1Effects of maternal diabetes on trophoblast cells显示文摘Diabetes mellitus(DM)is a health condition characterized by hyperglycemia over a prolonged period.There are three main types of DM:DM type 1(DM1),DM2 and gestational DM(GDM).Maternal diabetes,which includes the occurrence of DM1 and DM2 during pregnancy or GDM,increases the occurrence of gesttional complications and adverse fetal outcomes.The hyperglycemic intrauterine environment affects not only the fetus but also the placental development and function in humans and experimental rodents.The underlying mechanisms are still unclear,but some evidence indicates alterations in trophoblast proliferation,apoptosis and cell cycle control in diabetes.A proper coordination of trophoblast proliferation,differentiation and invasion is required for placental development.Initially,increased expression of proliferative markers in junctional and labyrinth zones of rat placentas and villous cytotrophoblast,syncytiotrophoblast,stromal cells and fetal endothelial cells in human placentas is reported among diabetics.Moreover,reduced apoptotic index and expression of some apoptotic genes are described in placentas of GDM women.In addition,cell cycle regulators including cyclins and cyclin-dependent kinase inhibitors seem to be affected by the hyperglycemic environment.More studies are necessary to check the balance between proliferation,apoptosis and differentiation in trophoblast cells during maternal diabetes.Marlúcia Bastos Aires Anne Carolline Veríssimo dos Santos 2015World Journal of Diabetes2015,6,2:9
2Short-term complementary and alternative medicine on quality of life in women with fibromyalgia显示文摘BACKGROUND:Fibromyalgia(FMS)is a syndrome characterized by chronic widespread musculoskeletal pain,whose etiology is not completely understood.Different therapeutic approaches have been used with inconsistent results.This observation does not invalidate the continued search for alternative treatments aimed at improving quality of life(Qo L)in FMS.OBJECTIVE:This study compared three classical traditional Chinese medicine(TCM)therapies:acupuncture(AC),electroacupuncture(EAC)and moxibustion(MX)in the management of pain and promotion of Qo L in FMS patients.DESIGN,SETTING,PARTICIPANTS AND INTERVENTIONS:A preliminary,group-assigned,comparative study enrolled 30 women,mean age(46.90±9.24)years(range 20–60 years),who met the 1990 American College of Rheumatology criteria for FMS diagnosis and a pain-pressure threshold(PPT)< 4 kg/cm^2.The study was conducted in a teaching tertiary-care medical institution from May 2010 through April 2012.AC,EAC and MX were delivered for 30 min,once a week,for 8 weeks,bilaterally at Neiguan(PC6),Hegu(LI4),Yanglingquan(GB34),Sanyinjiao(SP6)and Taichong(LR3)acupoints.MAIN OUTCOME MEASURES:Each week,immediately before treatment and after treatment,subjects were tested for PPTs,Wong-Baker Faces Pain Scale(WBFPS;for pain intensity)and Medical Outcomes Study 36-item Short Form Health Survey(SF-36:for Qo L).RESULTS:There was no signifi cant improvement in pain or reduction of tender points in any of the groups studied,at the end of the 8th session.Signifi cant improvement of Qo L was perceived in vitality(after AC treatment)and in mental health(after EAC and MX treatments).CONCLUSION:TCM therapies(AC,EAC and MX)promoted an improvement in the Qo L in two areas(vitality and mental health)in FMS women.Further large-scale clinical trials are required to confi rm this effect.Paulo Araujo Dias Andre Brito Bastos Guimaraes Andrea de Oliveira Albuquerque Karoline Lucas de Oliveira Maria Luzete Costa Cavalcante Sergio Botelho Guimaraes 2016Journal of Integrative Medicine2016,14,1:7

4Cumulative positive fluid balance is a risk factor for acute kidney injury and requirement for renal replacement therapy after liver transplantation显示文摘AIM To analyze whether fluid overload is an independent risk factor of adverse outcomes after liver transplantation(LT).METHODS One hundred and twenty-one patients submitted to LT were retrospectively evaluated.Data regarding perioperative and postoperative variables previously associated with adverse outcomes after LT were reviewed.Cumulative fluid balance(FB) in the first 12 h and 4 d after surgery were compared with major adverse outcomes after LT.RESULTS Most of the patients were submitted to a liberal approach of fluid administration with a mean cumulative FBover 5 L and 10 L,respectively,in the first 12 h and 4 d after LT.Cumulative FB in 4 d was independently associated with occurrence of both AKI and requirement for renal replacement therapy(RRT)(OR = 2.3;95%CI:1.37-3.86,P = 0.02 and OR = 2.89;95%CI:1.52-5.49,P = 0.001 respectively).Other variables on multivariate analysis associated with AKI and RRT were,respectively,male sex and Acute Physiology and Chronic Health Disease Classification System(APACHE II) levels and sepsis or septic shock.Mortality was shown to be independently related to AST and APACHE II levels(OR = 2.35;95%CI:1.1-5.05,P = 0.02 and 2.63;95%CI:1.0-6.87,P = 0.04 respectively),probably reflecting the degree of graft dysfunction and severity of early postoperative course of LT.No effect of FB on mortality after LT was disclosed.CONCLUSION Cumulative positive FB over 4 d after LT is independently associated with the development of AKI and the requirement of RRT.Survival was not independently related to FB,but to surrogate markers of graft dysfunction and severity of postoperative course of LT.Liana Codes Ygor Gomes de Souza Ricardo Azevedo Cruz D'Oliveira Jorge Luiz Andrade Bastos Paulo Lisboa Bittencourt 2018World Journal of Transplantation2018,8,2:4
5Coating independent cytotoxicity of citrate-and PEG-coated silver nanoparticles on a human hepatoma cell line显示文摘The antibacterial potential of silver nanoparticles(AgNPs) resulted in their increasing incorporation into consumer,industrial and biomedical products.Therefore,human and environmental exposure to AgNPs(either as an engineered product or a contaminant)supports the emergent research on the features conferring them different toxicity profiles.In this study,30 ran AgNPs coated with citrate or poly(ethylene glycol)(PEG) were used to assess the influence of coating on the effects produced on a human hepatoma cell line(HepG2),namely in terms of viability,apoptosis,apoptotic related genes,cell cycle and cyclins gene expression.Both types of coated AgNPs decreased cell proliferation and viability with a similar toxicity profile.At the concentrations used(11 and 5 μg/mL corresponding to IC50 and-IC10 levels,respectively) the amount of cells undergoing apoptosis was not significant and the apoptotic related genes BCL2(anti-apoptotic gene)and BAX(pro-apoptotic gene) were both downregulated.Moreover,both AgNPs affected HepG2 cell cycle progression at the higher concentration(11 μg/mL) by increasing the percentage of cells in S(synthesis phase) and G2(Gap 2 phase) phases.Considering the cell-cycle related genes,the expression of cyclin B1 and cyclin E1 genes were decreased.Thus,this work has shown that citrate- and PEG-coated AgNPs impact on HepG2 apoptotic gene expression,cell cycle dynamics and cyclin regulation in a similar way.More research is needed to determine the properties that confer AgNPs at lower toxicity,since their use has proved helpful in several industrial and biomedical contexts.Veronica Bastos Jose M.P.Ferreira-de-Oliveira Joana Carrola Ana L.Daniel-da-Silva Iola F.Duarte Conceicao Santos Helena Oliveira 2017Journal of Environmental Sciences2017,29,1:4

7Association of Helicobacter pylori infection and giardiasis: Results from a study of surrogate markers for fecal exposure among children显示文摘AIM: To investigate whether Helicobacter pylori (H pykori)infection is associated with hepatitis A virus (HAV) infection, presence of enteroparasites, and other surrogates of fecal exposure.METHODS: We conducted a cross-sectional study in 121 children consecutively admitted at a pediatric hospital inSalvador, Brazil.H pylori and HAV infection were identified by the presence of serum antibodies. Stool specimens were examined for the presence of ova and parasites. A structured questionnaire inquiring about sanitary conditions and life style was applied to each subject. RESULTS: Fifty-one of the 121 children (42.1%) werefound to be seropositive for H pylori, and 45 (37.2%) for HAV. The seroprevalence ofHpyloriand HAV both increased significantly with age. Cross-tabulation of data showed that 26 (21.5%) were seropositive and 51 (42.1%) were negative for bothHpyloriand HAV antibodies (χ2 = 7.18,OR = 2.8, CI 1.30-5.97). The age adjusted OR for an HAV-infected child being H pylori positive was 2.3 (CI1.02-5.03). The agreement between H pylori and HAVseropositivity was fair (κ = 0.24). After controlling for possible confounding, the variables remaining independently associated with seropositivity to H pyloriwere age,presence of Giardia lamblia in feces (OR = 3.2, 95%CI, 1.1-9.5) and poor garbage disposal quality (OR = 2.4,95%CI, 1.1-5.1).CONCLUSION: Our data suggest that H pylori infection is associated with surrogate markers of fecal exposure. Thus, we conclude that the fecal-oral route is relevant in the transmission of HP among children in an urban setting of a developing country. The association observed between G. lamblia and H pylori infection may have several explanations. 9Hepatitis C virus: Promising discoveries and new treatments显示文摘Despite advances in therapy, hepatitis C virus(HCV) infection remains an important global health issue. It is estimated that a significant part of the world population is chronically infected with the virus, and many of those affected may develop cirrhosis or liver cancer. The virus shows considerable variability, a characteristic that directly interferes with disease treatment. The response to treatment varies according to HCV genotype and subtype. The continuous generation of variants(quasispecies) allows the virus to escape control by antivirals. Historically, the combination of ribavirin and interferon therapy has represented the only treatment option for the disease. Currently, several new treatment options are emerging and are available to a large part of the affected population. In addition, the search for new substances with antiviral activity against HCV continues, promising future improvements in treatment.
