1Malignant biliary obstruction:From palliation to treatment显示文摘Malignant obstruction of the bile duct from cholan-giocarcinoma,pancreatic adenocarcinoma,or other tumors is a common problem which may cause debilitating symptoms and increase the risk of subsequent surgery.The optimal treatment-including the decision whether to treat prior to resection-depends on the type of malignancy,as well as the stage of disease.Preoperative biliary drainage is generally discouraged due to the risk of infectious complications,though some situations may benefit.Patients who require neoadjuvant therapy will require decompression for the prolonged period until attempted surgical cure.For pancreatic cancer patients,self-expanding metallic stents are superior to plastic stents for achieving lasting decompression without stent occlusion.For cholangiocarcinoma patients,treatment with percutaneous methods or nasobiliary drainage may be superior to endoscopic stent placement,with less risk of infectious complications or failure.For patients of either malignancy who have advanced disease with palliative goals only,the choice of stent for endoscopic decompression depends on estimated survival,with plastic stents favored for survival of < 4 mo.New endoscopic techniques may actually extend stent patency and patient survival for these patients by achieving local control of the obstructing tumor.Both photodynamic therapy and radiofrequency ablation may play a role in extending survival of patients with malignant biliary obstruction.Brian R Boulay Aleksandr Birg 2016World Journal of Gastrointestinal Oncology2016,8,6:24

9Functionality of a bicistronic construction containing HEXA and HEXB genes encoding β-hexosaminidase A for cell-mediated therapy of GM2 gangliosidoses显示文摘Tay-Sachs disease and Sandhoff disease are severe hereditary neurodegenerative disorders caused by a deficiency ofβ-hexosaminidase A(HexA)enzyme,which results in the accumulation of GM2 gangliosides in the nervous system cells.In this work,we analyzed the efficacy and safety of cell-mediated gene therapy for Sandhoff disease and Sandhoff disease using a bicistronic lentiviral vector encoding cDNA of HexAα-andβ-subunit genes separated by the nucleotide sequence of a P2A peptide(HEXA-HEXB).The functionality of the bicistronic construct containing the HEXA-HEXB genetic cassette was analyzed in a culture of HEK293T cells and human umbilical cord blood mononuclear cells(hUCBMCs).Our results showed that the enzymatic activity of HexA in the conditioned medium harvested from genetically modified HEK293T-HEXA-HEXB and hUCBMCs-HEXA-HEXB was increased by 23 and 8 times,respectively,compared with the conditioned medium of native cells.Western blot analysis showed that hUCBMCs-HEXA-HEXB secreted both completely separated HEXA and HEXB proteins,and an uncleaved protein containing HEXA+HEXB linked by the P2A peptide.Intravenous injection of genetically modified hUCBMCs-HEXA-HEXB to laboratory Wistar rats was carried out,and the HexA enzymatic activity in the blood plasma of experimental animals,as well as the number of live cells of immune system organs(spleen,thymus,bone marrow,lymph nodes)were determined.A significant increase in the enzymatic activity of HexA in the blood plasma of laboratory rats on days 6 and 9(by 2.5 and 3 times,respectively)after the administration of hUCBMCsHEXA-HEXB was shown.At the same time,the number of live cells in the studied organs remained unchanged.Thus,the functionality of the bicistronic genetic construct encoding cDNA of the HEXA and HEXB genes separated by the nucleotide sequence of the P2A peptide was shown in vitro and in vivo.We hypothesize that due to the natural ability of hUCBMCs to overcome biological barriers,such a strategy can restore the activity of the missing enzyme in the central nervous system of patients with GM2 gangliosidoses.Based on the obtained data,it can be concluded that intravenous administration of hUCBMCs with HexA overexpression is a promising method of the therapy for GM2 gangliosidoses.The animal protocol was approved by the Animal Ethics Committee of the Kazan Federal University(No.23)on June 30,2020.Alisa A.Shaimardanova Daria S.Chulpanova Valeriya V.Solovyeva Aleksandr M.Aimaletdinov Albert A.Rizvanov 2022Neural Regeneration Research2022,17,1:3

15Tailoring the LiNbO_(3)coating of Ni-rich cathode materials for stable and high-performance all-solid-state batteries显示文摘The research and development of advanced nanocoatings for high-capacity cathode materials is currently a hot topic in the field of solid-state batteries(SSBs).Protective surface coatings prevent direct contact between the cathode material and solid electrolyte,thereby inhibiting detrimental interfacial decomposition reactions.This is particularly important when using lithium thiophosphate superionic solid electrolytes,as these materials exhibit a narrow electrochemical stability window,and therefore,are prone to degradation during battery operation.Herein we show that the cycling performance of LiNbO_(3)-coated Ni-rich LiNi_(x)Co_(y)Mn_(z)O_(2)cathode materials is strongly dependent on the sample history and(coating)synthesis conditions.We demonstrate that post-treatment in a pure oxygen atmosphere at 350℃results in the formation of a surface layer with a unique microstructure,consisting of LiNbO_(3)nanoparticles distributed in a carbonate matrix.If tested at 45℃and C/5 rate in pellet-stack SSB full cells with Li_(4)Ti_(5)O_(12)and Li_(6)PS_(5)Cl as anode material and solid electrolyte,respectively,around 80%of the initial specific discharge capacity is retained after 200 cycles(~160 mAh·g^(−1),~1.7 mAh·cm^(−2)).Our results highlight the importance of tailoring the coating chemistry to the electrode material(s)for practical SSB applications.Seyedhosein Payandeh Florian Strauss Andrey Mazilkin Aleksandr Kondrakov Torsten Brezesinski 2022Nano Research Energy2022,1,3:2

20Variable retention forestry in European boreal forests in Russia显示文摘We explored whether,and to what extent,variable retention(VR)forestry has been applied in European boreal forests in northwestern Russia.Our survey revealed VR since 1910.Between 1910 and the 1960s,the statistics showing how much was retained are largely missing.However,for example,in the 1950s,on a large scale in the Republic of Karelia,up to 200-ha-sized harvesting areas,18–33%,were retention patches with a mean growing stock of 30–40m3 ha−1.In the fellings defined as“incomplete clear fellings,”which were the most common final felling type at that time,11–40%of the growing stock was left.Between the 1960s and the early 1990s,with more efficient harvesting and skidding techniques,conventional clear fellings with a much lower amount of retention were practiced.Concern about the regeneration of harvested areas gradually led to smaller(maximum 50 ha)harvesting areas and the increase of silvicultural activities.Until now,to ensure natural regeneration,patches of understory and 20–25 seed trees(i.e.,ca.15–25m3)per ha have been left permanently in harvesting areas.Landscape-scale retention for protecting ecosystem functions and biodiversity was legislated in 1978 by preserving key biotopes up to 1000 ha in size.Since 2001,promoted by forest certification,the key biotopes,such as paludified forest patches,buffers around water bodies,and habitats of red-listed species,have also been retained in harvesting areas,together with a dispersed retention of different elements.Quantitative estimates of the amount of key biotopes are largely missing.However,estimates of 1–13%in harvesting areas and 23%in whole managed landscapes have been given.VR applied during the last century has emulated natural disturbances and created diverse uneven-aged forest structures with high amounts of diverse coarse woody debris.We conclude that an analysis of past and current retention practices is essential for estimating the global role of Russian forestry.Further decisions on the general direction of Russian forestry and,specifically,retention practices are important to address the global challenges of biodiversity loss and climate change.Ekaterina Shorohova Sergey Sinkevich Aleksandr Kryshen Ilkka Vanha-Majamaa 2019Ecological Processes2019,8,1:1
