| 2 | Reproduction and beyond,kisspeptin in ruminants显示文摘Kisspeptin(Kp) is synthesized in the arcuate nucleus and preoptic area of the hypothalamus and is a regulator of gonadotropin releasing hormone in the hypothalamus.In addition,Kp may regulate additional functions such as increased neuropeptide Y gene expression and reduced proopiomelanocortin(POMC) gene expression in sheep.Other studies have found a role for Kp to release growth hormone(GH),prolactin and luteinizing hormone(LH)from cattle,rat and monkey pituitary cells.Intravenous injection of Kp stimulated release LH,GH,prolactin and follicle stimulating hormone in some experiments in cattle and sheep,but other studies have failed to find an effect of peripheral injection of Kp on GH release.Recent studies indicate that Kp can stimulate GH release after intracerebroventricular injection in sheep at doses that do not release GH after intravenous injection.These studies suggest that Kp may have a role in regulation of both reproduction and metabolism in sheep.Since GH plays a role in luteal development,it is tempting to speculate that the ability of Kp to release GH and LH is related to normal control of reproduction. | Joseph A.Daniel Chad D.Foradori Brian K.Whitlock James L.Sartin | 2015 | Journal of Animal Science and Biotechnology2015,6,3: | 0 |
| 3 | CaMK4 overexpression in polycystic kidney disease promotes mTOR-mediated cell proliferation显示文摘Autosomal dominant polycystic kidney disease(ADPKD)is characterized by progressive enlargement of fluid-filled cysts,causing nephron loss and a decline in renal function.Mammalian target of rapamycin(mTOR)is overactive in cyst-lining cells and contributes to abnormal cell proliferation and cyst enlargement;however,the mechanism for mTOR stimulation remains unclear.We discovered that calcium/calmodulin(CaM)dependent kinase IV(CaMK4),a multifunctional kinase,is overexpressed in the kidneys of ADPKD patients and PKD mouse models.In human ADPKD cells,CaMK4 knockdown reduced mTOR abundance and the phosphorylation of ribosomal protein S6 kinase(S6K),a downstream target of mTOR.Pharmacologic inhibition of CaMK4 with KN-93 reduced phosphorylated S6K and S6 levels and inhibited cell proliferation and in vitro cyst formation of ADPKD cells.Moreover,inhibition of calcium/CaM-dependent protein kinase kinase-βand CaM,two key upstream regulators of CaMK4,also decreased mTOR signaling.The effects of KN-93 were independent of the liver kinase B1-adenosine monophosphate-activated protein kinase(AMPK)pathway,and the combination of KN-93 and metformin,an AMPK activator,had additive inhibitory effects on mTOR signaling and in vitro cyst growth.Our data suggest that increased CaMK4 expression and activity contribute to mTOR signaling and the proliferation of cystic cells of ADPKD kidneys. | Yan Zhang Emily A.Daniel July Metcalf Yuqiao Dai Gail A.Reif Darren P.Wallace | 2022 | Journal of Molecular Cell Biology2022,14,7: | 0 |

| 6 | Kernel-Based State-Space Kriging for Predictive Control显示文摘In this paper, we extend the state-space kriging(SSK) modeling technique presented in a previous work by the authors in order to consider non-autonomous systems. SSK is a data-driven method that computes predictions as linear combinations of past outputs. To model the nonlinear dynamics of the system, we propose the kernel-based state-space kriging(K-SSK), a new version of the SSK where kernel functions are used instead of resorting to considerations about the locality of the data. Also, a Kalman filter can be used to improve the predictions at each time step in the case of noisy measurements. A constrained tracking nonlinear model predictive control(NMPC) scheme using the black-box input-output model obtained by means of the K-SSK prediction method is proposed.
