| 1 | Hepatoprotective and in vivo antioxidant activities of the hydroethanolic leaf extract of Mucuna pruriens (Fabaceae) in antitubercular drugs and alcohol models显示文摘AIM: Hepatotoxicity is a significantly increasing health problem worldwide, and the extent of the problem has stimulated interest in the search for hepatotherapeutic agents from plants. This study investigated the hepatoprotective and in vivo antioxidant activities of the hydroethanolic extract of Mucuna pruriens leaves in antitubercular and alcohol-induced hepatotoxicity assays in rats. METHOD: In each of the models used, seven groups were allotted. The different groups received normal saline(10 mL·kg-1, p.o.); hepatotoxicant(isoniazid-rifampicin, INH-RIF, 100 mg·kg-1, i.p. or 20% ethanol 5 g·kg-1, p.o.) and normal saline(10 mL·kg-1, p.o.); hepatotoxicant and extract at doses of 100, 200, and 400 mg·kg-1 p.o.; hepatotoxicant and silymarin 50 mg·kg-1 p.o.; and extract at 400 mg·kg-1 p.o.. On the 21st day of treatment, blood was collected for assessment of serum biochemical parameters and harvested liver samples were assessed for antioxidants. RESULTS: The hepatotoxicants significantly(P < 0.05-0.001) increased the levels of alanine transaminase(ALT), aspartate transaminase(AST), alkaline phosphatase(ALP), bilirubin, and malondialdehyde(MDA); and reduced the levels of catalase(CAT), superoxide dismutase(SOD), glutathione peroxidase(GPx), and reduced glutathione GSH compared to control. M. pruriens significantly reversed(P < 0.05-0.001) the elevation in the level of ALT, AST, ALP, and bilirubin caused by the hepatotoxicants. The extract(200 and 400 mg·kg-1) significantly reversed(P < 0.05) the diminution in the level of in vivo antioxidants and increased the level of MDA produced by INH-RIF. M. pruriens(100-400 mg·kg-1) elicited significant reduction(P < 0.001) in the level of MDA compared to the alcohol group. Silymarin also reversed the deleterious effects of the hepatotoxicants. | 5 | In vivo antimalarial activity and toxicological effects of methanolic extract of Cocos nucifera(Dwarf red variety) husk fibre显示文摘OBJECTIVE: Phytochemical constituents as well as antimalarial and toxicity potentials of the methanolic extract of the husk fi bre of Dwarf Red variety of Cocos nucifera were evaluated in this study.METHODS: The dried powdered husk fi bre was exhaustively extracted with hexane, ethyl acetate and methanol successively and the methanolic extract was screened for fl avonoids, phenolics, tannins, alkaloids, steroids, triterpenes, phlobatannins, anthraquinones and glycosides. A 4-day suppressive antimalarial test was carried out using Plasmodium berghei NK65-infected mice, to which the extract was administered at doses of 31.25, 62.5, 125, 250 and 500 mg/kg body weight(BW). Toxicity of the extract was evaluated in rats using selected hematological parameters and organ function indices after orally administering doses of 25, 50 and 100 mg/kg BW for 14 d.RESULTS: Phytochemical analysis revealed the presence of alkaloids, tannins, phenolics, saponins, glycosides, steroids and anthraquinones in the extract. Moreover, the extract reduced parasitemia by 39.2% and 45.8% at doses of 250 and 500 mg/kg BW respectively on day 8 post-inoculation. Various hematological parameters evaluated were not significantly altered(P>0.05) at all doses of the extract, except red blood cell count which was signifi cantly elevated(P<0.05) at 100 mg/kg BW. The extract significantly increased(P<0.05) urea, creatinine, cholesterol, high-density lipoprotein-cholesterol and bilirubin concentrations in the serum as well as atherogenic index, while it reduced albumin concentration significantly(P<0.05) at higher doses compared to the controls. | 7 | Anti-inflammatory and membrane stabilizing properties of methyl jasmonate in rats显示文摘The present investigation was carried out to evaluate anti-inflammatory and membrane stabilizing properties of methyl jasmonate(MJ) in experimental rat models of acute and chronic inflammation.The effects of MJ on acute inflammation were assessed using carrageenan-induced rat's paw edema model.The granuloma air pouch model was employed to evaluate the effects of MJ on chronic inflammation produced by carrageenan in rats.The number of white blood cells(WBC) in pouch exudates was estimated using light microscopy.The levels of biomarkers of oxidative stress,such as malondialdehyde(MDA),glutathione(GSH) and activity of antioxidant enzymes in the exudates,were determined using spectrophotometry.The membrane stabilizing property of MJ was assessed based on inhibition of hemolysis of rat red blood cells(RBC) exposed to hypotonic medium.Our results indicated that MJ(25-100 mg·kg-1,i.p.) produced significant anti-inflammatory activity in carrageenan-induced paw edema in rats(P < 0.05).MJ reduced the volume of pouch exudates and the number of WBC in carrageenan-induced granulomatous inflammation.It also exhibited potent antioxidant and membrane stabilizing activities.In conclusion,these findings suggest the therapeutic potentials of methyl jasmonate in disease conditions associated with inflammation and its anti-inflammatory activity may be related to its antioxidant and membrane stabilizing activities. | UMUKORO Solomon ALABI Akinyinka Oladipo EDUVIERE Anthony Taghogho AJAYI Abidemi Mayowa OLUWOLE Oluwafemi Gabriel | 2017 | Chinese Journal of Natural Medicines2017,15,3: | 0 |
| 8 | Byrsocarpus coccineus Schumach.&Thonn.(Connaraceae)叶子提取物的降血糖和抗氧化活性(英文)显示文摘AIM: Diabetes Mellitus is associated with significant morbidity and mortality worldwide. The need for enhanced efficacy and safety, and cheaper and more readily available new drugs has increased the search for new antidiabetic drugs from plants. This study was conducted to investigate the antidiabetic activity of the hydroethanolic leaf extract of B. coccineus in rats. METHODS: The effect of B. coccineus extract(100–800 mg·kg–1, p.o.) on blood glucose levels in normal and glucose loaded rats, and alloxan-induced diabetic rats was determined. After 10 days of treatment, blood samples were collected from rats for lipid and insulin profiling. Animals were thereafter sacrificed and the kidneys, heart, and liver were harvested for antioxidant indices assay. RESULTS: In normal rats, B. coccineus did not cause significant reduction in blood glucose. At the dose of 800 mg·kg–1, significant increase in blood glucose level was not observed 30 min. after glucose load. B. coccineus administered acutely did not generally produce significant reduction in blood glucose level in diabetic rats. Administered subacutely, the extract significantly reduced blood glucose level in diabetic rats from the 3rdday with peak effect observed at the dose of 800 mg·kg–1on the 10thday. The extract generally preserved in vivo antioxidant levels in the kidneys, heart, and liver, increased the level of high density lipoprotein and insulin, and reduced the level of triglycerides and low density lipoprotein compared to diabetic control.
