| 1 | Effect of Ionizing Radiation on the Expression of p16, CyclinDI and CDK4 in Mouse Thymocytes and Splenocytes显示文摘Objective:To investigate the effect of ionizing radiation on the expression of p16,CyclinD1,and CDK4 in mouse thymocytes and splenocytes.Methods:Fluorescent staining and flow cytometry analysis were employed for the measurement of protein expression.Results:In time course experiments,it was found that the expression of p16 protein was significantly increased at 8,24,and 48h for thymocytes(P<0.05,P<0.01,and P<0.05,respectively)and at 24h for splenocytes (P<0.05) after whole body irradiation(WBI) with 2.0Gy X-rays.However,the expression of CDK4 protein was significantly decreased from 8h to 24h for thymocytes(P<0.05-P<0.01) and from 8h to 72h for splenocytes(P<0.05-P<0.01).In dose effect experiments,it was found that the expression of P16 protein in thymocytes and splenocytes was significantly increased at 24h after WBI with 1.0,2.0 ,and 4.0GY(P<0.05-P<0.01),whereas the expression of CDK4 protein was significantly decreased with 2.0Gy for thymocytes(P<0.05)and 0.5-6.0 Gy for splenocytes(P<0.05-P<0.01),Results also showed that the expression of CyclinD1 protein decreased markedly in both thymocytes and splenocytes after exposure.Conclusion:the results indicate that the expression of p16 protein in thymocytes and splenocytes can be induced by ionizing radiation,and the p16-CyclinD1/CDK4 pathway may play an important role for G1 arrest of thymocytes induced by X-rays. | GUI-ZHI JU, XIAO-MEI WANG, SHI-Bo FU, AND SHU-ZHENG LIUDepartment of Radiation Biology, Medical Center in Memorial of Norman Bethune, Jilin University, Changchun 130021, China | 2003 | Biomedical and Environmental Sciences2003,16,1: | 7 |

| 6 | Effect of Low Dose Radiation on Intracellular Calcium and Protein Kinase C in Lymphocytes显示文摘It is first reported in the present paper that whole-body irradiation (WBI) with low dose X-rays could increase intracellular calcium ions ([Ca2+]i) and stimulate protein kinase C (PKC) activity of mouse lymphocytes. Following WBI of male Kunming micc With 75 mGy X-rays at a dose rate of 12.5 mGy/min the mobilization of [Ca2+]i with Con A in CD4+ and CD8+ Cells in the thymus and spleen was potentiated and the amplitude of [Ca2+], mobilization in thymocytes in response to anti-CD3 monoclonal antibody increased with time from 4 to 24 h following low dose radiation. The PKC activity in the homogenate of spleen was markedly stimulated 12 h after WBl with 75 mGy, reaching its peak value at 24-48 h and coming down to lower than normal on day 7. However, the PKC activity in the separated T lymphocytes reached its peak value at 12 h and that in the B lymphocytes reached its peak value on day 4, both coming down to below control on day 7. | 8 | Effect of methylmercury on LPO levels, SH groups and activities of GSH-Px, SOD, XOD in liver of rats显示文摘In recent years, lipid peroxidation has been considered as the initial step for foreign toxic substances to damage cells. In this paper, the effects of methylmercury (MeHg) on LPO (lipid peroxide) levels, SH (sulfhydryl) groups and activities of GSH\|Px (glutathione peroxidase), SOD (superoxide dismutase), XOD (xanthine oxidase) in liver of rats treated with doses of MeHg at different intervals were studied by TBA, Ellman Reagent, NBT colorimetric methods and chemilluminescence. Meanwhile, the contents of FR (free radical)and MeHg in the liver of female rats were measured by ESR and GC. Results showed that the LPO levels in the experimental group increased significantly over those of the control group ( p<0.05— p<0.001) , reaching maximum point on the first poisoning day. Both male and female rats showed marked positive dose\|effect relations (Male: Y=261.4±49.4X, r=0.94, p<0.02; Female : Y=255.0±73.0X, r=0.99, p<0.001 ). N-SH (nonprotein sulfhydryl), P\|SH (protein sulfhydryl), T-SH (total sulfhydryl) contents and GSH\|Px activities decreased significantly (p<0.05-p<0.001). For GSH\|Px, both groups reached minimum value on the first poisoning day. In female rats, activities of SOD and XOD increased markedly ( p<0.02-p<0.01 ) although the contents of FR showed no significant changes. | 9 | Taurine Inhibits Myocardial Fibrosis via PKC-ERK1/2 Signaling Pathways显示文摘Previous studies have demonstrated the important role of taurine in inhibiting proliferation of myofibrob lasts(myoFb) and myocardial fibrosis. However, the underlying mechanisms are unclear. The present study was de signed to shed light on this issue through exploring the signal pathways via in vitro experiments. Angiotension II (AngII) treatment significantly increased myoFb proliferation and the levels of collagens I and III(P<0.05), whereas taurine, PKCα(PKC: protein kinase C) specific inhibitor L-threo-dihydro-sphingosine(D4681), ERK1/2 inhibitor (PD98095) abrogated myoFb proliferation and collagen levels(P<0.05, P<0.01, respectively), and increased the G0/G1 phase rate and decreased S phase rate. Immunocytochemistry, confocal fluorescence staining and image analy sis showed that taurine could inhibit the translocation and expression of p-PKCαin membrane, and then inhibit nuc lear translocation and expression of p-ERK1/2. These results have statistically significant differences compared with those of AngII group(P<0.01). Western blot results also show that taurine could inhibit the protein expression of p-PKCα and p-ERK1/2. We used p-PKCα specific inhibitor D4681 in order to elucidate the relationship between p-PKCα and p-ERK1/2 in signal transduction pathways. | 12 | 表皮生长因子受体、血管内皮生长因子受体和BAD在非小细胞肺癌中的表达显示文摘背景与目的:表皮生长因子受体(epidermal growth factor receptor,EGFR)和血管内皮生长因子受体(vascular endothelial growth factor receptor,VEGFR)均属酪氨酸激酶受体(receptor tyrosine kinase,RTK),可调控细胞的增殖、分化与生存。BAD是Bcl-2家族中的促凋亡信号成分,在调控细胞凋亡特别是肿瘤细胞凋亡过程中发挥重要作用。但目前人们对上述这些重要蛋白在非小细胞肺癌(non-small-cell lung cancer,NSCLC)中的表达与肿瘤病理学的关系所知甚少。本研究探讨ECFR、VEGFR、BAD和磷酸化BAD在NSCLC中的表达情况以及与肿瘤病理的关系。方法:使用组织微阵列(tissue microarray,TMA)切片的免疫组织化学法,NSCLC患者51例(26例腺癌,16例鳞癌,8例大细胞癌,1例大细胞神经内分泌癌)。结果:51例患者中EGFR和VEGFR分别在10例(20%)和14例(27%)中出现过度表达。大细胞癌中未见VEGFR表达(0/8例),而鳞癌和腺癌患者中VEGFR表达分别为44%(7/16)和27%(7/26)。EGFR和VEGFR的表达与性别,肿瘤细胞分化及肿瘤浸润程度(包括胸膜浸润,血管浸润,淋巴结转移,肺内播散,脑转移情况)无关。51例患者中22例(43%)出现BAD蛋白表达缺失,且NSCLC的不同病理类型间差异有显著性。BAD蛋白表达缺失在16例鳞癌患者中10例(63%),8例大细胞癌患者中5例(63%),26例鳞癌患者中有7例(27%)(P=0.04)。51例患者中25例(49%)出现磷酸化BAD蛋白过度表达[其中26例腺癌患者中有13例(50%),16例鳞癌患者中有8例(50%),8例大细胞癌患者中有4例(50%)]。BAD蛋白的表达缺失与磷酸化BAD蛋白的过度表达经统计检验与上述肿瘤浸润程度无相关性。结论:肺鳞癌出现VEGFR表达增高的可能较大,而大细胞癌出现VEGFR表达增高的可能最小。在鳞癌和大细胞癌中可见BAD蛋白表达的显著缺失。NSCLC患者EGFR,VEGFR,磷酸化BAD蛋白的过度表达以及BAD蛋白表达的缺失与病理浸润程度无关。但这些受体酪氨酸激酶表达以及与NSCLC凋亡直接相关的媒介因子可能成为未来多靶向治疗中的候选靶标。 | Drew Bethune Neale Ridgway 罗晓阳 | 2006 | 中国癌症杂志2006,16,8: | 2 |
| 13 | Harmful Effects of MSG on Function of Hypothalamus-Pituitary-Target Gland System显示文摘It has been demonstrated that neonatal administration of monosodium glutamate (MSG) results in a clearly defined lesion of the arcuate nucleus (AN) of the hypothalamus.The present study shows that fat was accumulated in the abdomen of male rats treated with MSG; weights of the body, pituitary and testis were lower, 6-EP content in hypothalamus decreased while L-EnK content increased, serum LH, FSH, TSH, GH and TS levels all decreased in varying degrees while serum PRL level significantly increased. The cAMP content lowered in pituitary, but nor in testes; clear histological changes occurred in testicular tissue; Se-GSH-Px activity in both testis and adrenal gland lowered while LPO level significantly increased.
