1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 2Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. 3The Bedload Movement in the Changjiang Estuary显示文摘- Sandwaves in the Changjiang estuary were measured with a shallow sediment profiler and an echosounder from 1978 to 1988. The data, together with grain size and bedform of sediment indicates that the bedload movement by rolling and saltation is of great significance to sediment transport and is the principal factor responsible for sandwave and sandbody development in the estuary. The sandwaves were found well-developed, which is related to the tidal range and the velocity of ebb current. However, the further growth is restricted by strong flood current prevailing in the estuary. Because of the significant bedload, the sandbodies shift obviously and frequently, and sometimes the exchange of position occurs between the sandbodies and tidal channels. 8VELOCITY PROFILE OF SEDIMENT-LADEN OPEN CHANNEL FLOW显示文摘A detailed literature survey of the existing models for vertical velocity profiles of open channel flow for both clear water and sediment-laden water is performed. It is found that flow resistance and suspended sediment particles affect velocity profiles significantly. The Karman constant K of the logarithmic law is equal to 0.41 near the channel bed but far from the bed the value will change due to flow resistance and sediment suspension. The exponent of the power law velocity profile is also affected by those two factors. A general logarithmic law and a general power law for the description of velocity profiles arc proposed herein and verified by the experiment data presented by several investigators. 9Evalution of Application of p-y Curve Method in Offshore Engineering Foundation显示文摘- Based on the Mohr-Coulomb failure principle and Rankine’s theory, the laterally loaded pile ultimate resistance formulas of sand and soft clay proposed by Reese and Matlock respectively are discussed in this paper. The authors put forward the modified ultimate resistance formulas on the basis of which the ultimate resistance formula is developed for horizontally loaded pile in multi-layer soil in consideration of the effect of the overburden soil pressure on the calculation of soil layer. It also examines the trends of industrial cluster, the formation of the agglomerative economy and their effects on private economy development. 12FIELD INVESTIGATION INTO DEBRIS FLOW MECHANISM显示文摘Debris flow carries huge amount of sediment, from clay finer than 10-3 mm to huge stonesof several meters in diameter. Clay, sand, gravels and boulders move in different mechanisms.Field investigations performed by the authors reveal that there are two kinds of debris flowswith different mechanismst viscous debris now and two-phase debris flow. viscous debrisflow consists of more fine material and exhibits intermittent flow, paving way process, dragreduction and high superelevation. Drag reduction is caused by the paving way process andaeration of the flowing mixture. 13ANALYSIS OF DYNAMIC STABILITY OF SUBMERGED STRUCTURE显示文摘The submerged structure is basically a large three-dimensional structure of few statically redundant members. The structure is subjected to vertical dead and live loads in addition to the wave forces. An analysis of dynamic stability of the submerged structure without damping has been made by J. Thomas and Abbas (1980). In this paper the analyses of dynamic stability of the sumberged structure with damping are conducted. The case structure with damping is more complicated ’than the case without it. According to the principle of perturbation, a new model for dynamic stability calculation in consideration of damping effect is developed. In this paper, the formulas are deduced, the computational program is compiled, the practical examples are analysed, and this problem is solved very satisfactorily. The computational results show that the shape and value of the regions of dynamic instability can be changed significantly by damping. 15BED FORMS AND RESISTANCE PROPERTIES OF LIGHT-WEIGHT BED MATERIAL显示文摘Based on experimental results from a large tilting flume, the characteristics and de- veloping processes of bed forms of plastic particles during scouring and deposition are systematically studied. The relationships of the occurence, development and disappearence of sand waves with the flow conditions are presented. Various statistical characteristics of sand waves with different sizes and the corresponding resistance properties of different bed forms of plastic particles are obtained. These results have filled in the gaps in bed forms and resistance properties of light sediment particles. 16Experimental Wave Power Plant at the Pearl River Estuary显示文摘The first Chinese experimental wave power plant, i. e., the shoremountcd experimental wave power plant at the Pearl River Estuary was successful in trial power generation on February 15, 1990. The plant is on the south shore of the Dawanshan Island in the Pearl River Estuary, facing the vast waving South China Sea. With a designed wave condition of H1/10= 1.5 m and T= 6.5 s, the planned installed capacity is 8 kW comprising the first unit of 3 kW brushless clawpole generator which delivers 110 V DC current and the second unit of 5 kW brushless single phase synchro AC generator which delivers 220 V AC current.
