| 1 | Effect of biofilm formation by clinical isolates of Helicobacter pylori on the efflux-mediated resistance to commonly used antibiotics显示文摘AIM To evaluate the role of biofilm formation on the resistance of Helicobacter pylori(H. pylori) to commonly prescribed antibiotics, the expression rates of resistance genes in biofilm-forming and planktonic cells were compared.METHODS A collection of 33 H. pylori isolates from children and adult patients with chronic infection were taken for the present study. The isolates were screened for biofilm formation ability, as well as for polymerase chain reaction(PCR) reaction with HP1165 and hp1165 efflux pump genes. Susceptibilities of the selected strains to antibiotic and differences between susceptibilities of planktonic and biofilm-forming cell populations were determined. Quantitative real-time PCR(qP CR) analysis was performed using 16 S r RNA gene as a H. pylori-specific primer, and two efflux pumps-specific primers, hp1165 and hefA.RESULTS The strains were resistant to amoxicillin, metronidazole, and erythromycin, except for one strain, but they were all susceptible to tetracycline. Minimum bactericidalconcentrations of antibiotics in the biofilm-forming cells were significantly higher than those of planktonic cells. qP CR demonstrated that the expression of efflux pump genes was significantly higher in the biofilm-forming cells as compared to the planktonic ones.CONCLUSION The present work demonstrated an association between H. pylori biofilm formation and decreased susceptibility to all the antibiotics tested. | 2 | Immunogenicity and protective efficacy of recombinant M2e.Hsp70c(Hsp70_(359–610)) fusion protein against influenza virus infection in mice显示文摘New strategies in vaccine development are urgently needed to combat emerging influenza viruses and to reduce the risk of pandemic disease surfacing. Being conserved, the M2 e protein, is a potential candidate for universal vaccine development against influenza A viruses. Mycobacterium tuberculosis Hsp70(mHsp70) is known to cultivate the function of immunogenic antigen-presenting cells, stimulate a strong cytotoxic T lymphocyte(CTL) response, and stop the induction of tolerance. Thus, in this study, a recombinant protein from the extracellular domain of influenza A virus matrix protein 2(M2e), was fused to the C-terminus of Mycobacterium tuberculosis Hsp70(Hsp70c), to generate a vaccine candidate. Humoral immune responses, IFN-γ-producing lymphocyte, and strong CTL activity were all induced to confirm the immunogenicity of M2 e.Hsp70c(Hsp70359–610). And challenge tests showed protection against H1N1 and H9N2 strains in vaccinated groups. | 3 | The Long-Sought-After Salicylic Acid Receptors显示文摘The plant hormone salicylic acid (SA) plays a prominent role in modulating plant immune responses against diverse pathogens.SA also influences other physiological processes in plants,such as senescence-associated gene expression,basal thermogenesis,and seed germination (Vlot et al.,2009).Because of the critical role of SA in regulating plant immunity,growth,and development,there has been immense research about SA,which has resulted in the discovery of numerous plant genes involved in SA biosynthesis or signal transduction.One of the most notable findings was the identification of NPR1 (non-expressor of pathogenesis protein 1) (Cao et al.,1994;Delaney et al.,1995),a gene that encodes a master regulatory protein of SA-dependent defense responses and is a transcriptional co-activator of the TGA clade of bZIP transcription factors (transcription factors that contain basic region/leucine zipper motif).NPR1 exists in at least two forms in the cell.When the SA level is low in the cell (e.g.in the absence of pathogen infection),NPR1 is localized in the cytoplasm as an oligomer through intermolecular disulphide bonds.However,when the SA level is high (e.g.after pathogen infection),redox changes in the cytosol trigger the reduction of disulphide bonds,and monomeric NPR1 enters the nucleus and functions as a transcriptional co-activator at the target gene promoter (Mou et al.,2003).The npr1 mutant could not initiate the SA-associated global transcriptional response program and is defective inall major SA-dependent defense responses,suggesting a central role in SA signal transduction (Cao et al.,1994;Delaney et al.,1995).Despite these impressive advances,however,one of the most fundamental questions-the identity of the SA receptor-has remained unanswered. | Elham Attaran ShengYang He | 2012 | Molecular Plant2012,5,5: | 2 |
