| 1 | Dense coding scheme using superpositions of Bell-states and its NMR implementation显示文摘Dense coding using superpositions of Bell-states is proposed. The generalized Grover's algorithm is used to prepare the initial entangled states, and the reverse process of the quantum algorithm is used to determine the entangled state in the decoding measurement. Compared with the previous schemes, the superpositions of two Bell-states are exploited. Our scheme is demonstrated using a nuclear magnetic resonance (NMR)quantum computer. The corresponding manipulations are obtained. Experimental results show a good agreement between theory and experiment. | 2 | Primary structure and configuration of tea polysaccharide显示文摘The monosaccharide composition of a tea polysaccharide (TGC) was determined by GC-MS method. Furthermore, the primary structure of tea polysaccharide and its configura-tion in the aqueous solution were investigated utilizing a combination of classical chemical methods and modern instrumental techniques including GC-MS, Proton NMR, UV and CD. The results indicate that TGC consists of 6 monosaccharides: Rha, Ara, Xyl, Glu, Man and Gal. The configuration of TGC in water solution is proposed to be an ordered helix. The possible primary structure of TGC was outlined as below: the basic structure of the main chain consists of Rha, Glu and Gal units. All three monosaccharides can potentially be connected to branch chains consisting of mainly Ara, and the linkages could be in b1→2, b 1→3, b 2→3 forms. | 3 | Excretion of Four Catechins in Tea Polyphenols in Rats显示文摘Objective To investigate excretion profiles of the four major anti-oxidant active catechins, (-) epigallo-catechin-3-gallate (EGCG), (-) epicatechin-3-gallate (ECG), (-) epigallocatechin (EGC), and epicatechin (EC) in tea polyphenols (TP) in rats in order to provide experimental data for clinical uses and development of TP as a novel drug. Methods The above four catechins in urine, bile, and feces were simultaneously determined by high performance liquid chromatography coupled with ultraviolet absorption detector (HPLC-UV) assay with a binary gradient elution. The samples were extracted by ethyl acetate prior to HPLC. The quantification was carried out by peak area internal standard method. Following iv dosing TP 100 mg/kg to rats, the samples were collected at different time intervals up to 8 h (urine and bile) and 24 h (feces). Results The urinary Ae, 0-8 h (cumulative excretion amount over 8 h) of EGCG, ECG, EGC, and EC were, on the average, 150.83, 30.75, 116.69, and 254.56 μg, corresponding to fe, 0-8 h (cumulative excretion fraction of dose over 8 h) of 1.45%, 0.84%, 7.88%, and 10.73%, respectively; the biliary Ae, 0-8 h were 12.61, 42.64, 6.61, and 1.24 μg, corresponding to the fe, 0-8 h of 0.12%, 1.16%, 0.45%, and 0.053%,respectively. For fecal excretion, only EGCG and EGC were detected with Ae, 0-24 h of 7.38 μg (fe, 0-24 h of 0.07%) and 157 μg (fe, 0-24 h of 9.99 %), respectively. The fe, total (the total fe of 3 excretory routes) were 18.32%, 10.78%, 2.00%, and 1.64% for EGC, EC, ECG, and EGCG, respectively. Conclusion EGCG and EC are mainly excreted in urine, ECG in bile, and EGC in feces by reference to their Ae and fe. The excretion of the four catechins based on fe, total is ranked in order of EGC > EC > ECG > EGCG. | 4 | LC-MS/MS for Simultaneous Determination of Four Major Active Catechins of Tea Polyphenols in Rat Plasma and Its Application to Pharmacokinetics显示文摘Objective To develop a liquid chromatography technique coupled with tandem mass spectrometry (LC-MS/MS) for simultaneous determination of four active catechins EGCG, ECG, EGC, and EC of tea polyphenols (TP) in rat plasma in order to further study its multi-component pharmacokinetics. Methods Following a single step liquid-liquid extraction of plasma samples with ethyl acetate, the four catechins were separated on a Hypersil ODS C18 column using an isocratic mobile phase composed of methanol-water (30︰70). The detection using a mass spectrometer was performed under negative ESI in the MRM mode. The analytes were identified by reference to both MRM and tR values and quantified using peak area internal standard method. Results The method was shown to be specific without interference from matrix, metabolites, and impurities present in TP raw material and to be sensitive with LOD and LOQ of 1.5 and 10 ng/mL (EGCG) as well as 0.75 and 5 ng/mL (ECG, EGC, and EC). A good linearity was obtained over a wide range of 10-10 000 ng/mL for EGCG and 5-5000 ng/mL for other three catechins (r > 0.996). The method was validated to be reproducible and reliable, as evidenced by intra-batch and inter-batch precision of less than 10% and 11%, accuracy of 97.13%-106.05% and 99.22%-103.14%, respectively. The recovery of extraction ranged from 72.74% to 89.13%, matrix effect from 88.76% to 105.97% for four catechins. The method was successfully used to study the pharmacokinetics of TP iv administered to rats at a dose of 100 mg/kg. | 5 | Method Development of Enantiomer Separations by Affinity Capillary Electrophoresis,Cyclodextrin Electrokinetic Chromatography and Capillary Electrophoresis-Mass Spectrometry显示文摘Capillary electrophoresis (CE) has become a powerful tool for enantiomer separations during the last decade. Since 1993, the author has investigated enantiomer separations by affinity capillary electrophoresis (affinity CE) with some proteins and by cyclodextrin electrokinetic chromatography (CDEKC) with some charged cyclodextrins (CDs). Many successful enantiomer separations are demonstrated from our study in this review article. In the enantiomer separations by affinity CE, the deterioration of detection sensitivity was observed under high concentration of the protein in running solutions. The partial filling technique was practically useful to solve the serious problem. It allowed operation at high protein concentrations, such as 500 μmol/L, without the detection problem. Charged CDs had several advantages for the enantiomer separations over neutral ones. Strong electrostatic interactions between a charged CD and oppositely charged analytes should be effective for the formation of the complex. A large difference in electrophoretic mobility between the free analyte and the inclusion complex should also enhance the enantiomeric resolution. In CE mass spectrometry (CE MS), the partial filling technique was applied to avoid the introduction of nonvolatile chiral selectors into the CE MS interface. The functional nanomaterials offer excellent platforms for fabrication of sensitive biosensing devices, including optical and electronic biosensors. A lot of works have fo- cused on the biofunctionalization of different nanomaterials, such as metal nanoparticles, semiconductor nanoparticles and carbon nanostructures, by physical adsorption, electrostatic binding, specific recognition or covalent coupling. This strategy utilized the magnetic particles (MNPs) for the immobilization of linker DNA and CdS NPs and the subsequent target DNA hybridization. Using the unique characteristic of nicking endonuclease for cutting one specific strand of double strand DNA (ds DNA), the linker DNA could be transected and the target DNA could be liberated for re-hybridization and hence the amount of released CdS NPs was enhanced. Due to the advantage of the MNPs and signal amplification from the target recycling, the analyte DNA could be detected by the square-wave stripping voltammetry (SWV) in a wide linear range from 0.4 fM to 100 fM with the detection limit down to 0.08 fM. | 14 | A strategy with label-free quantification of the targeted peptides for quantitative peptidome analysis of human serum显示文摘Peptidomics draws more and more attention in discovering useful biomarkers for early diagnosis of disease. However, there is lack of efficient quantification strategy in peptidome analysis. In this study, a strategy with label-free quantification of the targeted endogenous peptides based on peak intensity using μUPLC-Q-TOF-MS/MS was developed for quantitative peptidome analysis of human serum. Different amounts of standard BSA tryptic digesting peptides were added into the same serum extracts for evaluation of the developed strategy, and it was observed that the average relative error of the targeted peptides was 6.42%, which was superior to the result obtained directly by commercially available software PLGS. It was also demonstrated that this quantification strategy could obviously increase the detection sensitivity of the peptide by DDA analysis. Then, this strategy was applied to comparatively analyze the peptides extracted from the serum of HCC or breast cancer patients and healthy individuals, respectively. Peptides with charge states up to 5 and molecular weight over 4000 can be reliably identified and quantified. This quantitative analysis method based on μUPLC-Q-TOF-MS/MS exhibited superior sensitivity than that by MALDI-TOF-MS commonly used in peptidome analysis. | 15 | Immunomagnetic assay combined with CdSe/ZnS amplification of chemiluminescence for the detection of abscisic acid显示文摘Abscisic acid (ABA) is an important plant hormone. It plays a key role in regulating plant responses to abiotic stress and in controlling seed germination, growth, and stomatal aperture. A rapid, sensitive analytical method for the ABA detection is urgently required for further investigation of ABA signaling. In this work, an immunomagnetic assay combined with CdSe/ZnS amplification of chemiluminescence has been developed for the detection of ABA. The result could be read out in 30 min at least, with the simplified procedure of immunomagnetic assay. Under the optimized condition, a linear range from 1 pM to 10 nM was obtained. An unexpected result induced by the dose hook effect was discussed. | 16 | Expression and function analysis of the metallo-thionein-like (MT-like) gene from Festuca rubra in Chlamydomonas reinhardtii chloroplast显示文摘The cDNA of the metallothionein-like (MT-like) gene from Festuca rubra cv. Merlin was optimized with bias codon of Chlamydomonous reinhardtii chloroplast genome. The optimized MT-like gene was de-livered into C. reinhardtii chloroplast and the transgenic strains expressing MT-like gene was obtained. PCR-Southern blot and RT-PCR-Southern blot analysis demonstrated that the MT-like gene was inte-grated into chloroplast genome of C. reinhardtii and expressed at the transcriptional level. The cad-mium binding capacity of the transgenic C. reinhardtii was determined by hydride generation-atomic fluorescence spectrometry (HG-AFS) and the binding properties were analyzed. | 18 | Electrochemiluminescence from tris(2,2′-bipyridyl) ruthenium (Ⅱ) in the presence of aminocarboxylic acid co-reactants显示文摘Due to the highly sensitive electrochemiluminescence (ECL), tris(2,2′-bipyridyl) ruthenium(II) (Ru(bpy)32+) is often used in the field of bioarrays with the help of co-reactants. However, the generally used co-reactant, tripropylamine (TPA), is toxic, corrosive and volatile. Therefore, the search for safe, sensitive and economical co-reactants is critical. Herein, three aminocarboxylic acids, ethylenediamine-tetraacetic acid (EDTA), nitrilotriacetic acid (NTA), and 2-hydroxyethylethylene diaminetriacetic acid (HEDTA), have been investigated as potential co-reactants for promoting Ru(bpy)32+ ECL behaviour. A possible ECL mechanism is also presented. The experimental results suggested that the co-reactants have a different ECL behaviour compared to TPA, such as different pH- and surfactant-responses. The detection limits of Ru(bpy)32+ using NTA, EDTA and HEDTA as co-reactants are 1, 60 and 680 fmol·L-1, respectively. The results indicate that NTA has a much higher efficiency than TPA to excite Ru(bpy)3 2+ ECL under their own optimal conditions. NTA could be widely used in many fields because it is less toxic, corrosive and volatile than TPA. Moreover, using Ru(bpy)3 2+ ECL, a sensitive method for the detection of aminocarboxylic acids is also developed. DMS concentration in surface sea water and in the air were measured during a research cruise from Hawaii to Tahiti. The diurnal variation of air temperature over the sea surface differed from the diurnal cycle of sea surface temperature because of the high heat capacity of sea water. The diurnal cycle of average DMS concentration in the air was studied in relation to the atmospheric stability parameter and surface heat flux. All these parameters had minima at noon and maxima in the early morning. The correlation coefficient of the air DMS concentration with wind speed (at 15 m high) was 0. 64. The observed concentrations of DMS in the equatorial marine surface layer and their diurnal variability agree well with model simulations. | 20 | Fabrication of the DNA/poly(3-methylthiophene) composite film modified electrode and its application for the study on the voltammetric behavior and determination of 8-hydroxy-2'-deoxyguanosine显示文摘A composite film of DNA/poly(3-methylthiophene)(P3MT) modified glassy carbon electrode(GCE) has been fabricated by electro-deposition method.P3MT film was first electropolymerized at the GCE and the DNA layer was then immobilized on the P3MT layer by electrochemical method.The voltammetric behavior of 8-hydroxy-2'-deoxyguanosine(8-OH-dG) at the composite film modified electrode was studied.The effects of scan rates,pH and the interference of uric acid(UA) on the voltammetric behavior and detection of 8-OH-dG were also discussed.The experimental results suggest that the electrochemical behavior of 8-OH-dG at the composite film modified electrode was greatly improved due to the combination of the advantages of P3MT and DNA.In 0.1 M pH 7.0 phosphate buffer solution(PBS),the anodic peak currents of 8-OH-dG were linear with the 8-OH-dG concentration in two intervals,viz.0.28―4.2 μM and 4.2―19.6 μM.The detection limit of 56 nM 8-OH-dG could be estimated(S/N=3).This proposed composite film modified electrode shows excellent reproducibility and stability.It may have the potential application for the detection of 8-OH-dG in human urine. | WANG YanHuai,LI Jing,LIU Yan,MA RongNa,JIA WenLi,CUI Hui & WANG HuaiSheng College of Chemistry and Chemical Engineering,Institute of Analytical Chemistry,Liaocheng University,Liaocheng 252059,China | 2009 | Science China Chemistry2009,52,11: | 2 |
