1Stripe Rust Effector PstGSRE1 Disrupts Nuclear Localization of ROS-Promoting Transcription Factor TaLOL2 to Defeat ROS-Induced Defense in Wheat显示文摘Puccinia striiformis f.sp.tritici(Pst),a biotrophic plant pathogen,secretes numerous effectors to modulate host defense systems.Understanding the molecular mechanisms by which Pst effectors regulate wheat immunity is of great importance for the development of novel strategies for durable control of stripe rust.In this study,we identified a glycine-serine-rich effector gene,PstGSRE1,which is highly induced dur-ing early infection.Transgenic expression of PstGSRE1 RNAi constructs in wheat significantly reduced virulence of Pst and increased H2O2 accumulation in wheat.PstGSRE1 was shown to target the reactive ox-ygen species(ROS)-associated transcription factor TaLOL2,a positive regulator of wheat immunity.PstGSRE1 disrupted nuclear localization of TaLOL2 and suppressed ROS-mediated cell death induced by TaLOL2,thus compromising host immunity.This work reveals a previously unrecognized strategy whereby rust fungi exploit the PstGSRE1 effector to defeat ROS-associated plant defense by modulating the subcellular compartment of a host immune regulator and facilitate pathogen infection.Tuo Qi Jia Guo Peng Liu Fuxin He Cuiping Wan Md Ashraful Islam Brett M.Tyler Zhensheng Kang Jun Guo 2019Molecular Plant2019,12,12:11
2Tailored PEDOT:PSS hole transport layer for higher performance in perovskite solar cells: Enhancement of electrical and optical properties with improved morphology显示文摘Precise control over the charge carrier dynamics throughout the device can result in outstanding performance of perovskite solar cells(PSCs).Poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate)(PEDOT:PSS)is the most actively studied hole transport material in p-i-n structured PSCs.However,charge transport in the PEDOT:PSS is limited and inefficient because of its low conductivity with the presence of the weak ionic conductor PSS.In addition,morphology of the underlying PEDOT:PSS layer in PSCs plays a crucial role in determining the optoelectronic quality of the active perovskite absorber layer.This work is focused on realization of a non-wetting conductive surface of hole transport layer suitable for the growth of larger perovskite crystalline domains.This is accomplished by employing a facile solventengineered(ethylene glycol and methanol)approach resulting in removal of the predominant PSS in PEDOT:PSS.The consequence of acquiring larger perovskite crystalline domains was observed in the charge carrier dynamics studies,with the achievement of higher charge carrier lifetime,lower charge transport time and lower transfer impedance in the solvent-engineered PEDOT:PSS-based PSCs.Use of this solventengineered treatment for the fabrication of MAPbI3 PSCs greatly increased the device stability witnessing a power conversion efficiency of 18.18%,which corresponds to^37%improvement compared to the untreated PEDOT:PSS based devices.Khan Mamun Reza Ashim Gurung Behzad Bahrami Sally Mabrouk Hytham Elbohy Rajesh Pathak Ke Chen Ashraful Haider Chowdhury Md Tawabur Rahman Steven Letourneau Hao-Cheng Yang Gopalan Saianand Jeffrey WElam Seth BDarling Qiquan Qiao 2020Journal of Energy Chemistry2020,29,5:4
3Proposal for dispersion compensating square-lattice photonic crystal fiber显示文摘A dispersion-compensating square-lattice photonic crystal fiber for broadband compensation which covers the S, C and L communication bands, i.e. wavelength ranging from 1 460 nm to 1 625 nm is proposed in this paper. Theoretically, it is shown a negative dispersion coefficient of about-595 ps/(nm·km) to-1 288 ps/(nm·km) over S to L bands and-975 ps/(nm·km) at the operating wavelength 1 550 nm. The relative dispersion slope is perfectly matched to that of conventional single-mode fiber of about 0.003 6 nm^(-1). Besides the proposed photonic crystal fiber shows the large nonlinear coefficient of 61.88 W/km at the operating wavelength of 1 550 nm. 18A Simple,Efficient and Rapid Method for Good Quality DNA Extraction from Rice Grains显示文摘An efficient and good DNA extraction protocol should be simple, affordable and yield enough DNA with high quality. Rice(Oryza sativa L.) DNA extraction methods often use seedlings or leaves rather than the grains and tend to be time-consuming, involve multiple steps, and use hazardous chemicals and expensive enzymes. Rice grains offer several benefits over seedlings and leaves as a source of DNA for genetic analysis. However, these benefits are underutilized because the bulk of a rice grain is made up of starch. It is particularly important, but difficult to get rid of the starch while extracting DNA from rice grains. This co-precipitated polysaccharide is a known inhibitor of DNA polymerase activity in polymerase chain reaction(PCR). We describe here a very simple and highly affordable Chelex~?-100 based DNA extraction method from rice grains. It does not require any hazardous chemicals or enzymes. 20Identification of visible colored dissolved organic matter in biological and tertiary municipal effluents using multiple approaches including PARAFAC analysis显示文摘This study provided insights into the persistent yellowish color in biological and tertiary effluents of municipal wastewater through a multi-characterization approach and fluorescence excitation-emission matrix-parallel factor(EEM-PARAFAC)analysis.The characterization was performed on three to five full-scale municipal wastewater treatment plants(WWTPs),including differential log-transformed absorbance(DLn A)spectroscopy,resin fractionation,size-exclusion chromatography for apparent molecular weight analysis(SECAMW),and X-ray photoelectron spectroscopy(XPS)analysis.Hydrophobic acids(HPOA)were abundant in visible colored dissolved organic matter(DOM).The SEC-AMW result showed that the molecular weight of the colored substances in the secondary effluents is mainly distributed in the range of 2–3 k Da.Through XPS analysis,C-O/C-N and pyrrolic/pyridonic(N-5)were found to be positively correlated with chroma.PARAFAC component models were built on biological(two components)and tertiary effluent(three components)and the correlation analysis revealed that PARAFAC component 2 in biological effluent(BE-C2)and component 1in tertiary effluent(TE-C1),which were ascribed to Hydrophobic acids and Humic acid-like,were the responsible visible colored DOM components cause yellowish color.In addition,component similarity testing found that the identified visible colored DOM PARAFAC BE-C2,and PARAFAC TE-C1 were identical(0.96)in physicochemical properties,with 4%removal efficacy on average,compared with 11%for invisible colored DOM.This implied that tertiary effluents containing colorants(TE-C1)were resistant to degradation/removal using different disinfection and filtration processes in advanced treatments.This sheds light on many physicochemical aspects of PARAFAC-identified visible colored DOM components and provides spectral data to build an online monitoring system.Ashraful Islam Guangxi Sun Ayman NSaber Wei Shang Xingcan Zheng Yu Zhang Min Yang 2022Journal of Environmental Sciences2022,34,12:1
