1Hepatitis B virus,HBx mutants and their role in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is one of the leading causes of death induced by cancer in the modern world and majority of the cases are related to chronic hepatitis B virus(HBV)infection.HBV-encoded X protein(HBx)is known to play a pivotal role in the pathogenesis of viral induced HCC.HBx is a multifunctional protein of17 kDa which modulates several cellular processes by direct or indirect interaction with a repertoire of host factors resulting in HCC.HBX might interfere with several cellular processes such as oxidative stress,DNA repair,signal transduction,transcription,protein degradation,cell cycle progression and apoptosis.A number of reports have indicated that HBx is one of the most common viral ORFs that is often integrated into the host genome and its sequence variants play a crucial role in HCC.By mutational or deletion analysis it was shown that carboxy terminal of HBx has a likely role in protein-protein interactions,transcriptional transactivation,DNA repair,cell,signaling and pathogenesis of HCC.The accumulated evidence thus far suggests that it is difficult to understand the mechanistic nature of HBx associated HCC,and HBx mediated transcriptional transactivation and signaling pathways may be a major determinant.This article addresses the role of HBx in the development of HCC with particular emphasis on HBx mutants and their putative targets.Ashraf Ali Hany Abdel-Hafiz Mohd Suhail Amany Al-Mars Mohammad Khalid Zakaria Kaneez Fatima Sultan Ahmad Esam Azhar Adeel Chaudhary Ishtiaq Qadri 2014World Journal of Gastroenterology2014,20,30:29
2Epidemiological, laboratory, diagnostic and public health aspects of human brucellosis in western Iran显示文摘Objective:To determine brucellosis's epidemiologic,laboratory,diagnostic and public health features considering brucellosis is endemic in Azna County,western Iran.Methods:This descriptive cross-sectional study was investigated on 43 patients with brucellosis in Azna County. The subjects were the patients with symptoms correspondent with brucellosis and positive Wright and 2ME tests. A questionnaire about demographic,epidemiological and laboratory findings was filled in. Afterwards,patients were treated using usual antimicrobial drugs regimen. The collected data were analyzed by SPSS software version 16.Results:Forty-three subjects were found to be positive in laboratory tests. Incidence of Brucellosis was 59.31 per hundred thousand population. About 34.9% of patients were female and 65.1%male. Nearly 95.2% of human cases were living in rural and 4.8%in urban areas. Around 20.9%of patients had history of animal contact. The commonest transmission was unpasteurized dairy products(79.1%). The most contagious seasons were summer and spring(60.3%). The most common age group was 15-24(27.9%),and about 60.5%of the patients were between 15-44 years old. Disease was more common among housewives(30.2%) and farmers(20.9%). The majority of the patients had Wright test titre=1:320(54.1%) and 2ME test titre=1:160(56.1%) in serological titration.Doxycycline with Rifampin was used for treatment of the greatest of patients(60.4%).Conclusions:In order to control this zoonotic disease,close cooperation of health and veterinary organizations is necessary.Hamid Kassiri Hamid Amani MassoudLotfi 2013Asian Pacific Journal of Tropical Biomedicine2013,3,8:19
3Treatment responses in Asians and Caucasians with chronic hepatitis C infection显示文摘AIM:To conduct a multicentre retrospective review of virological response rates in Asians infected with genotype 1 chronic hepatitis C(CHC) treated with combination interferon and ribavirin and then to compare their responses to that among Caucasians.METHODS:Asian patients infected with genotype 1 CHC treated at 4 Australian centres between 2001 to 2005 were identified through hospital databases.Baseline demographic characteristics,biochemical,virological and histological data and details of treatment were collected.Sustained virological responses(SVR) in this cohort were then compared to that in Caucasian subjects,matched by genotype,age,gender and the stage of hepatic fibrosis.RESULTS:A total of 108 Asians with genotype 1 CHC were identified.The end of treatment response(ETR) for the cohort was 79% while the SVR was 67%.Due to the relatively advanced age of the Asian cohort,only sixty-four subjects could be matched with Caucasians.The ETR among matched Asians and Caucasians was 81% and 56% respectively(P=0.003),while the SVR rates were 73% and 36%(P <0.001) respectively.This difference remained significant after adjusting for other predictive variables.Kenneth K Yan Marianne Guirgis Amany Zekry Thuy Dinh Anouk Dev Jacob George Alice Lee 2008World Journal of Gastroenterology2008,14,21:12
4Bone marrow-derived mesenchymal stem cells ameliorate sodium nitrite-induced hypoxic brain injury in a rat model显示文摘Sodium nitrite(Na NO2) is an inorganic salt used broadly in chemical industry. Na NO2 is highly reactive with hemoglobin causing hypoxia. Mesenchymal stem cells(MSCs) are capable of differentiating into a variety of tissue specific cells and MSC therapy is a potential method for improving brain functions. This work aims to investigate the possible therapeutic role of bone marrow-derived MSCs against Na NO2 induced hypoxic brain injury. Rats were divided into control group(treated for 3 or 6 weeks), hypoxic(HP) group(subcutaneous injection of 35 mg/kg Na NO2 for 3 weeks to induce hypoxic brain injury), HP recovery groups N-2 w R and N-3 w R(treated with the same dose of Na NO2 for 2 and 3 weeks respectively, followed by 4-week or 3-week self-recovery respectively), and MSCs treated groups N-2 w SC and N-3 w SC(treated with the same dose of Na NO2 for 2 and 3 weeks respectively, followed by one injection of 2 × 106 MSCs via the tail vein in combination with 4 week self-recovery or intravenous injection of Na NO2 for 1 week in combination with 3 week self-recovery). The levels of neurotransmitters(norepinephrine, dopamine, serotonin), energy substances(adenosine monophosphate, adenosine diphosphate, adenosine triphosphate), and oxidative stress markers(malondialdehyde, nitric oxide, 8-hydroxy-2′-deoxyguanosine, glutathione reduced form, and oxidized glutathione) in the frontal cortex and midbrain were measured using high performance liquid chromatography. At the same time, hematoxylin-eosin staining was performed to observe the pathological change of the injured brain tissue. Compared with HP group, pathological change of brain tissue was milder, the levels of malondialdehyde, nitric oxide, oxidized glutathione, 8-hydroxy-2′-deoxyguanosine, norepinephrine, serotonin, glutathione reduced form, and adenosine triphosphate in the frontal cortex and midbrain were significantly decreased, and glutathione reduced form/oxidized glutathione and adenosine monophosphate/adenosine triphosphate ratio were significantly increased in the MSCs treated groups. 5The aflatoxin B1 isolating potential of two lactic acid bacteria显示文摘Objective:To determine lactic acid bacteria's capability to enhance the process of binding and isolating aflatoxin B1 and to utilize such lactic acid bacteria as a food supplement or probiotic products for preventing absorption of aflatoxin Bl in human and animal bodies.Methods:In the present research,the bacteria were isolated from five different sources.For surveying the capability of the bacteria in isolating aflatoxin Bl,ELISA method was implemented,and for identifying the resultant strains through 16S rRNA sequencing method,universal primers were applied.Results:Among the strains which were isolated,two strains of Lactobacillus pentosus and Lactobacillus beveris exhibited the capability of absorbing and isolating aflatoxin Bl by respectively absorbing and discharging 17.4%and 34.7%of the aforementioned toxin existing in the experiment solution.Conclusions:Strains of Lactobacillus pentosus and Lactobacillus beveris were isolated from human feces and local milk samples,respectively.And both strains has the ability to isolate or bind with aflatoxin Bl.Adel Hamidi Reza Mirnejad Emad Yahaghi Vahid Behnod Ali Mirhosseini Sajad Amani Sara Sattari Ebrahim Khodaverdi Darian 2013Asian Pacific Journal of Tropical Biomedicine2013,3,9:8
6Visible light optical coherence tomography measures retinal oxygen metabolic response to systemic oxygenation显示文摘The lack of capability to quantify oxygen metabolism noninvasively impedes both fundamental investigation and clinical diagnosis of a wide spectrum of diseases including all the major blinding diseases such as age-related macular degeneration,diabetic retinopathy,and glaucoma.Using visible light optical coherence tomography(vis-OCT),we demonstrated accurate and robust measurement of retinal oxygen metabolic rate(rMRO2)noninvasively in rat eyes.We continuously monitored the regulatory response of oxygen consumption to a progressive hypoxic challenge.We found that both oxygen delivery,and rMRO2 increased from the highly regulated retinal circulation(RC)under hypoxia,by 0.2860.08 μL min(^-1)(p,0.001),and 0.2060.04 μL min(^-1)(p,0.001)per 100 mmHg systemic pO2 reduction,respectively.The increased oxygen extraction compensated for the deficient oxygen supply from the poorly regulated choroidal circulation.Results from an oxygen diffusion model based on previous oxygen electrode measurements corroborated our in vivo observations.We believe that vis-OCT has the potential to reveal the fundamental role of oxygen metabolism in various retinal diseases.Ji Yi Wenzhong Liu Siyu Chen Vadim Backman Nader Sheibani Christine M.Sorenson Amani A.Fawzi Robert A.Linsenmeier Hao F.Zhang 2015Light(Science & Applications)2015,4,1:8
7Recrystallization and mechanical properties of WE43 magnesium alloy processed via cyclic expansion extrusion显示文摘In this study, cyclic expansion extrusion(CEE), as a relatively new severe plastic deformation(SPD) process, is applied to a rare earth(RE) containing Mg alloy WE43. The effects of the processing temperature and the number of passes are also investigated. The results showed that dynamic recrystallization(DRX) occurred after CEE processing at 400°C, and a bimodal structure with ultrafine DRXed grains surrounded the unrecrystallized grains. However, the DRX at 330°C was retarded because of the existence of RE elements. The tensile tests showed that a simultaneous increase in the strength and the ductility of WE43 is obtained after CEE processing at 400°C via two passes. Furthermore, the highest ultimate tensile strength of 440 MPa was achieved after the second pass of CEE at 330°C, and the highest ductility of 21% was attained after the second pass of CEE at 400°C. The microhardness measurements showed that the hardness increased from HV 80 to HV 114 and HV 98 after two passes of CEE processing at 330 and 400°C, respectively. In conclusion, increasing the processing passes could increase the mechanical properties and the volume fraction of the recrystallized grains. 8Incidence and characteristics of HBV reactivation in hematological malignant patients in south Egypt显示文摘AIM:To investigate characteristics of hepatitis B virus(HBV)implicated in HBV reactivation in patients with hematological malignancies receiving immunosuppressive therapy.METHODS:Serum samples were collected from 53 patients with hematological malignancies negative for hepatitis B surface antigen(HBsAg)before the start of and throughout the chemotherapy course.HBV reactivation was diagnosed when the HBsAg status changed from negative to positive after the initiation of chemotherapy and/or when HBV DNA was detected by realtime detection polymerase chain reaction(RTD-PCR).For detecting the serological markers of HBV infection,HBsAg as well as antibodies to the core antigen(antiHBc)and to the surface antigen were measured in the sera by CEIA.Nucleic acids were extracted from sera,and HBV DNA sequences spanning the S gene were amplified by RTD-PCR.The extracted DNA was further subjected to PCR to amplify the complete genome as well as the specific genomic sequences bearing the enhancerⅡ/core promoter/pre-core/core regions(nt1628-2364).Amplicons were sequenced directly.RESULTS:Thirty-five(66%)of the 53 HBsAg-negative patients were found to be negative serologically for antiHBc,and the remaining 18(34%)patients were positive for anti-HBc.Five of the 53(9.4%)patients with hematologic malignancies experienced HBV reactivation.Genotype D1 was detected in all five patients.Four types of mutant strains were detected in the S gene product of HBV strains and were isolated from 3 patients with HBV reactivation:T/S120,L143,and I126.HBV DNA was detected in the pretreatment HBsAg-negative samples in one of the five patients with HBV reactivation.In this patient,sequences encompassing the HBV full genome obtained from sera before the start of chemotherapy and at the time of de novo HBV hepatitis were detected and it showed 100%homology.Furthermore,in the phylogenetic tree,the sequences were clustered together,thereby indicating that this patient developed reactivation from an occult HBV infection.CONCLUSION:Past infection with HBV is a risk factor for HBV reactivation in Egypt.Mandatory anti-HBc screening prior to chemotherapy in patients with hematological malignancies is recommended.Abeer Elkady Sahar Aboulfotuh Elsayed Mostafa Ali Douaa Sayed Nashwa M Abdel-Aziz Amany M Ali Shuko Murakami Sayuki Iijima Yasuhito Tanaka 2013World Journal of Gastroenterology2013,19,37:7
9Trauma patterns in patients attending the Emergency Department of Jazan General Hospital, Saudi Arabia显示文摘BACKGROUND: Modern civilization and the sharp rise in living standards have led to dramatic changes in trauma pattern in Saudi Arabia. This study aimed to describe the different patterns of injuries of patients attending the Emergency Department of Jazan General Hospital(JGH) in the southwest corner of Saudi Arabia.METHODS: A total number of 1 050 patients were enrolled in the study. A pre-organized data sheet was prepared for each patient attended the Emergency Department of JGH from February 2012 to January 2013. It contains data about socio-demographics, trauma data, clinical evaluation results, investigations as well as treatment strategies.RESULTS: The mean age of the patients was 25.3±16.8 years. Most(45.1%) of the patients were at age of 18–30 years. Males(64.3%) were affected by trauma more common than females. More than half(60.6%) of the patients were from urban areas. The commonest kind of injury was minor injury(60%), followed by blunt trauma(30.9%) and then penetrating trauma(9.1%). The mean time from the incident to arrival at hospital was 41.3±79.8 minutes. The majority(48.2%) of the patients were discharged after management of trivial trauma, whereas 2.3% were admitted to ICU, 7.7% transferred to inpatient wards, and 17.7% observed and subsequently discharged. The mortality rate of the patients was 2.6%.CONCLUSION: Trauma is a major health problem, especially in the young population in Saudi Arabia. 10Influence of bioactive sulphated polysaccharide-protein complexes on hepatocarcinogenesis, angiogenesis and immunomodulatory activities显示文摘Objective:To explore the in vivo anticancer,anti-angiogenesis and immunomodulatory efficacies of the bioactive polysaccharide isolated from cold aqueous extract of Jania rubens(JCEM) and Pterocladia capillacea(PCEM) as well as hot aqueous extract of Enteromorpha intestinalis(EHEM) against hepatocellular carcinoma rat model(HCC) and to study their chemical composition.Methods:The sugars and amino acids composition of the bioactive polysaccharides of JCEM,PCEM and EHEM were determined using gas liquid chromatography and amino acid analyzer,respectively.These polysaccharide extracts(20 mg/kg b.wt.for 5 weeks) were assessed on hepatocarcinogenesis in rats and α-fetoprotein(AFP),carcinoembryonic antigen(CEA),glypican-3(GPC-3),hepatocyte growth factor(HGF) and vascular endothelial growth factor(VEGF) and Ig G levels were evaluated.Results:The GLC analysis of JCEM,PCEM and EHEM polysaccharide revealed the presence of 10,9 and10 sugars,in addition the amino acid analyser enable identification of 16,15 and 15 amino acids,respectively.These polysaccharide extracts of JCEM,PCEM and EHEM produced significant decrease in serum AFP,CEA,GPC-3,HGF and VEGF compared with untreated HCC group.JCEM,PCEM and EHEM had an immunostimulatory responses by increasing the IgG levels as compared by naive value(1.23,1.53 and 1.17 folds),respectively.The bioactive polysaccharides in HCC induced rats improved the humoral immune response.The photomicrographs of liver tissue sections of the groups of HCC treated with polysaccharide extracts of Jania rubens and Enteromorpha intestinalis showed intact histological structure.Moreover,fractions HE1,HE4,HE7 obtained from polysaccharide of EHEM showed moderate cytotoxic activity against Hep G2 in vitro with IC_(50) 73.1,42.6,76.2 μg/mL.However,fractions of PCEM and JCEM show no or weak cytotoxicity against Hep G2 in vitro where the cytotoxic activity of their crude polysaccharide extract proved synergetic effect.Conclusions:The pronounced antitumor activity of sulphated polysaccharide-protein complexes of JCEM and EHEM is due to direct cytotoxic activity,anti-hepatocarcinogensis,and anti-angiogenesis.In addition,JCEM,PCEM and EHEM had an immunostimulatory response and improved the humoral immune response in HCC induced rats.Azza A.Matloub Hadeer A.Aglan Sahar Salah Mohamed El Souda Mona Elsayed Aboutabl Amany Sayed Maghraby Hanaa H.Ahmed 2016Asian Pacific Journal of Tropical Medicine2016,9,12:7

12Immunomodulatory effects of astragalus polysaccharide on Human peripheral blood mononuclear cells co-cultured with cervical cancer cell lineImmunomodulatory effects of astragalus polysaccharide on Human peripheral blood mononuclear cells co-cultured with cervical cancer cell line显示文摘OBJECTIVE:To investigate immunomodulatory effects of Astragalus polysaccharides(APS)on the co-culture of peripheral blood mononuclear cells(PBMCs)with He La cervical cancer cell line.METHODS:To assess the proliferation of PBMCs,carboxyfluorescein succinimidyl ester(CFSE)-labeled PBMCs were co-cultured with He La cells and treated with different concentrations of APS.Supernatants of cell culture were collected for cytokines assay via enzyme-linked immunosorbent assay(ELISA).The impact of APS on the proliferation of PBMCs,induction of regulatory T cells(Tregs),and myeloid-derived suppressor cells(MDSCs)was carried out by flow cytometry.RESULTS:It was observed that APS could increase the proliferation of PBMCs co-cultured with He La cells(P<0.05).However,APS had no significant effects on the induction of Tregs and MDSCs in the co-culture assay(P>0.05).Furthermore,ELISA results demonstrated that APS could decrease IL-10 and TGF-βconcentration(P<0.05).CONCLUSION:The above-mentioned characteristics showed that APS might be able to modulate immune responses and improve anti-tumor effects through increasing the proliferation of PBMCs and decreasing inhibitory cytokines secretion as critical mediators of immune suppression in the tumor microenvironment.Elham Shokati Mohammad-Reza Shokri Kobra Entezami Samaneh Khorrami Mona Amani Morteza Motallebnezhad Elahe Safari 2021Journal of Traditional Chinese Medicine2021,41,5:5
13Compliant plant wearables for localized microclimate and plant growth monitoring显示文摘The microclimate surrounding a plant has major effect on its health and photosynthesis process,where certain plants struggle in suboptimal environmental conditions and unbalanced levels of humidity and temperature.The ability to remotely track and correlate the effect of local environmental conditions on the healthy growth of plants can have great impact for increasing survival rate of plants and augmenting agriculture output.This necessitates the widespread distribution of lightweight sensory devices on the surface of each plant.Using flexible and biocompatible materials coupled with a smart compact design for a low power and lightweight system,we develop widely deployed,autonomous,and compliant wearables for plants.The demonstrated wearables integrate temperature,humidity and strain sensors,and can be intimately deployed on the soft surface of any plant to remotely and continuously evaluate optimal growth settings.This is enabled through simultaneous detection of environmental conditions while quantitatively tracking the growth rate(viz.elongation).Finally,we establish a nature-inspired origami-assembled 3D-printed“PlantCopter”,used as a launching platform for our plant wearable to enable widespread microclimate monitoring in large fields.Joanna M.Nassar Sherjeel M.Khan Diego Rosas Villalva Maha M.Nour Amani S.Almuslem Muhammad M.Hussain 2018npj Flexible Electronics2018,2,1:5
14Phytochemical studies and anti-ulcerative colitis effect of Moringa oleifera seeds and Egyptian propolis methanol extracts in a rat model显示文摘Objective: To analyze the phytochemical constituents, and to explore potential protective effect of the methanol extract of Moringa oleifera(M. oleifera) seeds and Egyptian propolis, each alone or concurrently administered on acetic acid-induced ulcerative colitis in rats. Methods: Eight groups of 5 rats each were used: normal control group with distilled water, model group, two groups with M. oleifera seeds(100 and 200 mg/kg), two groups with propolis(50 and 100 mg/kg), one group with concurrent administration of both, and one group with prednisolone(reference drug). Macro-and microscopic picture, ulcer index and lesion scores, oxidative markers, inflammatory mediators, in vitro activity of the inflammatory enzymes and 1, 1-diphenyl-2-picrylhydrazyl free radicals scavenging activity were evaluated. The phytochemical constituents of both extracts were explored by GC-MS analysis. Results: Both treatments modulated the macro-and microscopic picture, decreased the ulcerative index, lesion score, oxidative markers and inflammatory mediators, and inhibited the COX-1 and COX-2 enzymes. Propolis appeared to be powerful free radicals scavenger. A powerful synergistic effect of both treatments in modulating the course of the disease was reported. GCMS analysis of methanol extract of M. oleifera seeds and propolis revealed the presence of 50 and 34 compounds, respectively. Conclusions: M. oleifera seeds and propolis methanol extracts have modulated the course of acetic acid-induced ulcerative colitis. Moreover, both treatments induce a good synergistic effect against the disease. 15Acid leaching of heavy metals from contaminated soil collected from Jeddah,Saudi Arabia:kinetic and thermodynamics studies显示文摘Urban soils polluted with heavy metals are of increasing concern because it is greatly affecting human health and the ecological systems.Hence,it is mandatory to understand the reasons behind this pollution and remediate the contaminated solid.The removal of heavy metals from contaminated soil samples collected from the vicinity of the sewage lake in Jeddah,Saudi Arabia,was explored.The leaching process was studied kinetically and thermodynamically for better understanding of the remediation process.The results showed that the soil samples were slightly basic in nature,and tend to be more neutral away from the main contaminated sewage lake area.The total metal content in the soil samples was measured using the aqua regia extractions by ICPOES and the results showed that many of the heavy metals present have significant concentrations above the tolerable limits.In general,the metal concentrations at different sites indicated that the heavy metal pollution is mainly due to the sewage discharge to the lake.The results showed excellent correlation between the concentrations of Co,As,and Hg with the distance from the main contaminated area.The leaching of Co,As,and Hg using 1.0 M hydrochloric acid from the soil was studied kinetically at different temperatures and the experimental results were fitted using different kinetics models.The experimental data were best described with two-constant rate and Elovich equation kinetic models.Also,the thermodynamic study showed that the leaching process was spontaneous,endothermic and accompanied with increase in the entropy.In general,the polluted soil could be remediated successfully from the heavy metals using the acid leaching procedure in a short period of time.Shorouq I.Alghanmi Amani F.Al Sulami Tahani A.El-Zayat Basma G.Alhogbi Mohamed Abdel Salam 2015International Soil and Water Conservation Research2015,3,3:4
16A generalized supervised classification scheme to produce provincial wetland inventory maps:an application of Google Earth Engine for big geo data processing显示文摘Wetlands are important natural resources due to their numerous ecological services.Consequently,identifying their locations and extents is imperative.The stability,repeatability,cost-effectiveness,multi-scale coverage,and proper spatial resolution imagery of satellites provide a valuable opportunity for their use in various large-scale applications,such as provincial wetland mapping.To do so,it is required to(1)process and classify big geo data(i.e.a large amount of satellite datasets)in a time-and computationally-efficient approach and(2)collect a large amount of field samples.In this study,Google Earth Engine(GEE)and machine learning algorithms were utilized to process thousands of remote sensing images and produce provincial wetland inventory maps of the three Canadian provinces of Manitoba,Quebec,and Newfoundland and Labrador(NL).Additionally,using GEE,a generalized supervised classification method is proposed to produce a regional wetland map from a large area(e.g.,a province)when lacking field samples.In fact,using the field data from only Manitoba and assuming that all wetlands in Canada have similar characteristics,the wetland maps were generated for the other two provinces.The overall classification accuracies for Manitoba,Quebec,and NL were 84%,78%,and 82%,respectively,indicating the high potential of the proposed method for aiding provincial wetland inventory systems.Meisam Amani Brian Brisco Majid Afshar S.Mohammad Mirmazloumi Sahel Mahdavi Sayyed Mohammad Javad Mirzadeh Weimin Huang Jean Granger 2019Big Earth Data2019,3,4:4
17DNA extraction from fresh-frozen and formalin-fixed, paraffinembedded human brain tissue显示文摘Both fresh-frozen and formalin-fixed,paraffinembedded(FFPE)human brain tissues are invaluable resources for molecular genetic studies of central nervous system diseases,especially neurodegenerative disorders.To identify the optimal method for DNA extraction from human brain tissue,we compared methods on differently-processed tissues.Fragments of LRRK2 and MAPT(257 bp and 483 bp/245 bp)were amplified for evaluation.We found that for FFPE samples,the success rate of DNA extraction was greater when using a commercial kit than a laboratory-based method(successful DNA extraction from 76%versus 33%of samples).PCR amplicon size and storage period were key factors influencing the success rate of DNA extraction from FFPE samples.In the fresh-frozen samples,the DNA extraction success rate was 100%using either a commercial kit(QIAamp DNA Micro)or a laboratorybased method(sample boiling in 0.1 mol/L NaOH,followed by proteinase K digestion,and then DNA extraction using Chelex-100)regardless of PCR amplicon length or tissue storage time.Although the present results demonstrate that PCR-amplifiable genomic DNA can be extracted from both fresh-frozen and FFPE samples,fresh brain tissue is recommended for DNA extraction in future neuropathological studies.Jian-Hua Wang Amany Gouda-Vossos Nicolas Dzamko Glenda Halliday Yue Huang 2013Neuroscience Bulletin2013,29,5:3
18Nogo receptor expression in microglia/macrophages during experimental autoimmune encephalomyelitis progression显示文摘Myelin-associated inhibitory factors within the central nervous system(CNS) are considered to be one of the main obstacles for axonal regeneration following disease or injury. The nogo receptor 1(NgR1) has been well documented to play a key role in limiting axonal regrowth in the injured and diseased mammalian CNS. However, the role of nogo receptor in immune cell activation during CNS inflammation is yet to be mechanistically elucidated. Microglia/macrophages are immune cells that are regarded as pathogenic contributors to inflammatory demyelinating lesions in multiple sclerosis(MS). In this study, the animal model of MS, experimental autoimmune encephalomyelitis(EAE) was induced in ngr1^(+/+) and ngr1^(–/–) female mice following injection with the myelin oligodendrocyte glycoprotein(MOG_(35–55)) peptide. A fatemap analysis of microglia/macrophages was performed throughout spinal cord sections of EAE-induced mice at clinical scores of 0, 1, 2 and 3, respectively(increasing locomotor disability) from both genotypes, using the CD11 b and Iba1 cell markers. Western immunoblotting using lysates from isolated spinal cord microglia/macrophages, along with immunohistochemistry and flow cytometric analysis, was performed to demonstrate the expression of nogo receptor and its two homologs during EAE progression. Myelin protein engulfment during EAE progression in ngr1^(+/+) and ngr1^(–/–) mice was demonstrated by western immunblotting of lysates from isolated spinal cord microglia/macrophages, detecting levels of Nogo-A and MOG. The numbers of M1 and M2 microglia/macrophage phenotypes present in the spinal cords of EAE-induced ngr1^(+/+) and ngr1^(–/–) mice, were assessed by flow cytometric analysis using CD38 and Erg-2 markers. A significant difference in microglia/macrophage numbers between ngr1^(+/+) and ngr1^(–/–) mice was identified during the progression of the clinical symptoms of EAE, in the white versus gray matter regions of the spinal cord. This difference was unrelated to the expression of Ng R on these macrophage/microglial cells. We have identified that as EAE progresses, the phagocytic activity of microglia/macrophages with myelin debris, in ngr1^(–/–) mice, was enhanced. Moreover, we show a modulation from a predominant M1-pathogenic to the M2-neurotrophic cell phenotype in the ngr1^(–/–) mice during EAE progression. 20Hypoglycemic and antioxidant activities of Caesalpinia ferrea Martius leaf extract in streptozotocininduced diabetic rats显示文摘Objective: To evaluate the antidiabetic and antioxidant ef ects of aqueous ethanolic extract of Caesalpinia ferrea(C. ferrea) leaf in normal and streptozotocin(STZ) induced diabetic rats.Methods: Male Sprague-Dawley rats divided into 6 groups of 6 rats each were assigned into diabetic and non-diabetic groups. Diabetes was induced in rats by single intraperitoneal administration of STZ(65 mg/kg body weight). C. ferrea extract at the doses of 250 and 500 mg/kg body weight was orally administered to both diabetic and non-diabetic animals for a period of 30 days. After completion of experimental duration serum, liver and pancreas were used for evaluating biochemical and histopathological changes.Results: Oral administration of C. ferrea leaf extract significantly reduced elevated serum glucose, α-amylase, liver function levels and signii cantly increased serum insulin, total protein and body weight as well as improved lipid proi le due to diabetes. Furthermore, the treatment resulted in a marked increase in glutathione peroxidase, superoxide dismutase, catalase and reduced glutathione, and diminished levels of lipid peroxidation in liver and pancreas of diabetic rats. Histopathological studies demonstrated the reduction in the pancreas and liver damage and coni rmed the biochemical i ndings.Conclusions: From the present study, it can be concluded that the C. ferrea leaf extract ef ectively improved hyperglycaemia while inhibiting the progression of oxidative stress in STZ-induced diabetic rats.
