| 1 | Phytate:impact on environment and human nutrition.A challenge for molecular breeding显示文摘Phytic acid (PA) is the primary storage compound of phosphorus in seeds accounting for up to 80% of the total seed phosphorus and contributing as much as 1.5% to the seed dry weight. The negatively charged phosphate in PA strongly binds to metallic cations of Ca, Fe, K, Mg, Mn and Zn making them insoluble and thus unavailable as nutritional factors. Phytate mainly accumulates in protein storage vacuoles as globoids, predominantly located in the aleurone layer (wheat, barley and rice) or in the embryo (maize). During germination, phytate is hydrolysed by endogenous phytase(s) and other phosphatases to release phosphate, inositol and micronutrients to support the emerging seedling. PA and its derivatives are also implicated in RNA export, DNA repair, signalling, endocytosis and cell vesicular trafficking. Our recent studies on purification of phytate globoids, their mineral composition and dephytinization by wheat phytase will be discussed. Biochemical data for purified and characterized phytases isolated from more than 23 plant species are presented, the dephosphorylation pathways of phytic acid by different classes of phytases are compared, and the application of phytase in food and feed is discussed. | Lisbeth BOHN Anne S. | 3 | Investigation of the steam-cooled blade in a steam turbine cascade显示文摘With the increasing demand for electricity,an efficiency improvement and thereby reduced CO2 emissions of the coal-fired plants are expected in order to reach the goals set in the Kyoto protocol.It can be achieved by a rise of the process parameters.Currently,live steam pressures and temperatures up to 300 bars and 923 K are planned as the next step.Closed circuit steam cooling of blades and vanes in modern steam turbines is a promising technology in order to establish elevated live steam temperatures in future steam turbine cycles.In this paper,a steam-cooled test vane in a cascade with external hot steam flow is analyzed numerically with the in-house code CHTflow.A parametric analysis aiming to improve the cooling effectiveness is carried out by varying the cooling mass flow ratio.The results from two investigated cases show that the steam cooling technique has a good application potential in the steam turbine.The internal part of the vane is cooled homogeneously in both cases.With the increased cooling mass flow rate,there is a significant improvement of cooling efficiency at the leading edge.The results show that the increased cooling mass flow ratio can enhance the cooling effectiveness at the leading edge.With respect to trailing edge,there is no observable improvement of cooling effectiveness with the increased cooling mass flow.This implies that due to the limited dimension at the trailing edge,the thermal stress cannot be decreased by increasing the cooling mass flow rate.Therefore,impingement-cooling configuration at the trailing edge might be a solution to overcome the critical thermal stress there.It is also observed that the performance of the cooling effective differs on pressure side and suction side.It implicates that the equilibrium of the cooling effectiveness on two sides are influenced by a coupled relationship between cooling mass flow ratio and hole geometry.In future work,optimizing the hole geometry and cooling steam supply conditions might be the solutions for an equivalent cooling effectiveness along whole profile. | Dieter Bohn Jing Ren Karsten Kusterer | 2007 | 航空动力学报2007,22,5: | 6 |
| 4 | Identification of human papillomavirus in esophageal squamous papillomas显示文摘AIM:To investigate the presence of human papillomavirus (HPV) in esophageal squamous papilloma (ESP) and determine p16, p53 and Ki67 expression in a Mexican cohort. METHODS: Nineteen cases diagnosed as ESP, corresponding to 18 patients were reviewed; nineteen cases of normal esophageal mucosa were used as negative controls. HPV detection was performed by amplified chromogenic in situ hybridization (ACISH) using a wide spectrum-cocktail probe and PCR. RESULTS: The average age at presentation was 46.3 years (range 28-72 years). Patients included four (22.22%) males and 14 (77.77%) females. The most frequent location was upper third (11 cases), followed by middle third (3 cases) and unknown site (5 cases). Immunohistochemistry (IHC) revealed basal and focal p53 expression in 17 cases (89%); p16 was expressed in eight cases (42.10%) and the Ki67 index ranged from 10% to 30%. HPV was detected in 14 out of 16 cases (87.5%) by ACISH: Twelve showed diffuse nuclear patterns and two showed granular patterns. HPV DNA was identified by PCR in 12 out of 14 cases (85.7%). Low-risk HPV types were detected in the most of the cases. CONCLUSION: This study provides identification of HPV infection in almost 80% of ESP using either ACISH or PCR; overall, all of these lesions show low expression of cell-cycle markers. | 6 | Investigation on heat transfer enhancement and pressure loss of double swirl chambers cooling显示文摘By merging two standard swirl chambers,an alternative cooling configuration named double swirl chambers(DSC)has been developed.In the DSC cooling configuration,the main physical phenomena of the swirl flow in swirl chamber and the advantages of swirl flow in heat transfer augmentation are maintained.Additionally,three new physical phenomena can be found in DSC cooling configuration,which result in a further improvement of the heat transfer:(1)impingement effect has been observed,(2)internal heat exchange has been enhanced between fluids in two swirls,and(3)“∞”shape swirl has been generated because of cross effect between two chambers,which improves the mixing of the fluids.Because of all these improvements,the DSC cooling configuration leads to a higher globally-averaged thermal performance parameter(Nu/Nu_(∞)/(f/f0)^(1/3))than standard swirl chamber.In particular,at the inlet region,the augmentation of the heat transfer is nearly 7.5 times larger than the fully developed non-swirl turbulent flow and the circumferentially averaged Nusselt number coefficient is 41%larger than the standard swirl chamber.Within the present work,a further investigation on the DSC cooling configuration has been focused on the influence of geometry parameters e.g.merging ratio of chambers and aspect ratio of inlet duct on the cooling perfomance.The results show a very large influence of these geometry parameters in heat transfer enhancement and pressure drop ratio.Compared with the basic configuration of DSC cooling,the improved configuration with 20%to 23%merging ratio shows the highest globally-averaged themal performance parameter.With the same cross section area in tangential inlet ducts,the DSC cooling channel with larger aspect ratio shows larger heat transfer enhancement and at the same time reduced pressure drop ratio,which results in a better globally-averaged themal performance parameter. | Gang Lin Karsten Kusterer Dieter Bohn Takao Sugimoto Ryozo Tanaka Masahide Kazari | 2013 | Propulsion and Power Research2013,2,3: | 3 |
| 7 | Conjugate calculation of a film-cooled blade for improvement of the leading edge cooling configuration显示文摘Great efforts are still put into the design process of advanced film-cooling configurations.In particular,the vanes and blades of turbine front stages have to be cooled extensively for a safe operation.The conjugate calculation technique is used for the three dimensional thermal load prediction of a fim-cooled test blade of a modern gas turbine.Thus,it becomes possible to take into account the interaction of internal flows,external flow,and heat transfer without the prescription of heat transfer ooefficients.The focus of the investigation is laid on the leading edge part of the blade.The numerical model consists of all internal flow passages and cooling hole rows at the leading edge.Furthermore,the radial gap flow is also part of the model.The comparison with thermal pyrometer measurements shows that with respect to regions with high thermal load a qualitatively and quantitatively good agreement of the conjugate results and the measurements can be found.In particular,the region in the vicinity of the mid-span section is exposed to a higher thermal load,which requires further improvement of the cooling arrangement.Altogether the achieved results demonstrate that the conjugate calculation technique is applicable for reasonable prediction of three-dimensional thermal load of complex cooling configurations for blades. | Norbert Moritz Karsten Kusterer Dieter Bohn Takao Sugimoto Ryozo Tanaka Tomoki Taniguchi | 2013 | Propulsion and Power Research2013,2,1: | 3 |
