| 1 | 土霉素对柑橘黄龙病的防治效果及对柑橘产量和品质的影响显示文摘为有效防控柑橘黄龙病,于2017年在佛罗里达大学柑橘研究与教育中心的奥本代尔市柑橘试验场进行田间试验筛选黄龙病的防治药剂及其浓度,测定注射土霉素后叶片中柑橘黄龙病菌亚洲种Cadidatus Liberibacter asiaticus、土霉素、淀粉含量、柑橘产量、出汁率、可溶性固形物含量和酸度,显微镜下观察注射土霉素后淀粉粒的分布。结果表明,浓度为1.6 g/株土霉素处理180 d后柑橘叶片中黄龙病菌亚洲种含量减少幅度最大,为91.78%;注射浓度为1.6 g/株土霉素后7 d,叶片中土霉素含量达142.2μg/kg,注射后11 d叶片中土霉素含量达到最大,为239.8μg/kg,注射后45 d叶片中土霉素含量低至99.6μg/kg以下;注射后7~90 d,叶片中黄龙病菌亚洲种含量呈波浪形变化,注射90 d后叶片中黄龙病菌亚洲种含量一直处于增长趋势,叶片中黄龙病菌亚洲种含量总体随着土霉素含量的升高而降低;注射浓度为1.6 g/株土霉素后,叶片中淀粉含量大幅度下降,60 d时达到最低值,为3.3μg/mm2,90 d时达到峰值,为14.5μg/mm2;单株产量为16.7 kg,与注射浓度为0.8 g/株土霉素处理差异不显著,但均显著高于其它2个处理;柑橘出汁率、可溶性固形物和酸度均与清水对照差异不显著。表明土霉素可有效抑制黄龙病菌亚洲种,减少柑橘叶片内淀粉含量,增加柑橘产量,但对果实品质未产生显著影响。 | 姚廷山 周彦 ACHOR Diann LI Jinyun WANG Nian 周常勇 | 2020 | 植物保护学报2020,47,6: | 6 |
| 2 | 土霉素处理对Valencia夏橙中淀粉及相关基因表达的影响显示文摘黄龙病(Huanglongbing,HLB)会造成柑橘韧皮部坏死堵塞,导致光合同化物运输不畅,淀粉大量积累。在感染HLB的4年生Valencia夏橙病株上分别注入0.1和0.2 g·株^(-1)土霉素(Oxytetracycline,OTC),90 d后运用qPCR检测,病株中HLB病原菌(Candidatus Liberibacter asiaticus,Las)含量均明显降低,且0.2和0.1 g·株^(-1) OTC处理的效果相当。I2/KI显色及LM观测表明,0.2 g·株^(-1) OTC处理后植株的淀粉含量从注射前的18.58μg·mm^(-2)减少至90 d的5.24μg·mm^(-2),而0.1 g·株^(-1)的处理90 d仅降至11.88μg·mm^(-2)。高效液相色谱(HPLC)检测结果表明,注射后90 d内,试验用浓度0.2 g·株^(-1) OTC在植株体内可降解至200μg·kg^(-1)以下。基因表达结果表明,注射0.2 g·株^(-1) OTC后30和90 d,淀粉合成及分解相关基因表达量均下降,其中淀粉合成相关基因AGPase表达量下降最显著,这与OTC注射后30和90 d叶片内淀粉含量下降结果一致。 | 姚廷山 周彦 Diann Achor 周常勇 | 2019 | 园艺学报2019,46,2: | 3 |
| 3 | 土霉素处理对柑橘黄龙病的防治效果及PP2基因表达的影响显示文摘【目的】评价土霉素(oxytetracycline,OTC)药剂对不同发病程度柑橘黄龙病(Huanglongbing,HLB)的防治效果,并检测OTC处理后韧皮部关键基因——韧皮部蛋白2(PP2)的表达量变化,为HLB的有效防控提供科学依据,也为OTC作用机理研究提供参考。【方法】应用可有效抑制、杀死病原菌的OTC,树干定量注射0.1 g/树于不同发病程度HLB(初感染、轻度发病和重度发病3组)的4年生Valencia夏橙,注射后7、30、60、90 d定期采集Valencia叶片样品监测HLB致病菌Cadidatus Liberibacter asiaticus(Las)含量、淀粉含量及PP2基因表达量变化。树干注射OTC后90 d采集不同处理Valencia的嫩叶、成熟叶片及茎,进行淀粉染色光学显微观察(LM),直观反映植株累积淀粉的变化。树干注射OTC后80 d调查柑橘嫩叶抽发的情况,综合一系列指标评价OTC对HLB的防治效果。【结果】注射OTC 0.1 g/树对初感染HLB的4年生Valencia植株治疗作用最明显,7 d后检测即为阴性,并可保持90 d,其成熟叶片的淀粉含量明显减少,但茎内仍有淀粉粒富集;轻度发病夏橙注射90 d后叶片内的Las含量从(1.68×10~6±858884)cells/g叶片减至(7.21×10~4±30981)cells/g叶片,下降幅度极为明显,其成熟叶片内的淀粉含量在90 d内一直呈下降趋势;重度发病植株注射后叶片内的Las含量从(4.10×108±3.04×10~8)cells/g叶片减至(2.80×10~7±2.70×10~7)cells/g叶片,但从数量而言仍然有较多分布,除了新长出的秋梢,其成熟叶及茎内的淀粉粒含量仍旧很大,说明0.1 g/株OTC不足以治愈4年生重度发病Valencia夏橙。对PP2基因表达分析结果显示,注射OTC后30 d,Valencia夏橙体内的PP2基因表达量大幅度下降,随后90 d内表达量稳定,与注射后30 d的表达量较为一致。【结论】OTC可以用于HLB的防控,对初感染(Las含量为<9.00×10~5 cells/g叶片)及轻度发病(Las含量为9.00×10~5—9.00×10~7 cells/g叶片)的柑橘植株治疗作用较好,不适用于重度发病植株(Las含量为>9.00×10~7 cells/g叶片)的治疗。注射OTC后,PP2基因表达量下降明显,说明OTC可有效减少韧皮部病菌等的胁迫压力。 | 姚廷山 周彦 DIANN ACHOR 王年 周常勇 | 2019 | 中国农业科学2019,0,7: | 3 |
