| 2 | Tom20 senses iron-activated ROS signaling to promote melanoma cell pyroptosis显示文摘Iron has been shown to trigger oxidative stress by elevating reactive oxygen species (ROS) and to participate in different modes of cell death,such as ferroptosis,apoptosis and necroptosis. However,whether iron-elevated ROS is also linked to pyroptosis has not been reported. Here,we demonstrate that iron-activated ROS can induce pyroptosis via a Tom20-Bax-caspase-GSDME pathway. In melanoma cells,iron enhanced ROS signaling initiated by CCCP,causing the oxidation and oligomerization of the mitochondrial outer membrane protein Tom20. Bax is recruited to mitochondria by oxidized Tom20,which facilitates cytochrome c release to cytosol to activate caspase-3;eventually triggering pyroptotic death by inducing GSDME cleavage. Therefore,ROS acts as a causative factor and Tom20 senses ROS signaling for iron-driven pyroptotic death of melanoma cells. Since iron activates ROS for GSDME-dependent pyroptosis induction and melanoma cells specifically express a high level of GSDME,iron may be a potential candidate for melanoma therapy. Based on the functional mechanism of iron shown above,we further demonstrate that iron supplementation at a dosage used in iron-deficient patients is sufficient to maximize the anti-tumor effect of clinical ROS-inducing drugs to inhibit xenograft tumor growth and metastasis of melanoma cells through GSDME-dependent pyroptosis. Moreover,no obvious side effects are observed in the normal tissues and organs of mice during the combined treatment of clinical drugs and iron. | 4 | Stationarity Intervals of Time-Variant Channel in High Speed Railway Scenario显示文摘The non-stationary behavior, caused by the train movement, is the main factor for the variation of high speed railway channel. To measure the time-variant effect, the parameter of stationarity interval, in which the channel keeps constant or has no great change, is adopted based on Zhengzhou-Xi'an (Zhengxi) passenger dedicated line measurement with different train speeds. The stationarity interval is calculated through the definition of Local Region of Stationarity (LRS) under three train velocities. Furthermore, the time non-stationary characteristic of high speed railway channel is compared with five standard Multiple-Input Multiple-Output (MIMO) channel models, i.e. Spatial Channel Model (SCM), extended version of SCM (SCME), Wireless World Initiative New Radio Phase II (WINNERII), International Mobile Telecommunications-Advanced (IMT-Advanced) and WiMAX models which contain the high speed moving scenario. The stationarity interval of real channel is 9 ms in 80% of the cases, which is shorter than those of the standard models. Hence the real channel of high speed railway changes more rapidly. The stationarity intervals of standard models are different due to different modeling methods and scenario definitions. The corresponding sum-of-sinusoids(SoS) simulation models including both the deterministic model and statistical model with finite scatterers are also proposed for practicable implementation. The simulation results illustrate that the simulation models well reproduce the channel characteristics of the single sphere reference model with sufficient simulation scatterers. And the statistical model has a better approximation of the reference model in comparison with the deterministic one when the simulation trials of the stochastic model are sufficient. | 9 | Differentially expressed proteins of gamma-ray irradiated mouse intestinal epithelial cells by two-dimensional electrophoresis and MALDI-TOF mass spectrometry显示文摘AIM: To identify the differentially expressed proteins involved in ionizing radiation in mice and to explore new ways for studying radiation-related proteins.METHODS: Bal B/c mice grouped as sham-irradiation, 3h and 72 h irradiation were exposed to 9.0Gy single dose of γ-irradiation. Intestinal epithelia were isolated from mice,and total proteins were extracted with urea containing solution. A series of methods were used, including twodimensional electrophoresis, PDQuest 2-DE software analysis, peptide mass fingerprinting based on matrixassisted laser desorption/ionization time of flight masss pectrometry (MALDI-TOF-MS) and SWISS-PROT database searching, to separate and identify the differential proteins.Western blotting and RT-PCR were used to validate the differentially expressed proteins.RESULTS: Mouse intestine was severely damaged by 9.0 Gy γ-irradiation. Image analysis of two-dimensional gels revealed that averages of 638±39, 566±32 and 591±29 protein spots were detected in 3 groups, respectively, and the majority of these protein spots were matched. About 360 protein spots were matched between normal group and 3 h irradiation group, and the correlation coefficient was 0.78 by correlation analysis of gels. Also 312 protein spots matched between normal group and 72 h irradiation group, and 282 protein spots between 3 h and 72 h irradiation groups. Twenty-eight differential protein spots were isolated from gels, digested with trypsin, and measured with MALDI-TOF-MS. A total of 25 spots yielded good spectra, and 19 spots matched known proteins after database searching. These proteins were mainly involved in anti-oxidation, metabolism, signal transduction,and protein post-translational processes. Western-blotting confirmed that enolase was up-regulated by γ-irradiation. Up-regulation of peroxiredoxin I was verified by applying RTPCR technique, but no change occurred in QSVC72.CONCLUSION: These differentially expressed proteins might play important roles when mouse intestine was severely injured by γ-irradiation. | 11 | Effects of targeting magnetic drug nanoparticles on human cholangiocarcinoma xenografts in nude mice显示文摘BACKGROUND:Targeting is a new therapeutic tool for malignant tumor as a result of combining nanotechnology with chemotherapeutics.The aim of our study was to investigate the effects of magnetic nanoparticles enveloping a chemotherapeutic drug on human cholangiocarcinoma xenografts in nude mice. METHODS:The human cholangiocarcinoma xenograft model was established in nude mice with the QBC939 cell line.The nude mice were randomly assigned to 7 groups. 0.9%saline or magnetic nanoparticles,including high (group 2),medium(group 4)and low(group 5)dosages, were given to nude mice through the tail vein 20 days after the QBC939 cell line was implanted.Calculations were made 35 days after treatment in order to compare the volumes,inhibition ratios and growth curves of the tumors in each group.Mice in each group were sacrificed randomly to collect tumor tissues and other organs for electron microscopy and pathological examination. RESULTS:The high and medium dosage groups were significantly different from the control group(P<0.05). The tumor inhibition ratios for the high,medium and low dosage groups were 39.6%,14.6%and 7.9%,respectively. | 12 | Association between Plasma Metal Levels and Diabetes Risk: a Case-control Study in China显示文摘Objective Many metals, some of which have been classified as environmental endocrine disruptors, are used extensively in everyday consumer products and are ubiquitous in our living environment. In the present study, we aimed to explore the associations between the prevalence risk of type 2 diabetes and plasma levels of 20 trace elements as well as those of heavy metals in a Han Chinese population. Methods We conducted a case-control study to investigate the associations between plasma concentrations of 20 metals and diabetes in Jiangsu province. A total of 122 newly diagnosed cases of type 2 diabetes and 429 matched controls were recruited from community physical examinations in Suzhou City of Jiangsu Province. Plasma metal levels were measured by inductively-coupled plasma mass spectrometry. Results After adjusting for confounders, plasma vanadium, chromium, manganese, copper, zinc, arsenic, selenium, strontium, palladium, cadmium, cesium, and barium were associated with diabetes risk(P < 0.05). The adjusted OR increased with increasing concentration of vanadium, manganese, copper, zinc, and cesium. Conclusion Many metals, including manganese, copper, zinc, arsenic, selenium, and cadmium in plasma, are associated with the morbidity of diabetes. Thescreening protocol for bacterial infection consisted ofchest radiograph; blood, urine and ascitic fluid cul-tures; the severity of liver disease shown by Child-Pugh score.Results: Active bleeding was observed at endoscopyin 40 patients (53%). Failure to control bleedingWithin 5 days occurred in 36 patients (45%). Empir-ical antibiotic treatment was used in 53 patients(67%), whereas bacterial infections were documen-ted in 43 patients (57%). | 17 | Thermal performance analysis of non-uniform height rectangular fin based on constructal theory and entransy theory显示文摘A model of non-uniform height rectangular fin, in which the variation of base's thickness and width are taken into account, is established in this paper. The dimensionless maximum thermal resistance(DMTR) and the dimensionless equivalent thermal resistance(DETR) defined based on the entransy dissipation rate(EDR) are taken as performance evaluation indexes. According to constructal theory, the variations of the two indexes with the geometric parameters of the fin are analyzed by using a finite-volume computational fluid dynamics code, the effects of the fin-material fraction on the two indexes are analyzed. It is found that the two indexes decrease monotonically as the ratio between the front height and the back height of the fin increases subjected to the non-uniform height rectangular fin. When the model is reduced to the uniform height fin, the two indexes increase first and then decrease with increase in the ratio between the height of the fin and the fin space. The fin-material fraction has no effect on the change rule of the two indexes with the ratio between the height of the fin and the fin space. The sensitivity of the DETR to the geometric parameters of the fin is higher than that of the DMTR to the geometric parameters.
