| 1 | Epigenetic regulation: methylation of histone and non-histone proteins显示文摘Histone methylation is believed to play important roles in epigenetic memory in various biological processes. However, questions like whether the methylation marks themselves are faithfully transmit- ted into daughter cells and through what mechanisms are currently under active investigation. Previ- ously, methylation was considered to be irreversible, but the recent discovery of histone lysine de- methylases revealed a dynamic nature of histone methylation regulation on four of the main sites of methylation on histone H3 and H4 tails (H3K4, H3K9, H3K27 and H3K36). Even so, it is still unclear whether demethylases specific for the remaining two sites, H3K79 and H4K20, exist. Furthermore, be- sides histone proteins, the lysine methylation and demethylation also occur on non-histone proteins, which are probably subjected to similar regulation as histones. | 6 | Intrinsic control of axon regeneration显示文摘Spinal cord injury disrupts the connections between the brain and spinal cord, often resulting in the loss of sensory and motor function below the lesion site. The most important reason for such permanent functional deficits is the failure of injured axons to regenerate after injury. In principle, the functional recovery could be achieved by two forms of axonal regrowth: the regeneration of lesioned axons which will reconnect with their original targets and the sprouting of spared axons that form new circuits and compensate for the lost function. Our recent studies reveal the activity of the mammalian target of rapamycin (mTOR) pathway, a major regulator of new protein synthesis, as a critical determinant of axon regrowth in the adult retinal ganglion neurons[1]. | 7 | Second-order consensus in networks of dynamic agents with communication time-delays显示文摘This paper proposes second-order consensus protocols with time-delays and gives the measure of the robustness of the protocols to the time-delay existing in the network of agents with second-order dynamics. By employing a frequency domain method, it is proven that the information states and their time derivatives of all the agents in the network achieve consensus asymptotically, respectively, for appropriate communication timedelay if the topology of weighted network is connected. Particularly, a tight upper bound on the communication time-delay that can be tolerated in the dynamic network is found. The consensus protocols are distributed in the sense that each agent only needs information from its neighboring agents, which reduces the complexity of connections between neighboring agents significantly. | 8 | Inducible resistance to Fas-mediated apoptosis in B cells显示文摘Apoptosis produced in B cells through Fas (APO-1, CD95) triggering is regulated by signals derived from other surface receptors: CD40 engagement produces upregulation of Fas expres. sion and marked susceptibility to Fas-induced cell death, whereas antigen receptor engagement, or IL-4R engagement, inhibits Fas killing and in so doing induces a state of Fas-resistance, even in otherwise sensitive, CD40-stimulated targets. Surfaceim. munoglobulin and IL-4R utilize at least partially distinct pathways to produce Fas-resistance that differentially depend on PKC and STAT6, respectively. Further, surface immunoglob- 'lin signaling for inducible Fas-resistance bypasses Bib, requires NF-GB, and entails new macromolecular synthesis. Terminal effectors of B cell Fas-resistance include the known anti-apoptotic gene products, Bcl-xL and FLIP, and a novel anti-apoptotic gene that encodes FAIM (Fas Apoptosis Inhibitory Molecule). faim was identified by differential display and exists in two alternatively spliced forms; maim-S is broadly expressed, but faim-L expression is tissue-specific. The FAIM sequence is highly evolu tionarily conserved, suggesting an important role for this molecule throughout phylogeny. | 9 | Nonlinear dynamics of self-oscillating polymer gels显示文摘Self-oscillating polymer gels driven by Belousov-Zhabotinsky (BZ) chemical reaction are a new class of functional gels that have a wide range of potential applications (e.g., autonomously functioning membranes, actuate artificial muscles). However, the precise control of these gels has been an issue due to limited investigations of the influences of key system parameters on the characteristics of BZ gels. To address this deficiency, we studied the self-oscillating behavior of BZ gels using the nonline-ar dynamics theory and an Oregonator-like model, with focus placed upon the influences of various system parameters. The analysis of the oscillation phase indicated that the dynamic response of BZ gels represents the classical limit cycle oscillation. We then investigated the characteristics of the limit cycle oscillation and quantified the influences of key parameters (i.e., ini-tial reactant concentration, oxidation and reduction rate of catalyst, and response coefficient) on the self-oscillating behavior of BZ gels. The results demonstrated that sustained limit cycle oscillation of BZ gels can be achieved only when these key pa-rameters meet certain requirements, and that the pattern, period and amplitude of the oscillation are significantly influenced by these parameters. The results obtained in this study could enable the controlled self-oscillation of BZ gels system. | 10 | Age- and gender-dependent obesity in individuals with 16pl1.2 deletion显示文摘Recurrent genomic imbalances at 16p 11.2 are genetic risk factors of variable penetrance for developmental delay and autism.Recently, 16pl 1.2(chr16:29.5 Mb—30.1 Mb) deletion has also been detected in individuals with early-onset severe obesity.The penetrance of 16p11.2 deletion as a genetic risk factor for obesity is unknown.We evaluated the growth and body mass characteristics of 28 individuals with 16p11.2 (chr16:29.5 Mb—30.1 Mb) deletion originally ascertained for their developmental disorders by reviewing their medical records.We found that nine individuals could be classified as obese and six as overweight.These individuals generally had early feeding and growth difficulties,and started to gain excessive weight around 5—6 years of age.Thirteen out of the 18 deletion carriers aged 5 years and older(72%) were overweight or obese,whereas only two of 10 deletion carriers(20%) younger than five were overweight or obese.Males exhibited more severe obesity than females.Thus,the obesity phenotype of 16p11.2 deletion carriers is of juvenile onset,exhibited an age- and gender-dependent penetrance. 16p11.2 deletion appears to predispose individuals to juvenile onset obesity and in this case are similar to the well-described Prader-Willi syndrome(PWS).Early detection of this deletion will provide opportunity to prevent obesity. | David T.Miller James F.Gusella Orah S.Platt on behalf of the Children's Hospital Boston Genotype Phenotype Study Group | 2011 | Journal of Genetics and Genomics2011,38,9: | 2 |
