| 1 | Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. | 2 | Emerging role of Hippo pathway in gastric and othergastrointestinal cancers显示文摘More evidence has underscored the importance of Hippo signaling pathway in gastrointestinal tissue homeostasis, whereas its deregulation induces tumorigenesis. Yes-associated protein 1(YAP1) and its close paralog TAZ, transcriptional co-activator with a PDZbinding motif, function as key effectors negatively controlled by the Hippo pathway. YAP1/TAZ exerts oncogenic activities by transcriptional regulation via physical interaction with TEAD transcription factors. In various cancers, Hippo pathway cross-talks with pro- or anti-tumorigenic pathways such as GPCR, Wnt/β-catenin, Notch and TGF-β signaling and is deregulated by multiple factors including cell density/junction and micro RNAs. As YAP1 expression is significantly associated with poor prognosis of gastric and other gastrointestinal cancers, detailed delineation of Hippo regulation in tumorigenesis provides novel insight for therapeutic intervention. | 3 | The record of paleoclimatic change from stalagmites and the determination of termination II in the south of Guizhou Province, China显示文摘A high-resolution climate record from 163.00 kaBP to 113.80 kaBP has been obtained through TIMS-U series dating and carbon and oxygen isotope analysis of the three large stalagmites from two caves in the south of Guizhou Province, China. The record of the oxygen isotopes from the stalagmites reveals that the undulation characteristics between the cooling event of the glacial period and the warming event of the interglacial period in the research area can compare well to those of ice cores, lake sediments, loess and deep sea sediments on the scale of ten-thousand years or millennium time scale. The climate undulation provided by the record of the stalagmites has a coherence with the global changes and a tele-connection to the paleoclimate changes in the north polar region. Our results suggest that the direct dynamics of paleo-monsoon circulation changes reflected in the record of the stalagmites might be caused by changes of the global ice volume, and in turn related to various factors, including the solar radiation strength at the mid-latitudes in the Northern Hemisphere, the southern extension of the ice-rafted event in the North Atlantic, and changes of the equatorial Pacific sea surface temperature at the low-latitudes. Using δ18O values, we have calculated the temperatures and the results show that the temperature difference between the penultimate glacial period (with an average temperature of 8.1℃, and a minimum temperature range from 0.65℃ to -1.43℃ at stage 6 ) and the last interglacial period (with an average temperature of 18.24℃ at sub-stage 5e ) was about 10℃. This temperature difference from the record of the stalagmites corresponds in general to the record temperature variation (about 10℃) of measured ice cores. The climate records from the three stalagmites in the two caves have shown that the circulation strength of the Asian summer monsoon and the winter monsoon in the penultimate glacial period and the last interglacial period had a clear change. With the TIMS-U series method, termination II of the penultimate glacial period has been precisely dated at an age of (129.28±1.10) kaBP for the three stalagmites in the south of Guizhou Province, China. This borderline age represents the beginning of the last interglacial period or the boundary between the Middle Pleistocene and the Late Pleistocene, and corresponds to the beginning age of the last interglacial period shown by the ice cores and in the SPECMAP curve of the marine oxygen isotopes. | 6 | Synthesis of vertically aligned carbon nanotube arrays on silicon substrates显示文摘The growth of well-aligned carbon nanotube (CNT) arrays using a heat chemical vapor deposition system on silicon substrates is reported. The growth properties of CNT arrays are studied as a function of synthesis conditions. It is found that 750℃ and 10 nm Fe film are suitable conditions for the growth of well-aligned CNT arrays. CNT arrays with a uniform diameter, thick tube wall and firm cohesion to the Si substrate can be grown for C2H2 concentration of 27%.
