 underwent SEMS and 225 underwent emergent surgery. Seven studies reported difference of the one-stage stoma rates between the two groups (RR, 0.60; 95% CI:0.48-0.76; P < 0.0001). Only three RCTs described the follow-up stoma rates, which showed no significant difference between the two groups (RR, 0.80; 95% CI:0.59-1.08; P = 0.14). Difference was not significant in the mortality between the two groups (RR, 0.91; 95% CI:0.50-1.66; P = 0.77), but there was significant difference (RR, 0.57; 95% CI:0.44-0.74; P < 0.0001) in the overall morbidity. There were no significant differences between the two groups in the anastomotic leak rate (RR, 0.60; 95% CI: 0.28-1.28; P = 0.19), occurrence of abscesses, including peristomal abscess, intraperitoneal abscess and parietal abscess (RR, 0.83; 95% CI:0.36-1.95; P = 0.68), and other abdominal complications (RR: 0.67; 95% CI: 0.40-1.12; P = 0.13). | 14 | Flame-retardant concentrated electrolyte enabling a Li F-rich solid electrolyte interface to improve cycle performance of wide-temperature lithium–sulfur batteries显示文摘Lithium–sulfur batteries have been regarded as the most promising high-energy electrochemical energy storage device owing to the high energy density, low cost and environmental friendliness. However, traditional lithium–sulfur batteries using ether-based electrolytes often suffer from severe safety risks(i.e. combustion). Herein, we demonstrated a novel kind of flame-retardant concentrated electrolyte(6.5 M lithium bis(trifluoromethylsulphonyl)imide/fluoroethylene carbonate) for highly-safe and widetemperature lithium–sulfur batteries. It was found that such concentrated electrolyte showed superior flame retardancy, high lithium-ion transference number(0.69) and steady lithium plating/stripping behavior(2.5 m Ah cm^(-2) over 3000 h). Moreover, lithium–sulfur batteries using this flame-retardant concentrated electrolyte delivered outstanding cycle performance in a wide range of temperatures(-10 °C, 25 °C and 90 °C). This superior battery performance is mainly attributed to the LiF-rich solid electrolyte interphase formed on lithium metal anode, which can effectively suppress the continuous growth of lithium dendrites. | 16 | In vitro Anti-viral Activity of the Total Alkaloids from Tripterygium hypoglaucum against Herpes Simplex Virus Type 1显示文摘Herpes simplex virus type 1 (HSV-1) is a commonly occurring human pathogen worldwide. There is an urgent need to discover and develop new alternative agents for the management of HSV-1 infection. Tripterygium hypoglaucum (level) Hutch (Celastraceae) is a traditional Chinese medicine plant with many pharmacological activities such as anti-inflammation, anti-tumor and antifertility. The usual medicinal part is the roots which contain about a 1% yield of alkaloids. A crude total alkaloids extract was prepared from the roots of T. hypoglaucum amd its antiviral activity against HSV-1 in Vero cells was evaluated by cytopathic effect (CPE) assay, plaque reduction assay and by RT-PCR analysis. The alkaloids extract presented low cytotoxicity (CC50 = 46.6 μg/mL) and potent CPE inhibition activity, the 50% inhibitory concentration (IC50) was 6.5 μg/mL, noticeably lower than that of Acyclovir (15.4 μg /mL). Plaque formation was significantly reduced by the alkaloids extract at concentrations of 6.25 μg/mL to 12.5 μg/mL, the plaque reduction ratio reached 55% to 75% which was 35% higher than that of Acyclovir at the same concentration. RT-PCR analysis showed that, the transcription of two important delayed early genes UL30 and UL39, and a late gene US6 of HSV-1 genome all were suppressed by the alkaloids extract, the expression inhibiting efficacy compared to the control was 74.6% (UL30), 70.9% (UL39) and 62.6% (US6) respectively at the working concentration of 12.5μg/mL. | 17 | Is the Chinese Medicinal Formula Guipi Decoction(归脾汤) Effectiveas AnAdjunctive Treatment for Depression?AMeta-Analysis of Randomized Controlled Trials显示文摘Objectives: To evaluate the effectiveness and safety of Guipi Decoction(归脾汤, GPD) as an adjunctive in the treatment of depression. Methods: A review of all relevant studies retrieved from a search of the following databases were conducted without any language restriction: Excerpt Medica Database(EMBASE), Pub Med, Cochrane Central Register of Controlled Trials, China National Knowledge Infrastructure(CNKI), VIP Information, Wanfang Data, and the Chinese Biomedical Literature Database. Papers published until February 2013 were taken into consideration. The analysis was performed using the Cochrane software Revman 5.1. Results: Nine randomized controlled trials involving 620 patients with depression were included in this review. The meta-analysis revealed that compared with antidepressant therapy alone, treatment with a combination of GPD and an antidepressant drug significantly improved the symptoms of depression [weighted mean difference(WMD): –3.09; 95% confidence interval(CI): –4.11 to –2.07] and increased the rates of effectiveness(OR: 4.75; 95% CI: 2.66–8.51) as well as recovery(OR: 1.73; 95% CI: 1.17–2.56). The adverse effects of GPD were not found to be significant in these studies. Conclusions: The findings of this meta-analysis were in keeping with the notion that GPD formulations were effective in the treatment of depression without causing any severe adverse effects. | 18 | Characteristics of carbon storage in Shanghai's urban forest显示文摘Urban forest has undergone rapid development in China over the last three decades because of the acceleration of urbanization.Urban forest thus plays an increasingly important role in carbon sequestration at a regional and national scale.As one of the most urbanized cities in China,Shanghai showed an increase of forest coverage from 3% in the 1990s to 13% in 2009.Based on CITY-green model and the second soil survey of Shanghai,the forest biomass carbon(FBC) was estimated to be 0.48 Tg in the urban area and,forest soil organic carbon(SOC)(0-100 cm soil depth) is 2.48 Tg in the urban and suburban areas,respectively.These values are relatively within the median and lower level compared with other Chinese megacities,with the FBC of 0.02 Tg in Harbin to 47.29 Tg in Chongqing and the forest SOC of 1.74 Tg in Nanjing to 418.67 Tg in Chongqing.For the different land-use types in Shanghai,the SOC density ranges from 13.8(tidal field) to 38.6 t ha-1(agricultural land).On average,the forest SOC density(31.5 t ha-1) in Shanghai is lower than that in agricultural lands(38.6 t ha-1) and higher than that in lawns(26.5 t ha-1) and gardens(21.3 t ha-1).In Shanghai,the SOC density in newly established urban parks is generally lower than that in older parks.In the northern and southeastern suburban areas(e.g.,Baoshan,Yangpu,and Nanhui districts),greenspace SOC density is higher than that in the central commercial areas(Hongkou,Putuo,Changning,and Zhabei districts) and in newly developed district(Pudong District).Uncertainties still exist in the estimation of urban forest carbon in Shanghai,as well as in other Chinese cities.Thus,future research directions are also discussed in this paper. | WANG Zhe CUI Xuan YIN Shan SHEN GuangRong HAN YuJie LIU ChunJiang | 2013 | Chinese Science Bulletin2013,58,10: | 6 |
| 19 | Redo Coronary Artery Bypass Grafting: On-Pump and Off-Pump Coronary Artery Bypass Grafting Revascularization Techniques显示文摘Objective To analyze the short-term outcomes of redo coronary artery bypass grafting(CABG) using on-pump and off-pump CABG techniques. Methods From January 2003 to August 2013, non-randomized 80 patients were treated with redo CABG in the Department of Cardiac Surgery, Peking University Third Hospital. Among these patients, 40 underwent on-pump CABG technique(redo-ONCAB group) and 40 underwent off-pump CABG technique(redo-OPCAB group). Furthermore, transmyocardial laser revascularization was performed in high-risk patients who were not suitable to conventional grafting. Clinical data of the two groups were recorded and analyzed including operation time, coronary grafts, incomplete revascularization, postoperative ventilation, perioperative stroke, and low output syndrome, etc. Results There were no significantly differences in age, gender distribution, incidences of hypertension, stroke, and other clinical characteristics between redo-OPCAB group and redo-ONCAB group(all P>0.05), except for incidences of renal dysfunction and pulmonary disease(all P<0.05). The number of grafting vessels in the redo-ONCAB and redo-OPCAB groups was 2.1 ± 0.74 and 1.4 ±0.52 respectively. There was significant difference between the two groups(P=0.0243). Compared with the redo-ONCAB group, there was shorter operation time(P=0.0045), postoperative ventilation(P=0.0211) and intensive care unit stay(P=0.0400), as well as fewer use of platelet(P=0.0338) and blood transfusion(P=0.0034) in the redo-OPCAB group. The incidence of incomplete revascularization(P=0.0253) and the use of transmyocardial laser revascularization(P=0.0052) were higher in the redo-OPCAB group than those in the redo-ONCAB group(all P<0.05). However, no significant differences were showed for the incidence of the use of intra aortic balloon pump and continuous renal replacement therapy, perioperative stroke, low output syndrome, and in-hospital mortality between the two groups(all P>0.05). | 20 | Human tissue kallikrein-1 protects against the development of erectile dysfunction in a rat model of hyperhomocysteinemia显示文摘The aim of this study was to investigate the mechanism by which a diet inducing high hyperhomocysteinemia(HHcy)leads to the deterioration of erectile function in rats and whether this is inhibited by expression of the human tissue kallikrein-1(hKLKl)gene.We established a rat model of HHcy by feeding methionine(Met)-rich diets to male Sprague-Dawley(SD)rats.Male wild-type SD rats(WTRs)and transgenic rats harboring the hKLKl gene(TGRs)were fed a normal diet until 10 weeks of age.Then,30 WTRs were randomly divided into three groups as follows:the control(n=10)group,the low-dose(4%Met,n=10)group,and the high-dose(7%Met,n=10)group.Another 10 age-matched TGRs were fed the high-dose diet and designated as the TGR+7%Met group.After 30 days,in all four groups,erectile function was measured and penile tissues were harvested to determine oxidative stress,endothelial cell content,and penis fibrosis.Compared with the 7%Met group,the TGR+7%Met group showed diminished HHcy-induced erectile dysfunction(ED),indicating the improvement caused by hKLKl.Regarding corpus cavernosum endothelial cells,hKLKl preserved endothelial cell-cell junctions and endothelial cell content,and activated protein kinase B/endothelial nitric oxide synthase(Akt/eNOS)signaling.Fibrosis assessment indicated that hKLKl preserved normal penis structure by inhibiting apoptosis in the corpus cavernosum smooth muscle cells.Taken together,these findings showed that oxidative stress,impaired corpus cavernosum endothelial cells,and severe penis fibrosis were involved in the in duction of ED by HHcy in rats,whereas hKLKl preserved erectile function by inhibiting these pathophysiological changes. | Kai Cui Yang Luan Zhe Tang Chuan-Chang Li Tao Wang Shao-Gang Wang Zhong Chen Ji-Hong Liu | 2019 | Asian Journal of Andrology2019,21,5: | 4 |
