1Expression of lysosome-associated protein transmembrane 4B-35 in cancer and its correlation with the differentiation status of hepatocellular carcinoma显示文摘AIM: To produce high-quality polydonal antibody to lysosome associated protein transmembrane 4B-35 and to identify LAPTM4B-35 expression in cancer tissues and its correlation with differentiation status of hepatocellular carcinoma (HCC).METHODS: The 297 bp 5' end of LAPTM4BcDNA was obtained by PCR and inserted into prokaryotic expression vector pGEXKG. Then the recombinant pGEX-KG-N1-99 was transformedinto E. coli JM109 to express GST-fusion protein. The fusion protein was purified by glutathione sepharoseTM 4B agarose. The purified GST-LAPTM4B-N1-99 was characterized by SDSPAGE, and used to immunize rabbits. The titer and specificity of antisera were detected by ELISA and Western blot, respectively. The correlation between the expression levels of LAPTM4B-35 and the differentiation status of HCC was analyzed via Western blot. The expression of LAPTM4B35 in HCC and other six cancer tissues was investigated via tissue chip and immunohistochemical analysis. RESULTS: About 6.2 mg of pure GST-LAPTM4B-N1-99 was isolated from 1 L of bacteria. The GST-LAPTM4B-N1-99produced high titer antisera in rabbits and showed good immunity. Western blot showed specific reactions for the antibody to the LAPTM4B-35 in the total proteins from HCC tissues and BEL-7402 cells, also to the fusion protein purified or in the transformed bacteria. LAPTM4B-35 was remarkably expressed in several cancers, such as HCC, breast cancer, gastric carcinoma, lung cancer, and colon carcinoma, but not commonly expressed in esophageal cancer and rectum carcinoma. Notably, the expression levels of LAPTM4B-35 were significantly and inversely correlated to the differentiation of HCCs in a 20 case analysis. CONCLUSION: Specific polyclonal antibody (LAPTM4B-N1-99-pAb) to LAPTM4B-35 was produced. It identified the expression of LAPTM4B-35 in some cancer tissues originated from single layer cuboidal and columnar epithelial cells and firmly demonstrated that the expression of LAPTM4B-35 in HCC was inversely correlated with the differentiation of HCC.CongPeng Rou-LiZhou Gen-ZeShao Jing-AnRui Shao-BinWang MingLin ShaZhang Zi-FengGao 2005World Journal of Gastroenterology2005,11,18:9
2中国陆地生态系统近 2 0年碳空间动态的初步研究(英文)显示文摘陆地生态系统的净生产力 (NEP)是生态系统净初级生产力 (NPP)和异氧呼吸 (Rh)之差。在全球尺度上 ,反映NPP和Rh之差的NEP直接揭示陆地生态系统与大气系统之间的二氧化碳交换 ,即碳平衡 ,因此 ,意义重大。文章简短回顾了关于中国陆地生态系统碳平衡的研究状况。由于植物根部呼吸很难从土壤表面总二氧化碳 (CO2 )通量中分开 ,因此直接从野外测量土壤异氧呼吸几乎是不可能的。虽然由于像诸如火灾、森林砍伐、土地利用变化及气候和大气变化等干扰因素 ,全球陆地生态系统很大程度上处于非平衡态 ,利用生态系统在平衡态时NPP等于Rh这一事实 ,我们估算了全球和中国森林生态系统的土壤异氧呼吸。利用遥感和地面的NPP观测数据 ,我们也估算了中国森林生态系统逐月的净生产力 (1982— 1998)。NPP的估算主要采用NOAA卫星AVHRR 8km的NDVI数据 ,结合地面气候数据完成。土壤呼吸是通过地面观测数据与温度和降水的关系得到的。在此基础上我们得到了中国陆地生态系统在过去近 2 0年中碳的动态变化 ,并给出了初步结果。CONGPeng XUMing2002地学前缘2002,9,1:4
3Multiphase microstructure formation and its effect on fracture behavior of medium carbon high silicon high strength steel显示文摘This work investigated the evolution of multiphase microstructure and impact fracture behavior of medium carbon high silicon high strength steel subjected to the austempering treatment at 240,360,and400℃.The results show that martensite,bainite,and retained austenite(RA)are the main microstructural phases.The austempering treatments at 360 and 400℃ caused the formation of carbon-poor ferrite in the matrix,and the transformation of ultrafine bainite into coarse lath bainite and granular bainite,respectively.Thick filmy RA was distributed between bainite laths.The polygonal martensiteaustenite islands and blocky RA formed along the grain boundaries.The average carbon concentration in the matrix decreased with the temperature increase,while the impact toughness initially increased and then dropped with temperature.The quasi-cleavage brittle fracture dominated the impact fracture mechanism of the sample austempered at 240℃ by forming tearing surfaces and tearing steps.The microcracks disappeared in the RA on the prior austenite grain boundaries.On the other side,the fracture surface of the sample austempered at 360℃ exhibited ductile fracture with deep dimples and brittle fracture with cleavage river patterns.The polygonal martensite-austenite islands or blocky RA constrained the microcracks.After austempered at 400℃,the brittle fracture was dominant,showing river patterns,and the microcracks propagated through the granular bainite without any resistance.Kui Chen Huabing Li Zhouhua Jiang Fubin Liu Congpeng Kang Xiaodong Ma Baojun Zhao 2021Journal of Materials Science & Technology2021,,13:3

6Fine-tuning brassinosteroid biosynthesis via 3′UTR-dependent decay of CPD mRNA modulates wood formation in Populus显示文摘Brassinosteroids(BRs)are plant hormones that regulate wood formation in trees.Currently,little is known about the post-transcriptional regulation of BR synthesis.Here,we show that during wood formation,fine-tuning BR synthesis requires 3′UTR-dependent decay of Populus CONSTITUTIVE PHOTOMORPHOGENIC DWARF 1(PdCPD1).Overexpression of PdCPD1 or its 3′UTR fragment resulted in a significant increase of BR levels and inhibited secondary growth.In contrast,transgenic poplars repressing PdCPD13′UTR expression displayed moderate levels of BR and promoted wood formation.We show that the Populus GLYCINE-RICH RNA-BINDING PROTEIN1(PdGRP1)directly binds to a GU-rich element in 3′UTR of Pd CPD1,leading to its mRNA decay.We thus provide a post-transcriptional mechanism underlying BRs synthesis during wood formation,which may be useful for genetic manipulation of wood biomass in trees.Dian Wang Xiaoning Hao Li Xu Mengyan Zhao Congpeng Wang Xihao Yu Yingzhen Kong Mengzhu Lu Gongke Zhou Guohua Chai Xianfeng Tang 2023Journal of Integrative Plant Biology2023,65,8:0
7Investigation on the Metabolism of Salvianolic acid B, Tanshinone Ⅱ A, Ginsenoside Rg1 and Ginsenoside Rb1 from Danshen-Sanqi Herbal Pair in Zebrafish by Liquid Chromatography-Tandem Mass Spectrometry Analysis显示文摘Background:Salvianolic acid B(SAB),tanshinone IIA(TIIA),ginsenoside Rg1(GRg1)and ginsenoside Rb1(GRb1)are the main active components of Danshen-Sanqi herbal pair(DS-SQ),which is widely used in the treatment of cardiovascular diseases in Asia.The compatibility mechanisms of compound Chinese herbal preparation may be clarified to some degree by understanding the interactions of the active components based on their metabolic research.Furthermore,zebrafish is a convenient and feasible model to study the metabolism of drugs.Objective:To investigate the metabolism of SAB,TIIA,GRg1 and GRb1 in zebrafish by liquid chromatography-tandem mass spectrometry(LC-MS/MS)analysis.Methods:Zebrafish embryos after 48-hour hatching were divided into nine experimental groups.The blank group was exposed to 1 mL ultra-pure water.Eight drug-treated groups were exposed to 1 mL solution of SAB,TIIA,GRg1,GRb1,SAB and GRg1,SAB and GRb1,TIIA and GRg1,TIIA and GRb1,respectively.After homogenization,they were analyzed by LC-MS/MS.The Structures of the metabolites were determined by analyzing their corre-sponding mass spectra information and comparing the data with relevant literature.According to the change of the amount of metabolites,the metabolic effect between SAB,TIIA,GRg1 and GRb1 were investigated.Results:Eighteen metabolites and four parent drugs of SAB,TIIA,GRg1 and GRb1 in zebrafish were identified by LC-MS/MS,including methylation,degradation,reduction,dehydrogenation,hydroxylation,desugarization and glucuronidation metabolites.Furthermore,after being combined exposed of drugs,GRg1 and GRb1 could promote the metabolism of TIIA,while SAB and TIIA could promote the metabolism of GRg1 and GRb1 to different degree.Conclusion:By using LC-MS/MS analysis,zebrafish was successfully applied to study the metabolism of SAB,TIIA,GRg1 and GRb1,which is significance to explain the possible synergistic effect of components in DS-SQ through modifying the metabolism in zebrafish.This study can be helpful for the further research of DS-SQ.Shijun Yin Congpeng Zhao Guang Hu Hua Chen Fengqing Yang 2021Clinical Complementary Medicine and Pharmacology2021,1,1:0
