1Feasibility of endoscopic treatment and predictors of lymph node metastasis in early gastric cancer显示文摘BACKGROUND Endoscopic submucosal dissection (ESD) has been routinely performed in applicable early gastric cancer (EGC) patients as an alternative to conventional surgical operations that involve lymph node dissection. The indications for ESD have been recently expanded to include larger, ulcerated, and undifferentiated mucosal lesions, and differentiated lesions with slight submucosal invasion. The risk of lymph node metastasis (LNM) is the most important consideration when deciding on a treatment strategy for EGC. Despite the advantages over surgical procedures, lymph nodes cannot be removed by ESD. In addition, whether patients who meet the expanded indications for ESD can be managed safely remains controversial. AIM To determine whether the ESD indications are applicable to Chinese patients and to investigate the predictors of LNM in EGC. METHODS We retrospectively analyzed 12552 patients who underwent surgery for gastric cancer between June 2007 and December 2018 at the Affiliated Hospital of Qingdao University. A total of 1262 (10.1%) EGC patients were eligible for inclusion in this study. Data on the patients’ clinical, endoscopic, and histopathological characteristics were collected. The absolute and expanded indications for ESD were validated by regrouping the enrolled patients and determining the positive LNM results in each subgroup. Predictors of LNM in patients were evaluated by univariate and multivariate analyses. RESULTS LNM was observed in 182 (14.4%) patients. No LNM was detected in the patients who met the absolute indications (0/90). LNM occurred in 4/311 (1.3%) patients who met the expanded indications. According to univariate analysis, LNM was significantly associated with positive tumor marker status, medium (20-30 mm) and large (>30 mm) lesion sizes, excavated macroscopic-type tumors, ulcer presence, submucosal invasion (SM1 and SM2), poor differentiation, lymphovascular invasion (LVI), perineural invasion, and diffuse and mixed Lauren’s types. Multivariate analysis demonstrated SM1 invasion (odds ration [OR]= 2.285, P = 0.03), SM2 invasion (OR = 3.230, P < 0.001), LVI (OR = 15.702, P < 0.001), mucinous adenocarcinoma (OR = 2.823, P = 0.015), and large lesion size (OR = 1.900, P = 0.006) to be independent risk factors. CONCLUSION The absolute indications for ESD are reasonable, and the feasibility of expanding the indications for ESD requires further investigation. 3Expansion of the hepatocellular carcinoma Milan criteria in liver transplantation: Future directions显示文摘Milan criteria are currently the benchmark related to liver transplantation(LT) for hepatocellular carcinoma. However, several groups have proposed different expanded criteria with acceptable results. In this article, we review the current status of LT beyond the Milan criteria in three different scenarios-expanded criteria with cadaveric LT, downstaging to Milan criteria before LT, and expansion in the context of adult living donor LT. 4Stent-grafts for the treatment of TIPS dysfunction:Fluency stent vs Wallgraft stent显示文摘AIM:To evaluate the clinical efficacy of an expanded polytetrafluoro-ethylene-covered Fluency stent compared with that of a polyethylene terephthalate-covered Wallgraft stent for the management of transjugular intrahepatic portosystemic shunt(TIPS)dysfunction.METHODS:A retrospective review of patients who underwent TIPS revision with stent-grafts between May 2007 and June 2011 was conducted.The patients were divided into two groups according to the stentgrafts implanted:the Fluency stent(Bard Incorporated,Karlsruhe,Germany)and the Wallgraft stent(Boston Scientific,Galway,Ireland).The primary patency rates were calculated and compared using the Kaplan-Meier method.RESULTS:A total of 73 patients were evaluated in this study:33 with Fluency stents and 40 with Wallgraft stents.The primary patency rates at 12 and 24 mo were 91% and 85%,respectively,in the Fluency stent group and 78% and 63%,respectively,in the Wallgraft stent group.The primary shunt patency rates after TIPS revision were significantly better with the Fluency stent than with the Wallgraft stent(P = 0.033).CONCLUSION:TIPS revision with the Fluency stent has higher medium-term patency rates than that with the Wallgraft stent.Xue-Feng Luo Ling Nie Zhu Wang Jiaywei Tsauo Ling-Jun Liu Yang Yu Biao Zhou Cheng-Wei Tang Xiao Li 2013World Journal of Gastroenterology2013,19,30:9
5The apricot(Prunus armeniaca L.)genome elucidates Rosaceae evolution and beta-carotenoid synthesis显示文摘Apricots,scientifically known as Prunus armeniaca L,are drupes that resemble and are closely related to peaches or plums.As one of the top consumed fruits,apricots are widely grown worldwide except in Antarctica.A high-quality reference genome for apricot is still unavailable,which has become a handicap that has dramatically limited the elucidation of the associations of phenotypes with the genetic background,evolutionary diversity,and population diversity in apricot.DNA from P.armeniaca was used to generate a standard,size-selected library with an average DNA fragment size of~20 kb.The library was run on Sequel SMRT Cells,generating a total of 16.54 Gb of PacBio subreads(N50=13.55 kb).The high-quality P.armeniaca reference genome presented here was assembled using long-read single-molecule sequencing at approximately 70×coverage and 171×Illumina reads(40.46 Gb),combined with a genetic map for chromosome scaffolding.The assembled genome size was 221.9 Mb,with a contig NG50 size of 1.02 Mb.Scaffolds covering 92.88%of the assembled genome were anchored on eight chromosomes.Benchmarking Universal Single-Copy Orthologs analysis showed 98.0%complete genes.We predicted 30,436 protein-coding genes,and 38.28%of the genome was predicted to be repetitive.We found 981 contracted gene families,1324 expanded gene families and 2300 apricot-specific genes.The differentially expressed gene(DEG)analysis indicated that a change in the expression of the 9-cis-epoxycarotenoid dioxygenase(NCED)gene but not lycopene beta-cyclase(LcyB)gene results in a lowβ-carotenoid content in the white cultivar“Dabaixing”.This complete and highly contiguous P.armeniaca reference genome will be of help for future studies of resistance to plum pox virus(PPV)and the identification and characterization of important agronomic genes and breeding strategies in apricot.Fengchao Jiang Junhuan Zhang Sen Wang Li Yang Yingfeng Luo Shenghan Gao Meiling Zhang Shuangyang Wu Songnian Hu Haoyuan Sun Yuzhu Wang 2019Horticulture Research2019,6,1:9
6A chromosome-scale genome assembly of Isatis indigotica,an important medicinal plant used in traditional Chinese medicine显示文摘Isatis indigotica(2n=14)is an important medicinal plant in China.Its dried leaves and roots(called Isatidis Folium and Isatidis Radix,respectively)are broadly used in traditional Chinese medicine for curing diseases caused by bacteria and viruses such as influenza and viral pneumonia.Various classes of compounds isolated from this species have been identified as effective ingredients.Previous studies based on transcriptomes revealed only a few candidate genes for the biosynthesis of these active compounds in this medicinal plant.Here,we report a high-quality chromosome-scale genome assembly of I.indigotica with a total size of 293.88 Mb and scaffold N50=36.16 Mb using single-molecule real-time long reads and high-throughput chromosome conformation capture techniques.We annotated 30,323 highconfidence protein-coding genes.Based on homolog searching and functional annotations,we identified many candidate genes involved in the biosynthesis of main active components such as indoles,terpenoids,and phenylpropanoids.In addition,we found that some key enzyme-coding gene families related to the biosynthesis of these components were expanded due to tandem duplications,which likely drove the production of these major active compounds and explained why I.indigotica has excellent antibacterial and antiviral activities.Our results highlighted the importance of genome sequencing in identifying candidate genes for metabolite synthesis in medicinal plants.Minghui Kang Haolin Wu Qiao Yang Li Huang Quanjun Hu Tao Ma Zaiyun Li Jianquan Liu 2020Horticulture Research2020,7,1:9

8Measurement of the echo reduction for underwater acoustic passive materials by using the time reversal technique显示文摘A measuring method of the echo reduction of passive materials by using the time reversal(TR) technique is presented. To measure the echo reduction of a sample with this approach, the received signals are firstly focused according to the TR theory. Then, the sample is removed and the TR processing is again employed to realize the focus of the received signal.Finally, the echo reduction of the sample is evaluated with these focusing signals. Besides, to calibrate the measured echo reduction via the TR technique, a standard sample is employed to measure a constant coefficient that only depends on the measurement environment. An aluminum plate sample and a steel plate sample with the same size of 1.1 mxl.O m x0.005 m axe tested in a wave guide tank. The experimental results show that the calibrated values are well consistent with theoretical results under the free field at the measured frequency range of0.5-20 kHz. The relative errors of all the measured values are less than 10% and the values of the expanded uncertainty are less than 1.5 dB. 9Prolonged overall survival in gastric cancer patients after adoptive immunotherapy显示文摘AIM:To assess the efficacy of immunotherapy with expanded activated autologous lymphocytes(EAALs) in gastric cancer.METHODS:An observational study was designed to retrospectively analyze the clinical data of 84 gastric cancer patients,of whom 42 were treated by EAAL immunotherapy plus conventional treatment and another 42 only received conventional treatment(control group).EAALs were obtained by proliferation of peripheral blood mononuclear cells from patients followed by phenotype determination.Clinical data including age,gender,clinical stage,chemotherapeutic regimens,hospitalization,surgical,radiotherapy,and survival data were collected along with EAAL therapy details and side effects.Patients were followed and the relationship between treatment and overall survival(OS) data obtained for the immunotherapy and control groups were compared retrospectively.The safety of EAAL immunotherapy was also evaluated.RESULTS:After in vitro culture and proliferation,the percentages of CD3+,CD3+CD8+,CD8+CD27+,CD8+CD28+,and CD3+CD16+/CD56+cells increased remarkably(P < 0.05),while the percentages of CD3+CD4+,CD4+CD25+,and CD3-CD16+/CD56+(natural killer cells) were overtly decreased(P < 0.05); no significant change was observed in CD4+CD25+CD127- cells(P =0.448).Interestingly,OS in the immunotherapy group was significantly higher than that in the control group,with 27.0 and 13.9 mo obtained for the two groups,respectively(P =0.028,HR =0.573,95%CI:0.347-0.945).These findings indicated a 42.7% decrease in the risk of death.In addition,we found that clinical stage and application of EAAL immunotherapy wereindependent prognostic factors for gastric cancer patients.Indeed,the OS in stage Ⅲc and Ⅳ patients that had received surgery was prolonged after EAAL immunotherapy(P < 0.05).Importantly,in vitro induction and proliferation of EAAL were easy and biologically safe.CONCLUSION:Overall,EAAL adoptive immunotherapy might prolong the OS in gastric cancer patients.Guo-Qing Zhang Hong Zhao Jian-Yu Wu Jin-Yu Li Xiang Yan Gang Wang Liang-Liang Wu Xiao-Gang Zhang Yi Shao Yu Wang Shun-Chang Jiao 2015World Journal of Gastroenterology2015,21,9:6
10Functional analysis of a novel C-glycosyltransferase in the orchid Dendrobium catenatum显示文摘Flavonoids,which are a diverse class of phytonutrients,are used by organisms to respond to nearly all abiotic stresses and are beneficial for human health.Glycosyltransferase,used during the last step of flavonoid biosynthesis,is important in flavonoid enrichment.However,little is known about glycosyltransferase in the orchid Dendrobium catenatum(D.officinale).In this study,we isolated a novel C-glycosyltransferase(designated DcaCGT)from the orchid D.catenatum by identifying and analyzing 82 putative genes in the GT1 family.DcaCGT could specifically catalyze not only di-C-glycosylation but also O-glycosylation.Apart from the normal function of catalyzing 2-hydroxynaringenin and phloretin to the respective di-C-glycosides,DcaCGT also catalyzes apigenin to cosmosiin.Targeted metabolic profiling of the substrates(2-hydroxynaringenin,phloretin,and apigenin)and products(vitexin,isovitexin,vicenin-2,nothofagin,3’,5’-di-C-glucosylphloretin,and cosmosiin)in different tissues showed that vicenin-2 was the most abundant product of this novel enzyme.Cosmosiin was detected in flowers and flower buds.We also established that DcaCGT functions expanded throughout the evolution of D.catenatum.Residual OGT activity may help D.catenatum resist drought stress.Our study illustrates the function,origin,and differentiation of DcaCGT and provides insights into glycosylation and molecular propagation processes,which can be used to improve the production of flavonoids by the cultivated medicinal plant D.catenatum.Zhiyao Ren Xiaoyu Ji Zhenbin Jiao Yingyi Luo Guo-Qiang Zhang Shengchang Tao Zhouxi Lei Jing Zhang Yuchen Wang Zhong-Jian Liu Gang Wei 2020Horticulture Research2020,7,1:5
11Insights into triterpene synthesis and unsaturated fatty-acid accumulation provided by chromosomallevel genome analysis of Akebia trifoliata subsp . australis显示文摘Akebia trifoliata subsp.australis is a well-known medicinal and potential woody oil plant in China.The limited genetic information available for A.trifoliata subsp.australis has hindered its exploitation.Here,a high-quality chromosomelevel genome sequence of A.trifoliata subsp.australis is reported.The de novo genome assembly of 682.14 Mb was generated with a scaffold N50 of 43.11 Mb.The genome includes 25,598 protein-coding genes,and 71.18%(485.55 Mb)of the assembled sequences were identi fi ed as repetitive sequences.An ongoing massive burst of long terminal repeat(LTR)insertions,which occurred~1.0 million years ago,has contributed a large proportion of LTRs in the genome of A.trifoliata subsp.australis.Phylogenetic analysis shows that A.trifoliata subsp.australis is closely related to Aquilegia coerulea and forms a clade with Papaver somniferum and Nelumbo nucifera,which supports the well-established hypothesis of a close relationship between basal eudicot species.The expansion of UDP-glucoronosyl and UDP-glucosyl transferase gene families and fi-amyrin synthase-like genes and the exclusive contraction of terpene synthase gene families may be responsible for the abundant oleanane-type triterpenoids in A.trifoliata subsp.australis.Furthermore,the acyl-ACP desaturase gene family,including 12 stearoyl-acyl-carrier protein desaturase(SAD)genes,has expanded exclusively.A combined transcriptome and fatty-acid analysis of seeds at fi ve developmental stages revealed that homologs of SADs,acyl-lipid desaturase omega fatty acid desaturases(FADs),and oleosins were highly expressed,consistent with the rapid increase in the content of fatty acids,especially unsaturated fatty acids.The genomic sequences of A.trifoliata subsp.australis will be a valuable resource for comparative genomic analyses and molecular breeding.Hui Huang Juan Liang Qi Tan Linfeng Ou Xiaolin Li Caihong Zhong Huilin Huang Ian Max Moller Xianjin Wu Songquan Song 2021Horticulture Research2021,8,1:5

13The chromosome-based lavender genome provides new in sights into Lamiaceae evolution and terpenoid biosynthesis显示文摘The aromatic shrub Lavandula angustifolia produces various volatile terpenoids that serve as resources for essential oils and function in plant-insect communication.To better understand the genetic basis of the terpenoid diversity in lavender,we present a high-quality reference genome for the Chinese lavender cultivar‘Jingxun 2'using PacBio and Hi-C technologies to anchor the 894.50 Mb genome assembly into 27 pseudochromosomes.In addition to the y triplication event,lavender underwent two rounds of whole-genome duplication(WGD)during the Eocene-Oligocene(29.6 MYA)and Miocene-Pliocene(6.9 MYA)transitions.As a result of tandem duplications and lineage-specific WGDs,gene families related to terpenoid biosynthesis in lavender are substantially expanded compared to those of five other species in Lamiaceae.Many terpenoid biosynthesis transcripts are abundant in glandular trichomes.We further integrated the contents of ecologically functional terpenoids and coexpressed terpenoid biosynthetic genes to construct terpenoid-gene networks.Typical gene clusters,including TPS-TPS,TPS-CYP450,and TPS-BAHD,linked with compounds that primarily function as attractants or repellents,were identified by their similar patterns of change during flower development or in response to methyl jasmonate.Comprehensive analysis of the genetic basis of the production of volatiles in lavender could serve as a foundation for future research into lavender evolution,phytochemistry,and ecology.Jingrui Li Yiming Wang Yanmei Dong Wenying Zhang Di Wang Hongtong Bai Kui Li Hui Li Lei Shi 2021Horticulture Research2021,8,1:5
14A high-quality genome assembly of Morinda officinalis,a famous native southern herb in the Lingnan region of southern China显示文摘Morinda officinalis is a well-known medicinal and edible plant that is widely cultivated in the Lingnan region of southern China.Its dried roots(called bajitian in traditional Chinese medicine)are broadly used to treat various diseases,such as impotence and rheumatism.Here,we report a high-quality chromosome-scale genome assembly of M.officinalis using Nanopore single-molecule sequencing and Hi-C technology.The assembled genome size was 484.85Mb with a scaffold N50 of 40.97 Mb,and 90.77%of the assembled sequences were anchored on eleven pseudochromosomes.The genome includes 27,698 protein-coding genes,and most of the assemblies are repetitive sequences.Genome evolution analysis revealed that M.officinalis underwent core eudicotγgenome triplication events but no recent whole-genome duplication(WGD).Likewise,comparative genomic analysis showed no large-scale structural variation after species divergence between M.officinalis and Coffea canephora.Moreover,gene family analysis indicated that gene families associated with plant–pathogen interactions and sugar metabolism were significantly expanded in M.officinalis.Furthermore,we identified many candidate genes involved in the biosynthesis of major active components such as anthraquinones,iridoids and polysaccharides.In addition,we also found that the DHQS,GGPPS,TPS-Clin,TPS04,sacA,and UGDH gene families—which include the critical genes for active component biosynthesis—were expanded in M.officinalis.This study provides a valuable resource for understanding M.officinalis genome evolution and active component biosynthesis.This work will facilitate genetic improvement and molecular breeding of this commercially important plant.Jihua Wang Shiqiang Xu Yu Mei Shike Cai Yan Gu Minyang Sun Zhan Liang Yong Xiao Muqing Zhang Shaohai Yang 2021Horticulture Research2021,8,1:4
15Burgeoning study of sentinel-node analysis on management of early gastric cancer after endoscopic submucosal dissection显示文摘Endoscopic submucosal dissection(ESD)represents an organ-preserving alternative to surgical resection of early gastric cancer.However,even with ESD yielding en-bloc resection specimens,there are concerns regarding tumor spread such as with larger lesions,ulcerated lesions,undifferentiated pathology and submucosal invasion.Sentinel node navigational surgery(SNNS)when combined with ESD offers a minimally invasive alternative to the traditional extended gastrectomy and lymphadenectomy if lack of lymph node spread can be confirmed.This would have a clear advantage in terms of potential complications and quality of life.However,SNNS,though useful in other malignancies such as breast cancer and melanoma,may not have a sufficient sensitivity for malignancy and negative predictive value in EGC to justify this as standard practice after ESD.The results of SNNS may improve with greater standardization and more involved dissection,technological innovations and more experience and validation such that the paradigm for post-ESD resection of EGC may change and include SNNS.David Friedel Xiaocen Zhang Stavros Nicholas Stavropoulos 2020World Journal of Gastrointestinal Endoscopy2020,12,4:3
16The genome of Chinese flowering cherry(Cerasus serrulata)provides new insights into Cerasus species显示文摘Cerasus serrulata is a flowering cherry germplasm resource for ornamental purposes.In this work,we present a de novo chromosome-scale genome assembly of C.serrulata by the use of Nanopore and Hi-C sequencing technologies.The assembled C.serrulata genome is 265.40 Mb across 304 contigs and 67 scaffolds,with a contig N50 of 1.56 Mb and a scaffold N50 of 31.12 Mb.It contains 29,094 coding genes,27,611(94.90%)of which are annotated in at least one functional database.Synteny analysis indicated that C.serrulata and C.avium have 333 syntenic blocks composed of 14,072 genes.Blocks on chromosome 01 of C.serrulata are distributed on all chromosomes of C.avium,implying that chromosome 01 is the most ancient or active of the chromosomes.The comparative genomic analysis confirmed that C.serrulata has 740 expanded gene families,1031 contracted gene families,and 228 rapidly evolving gene families.By the use of 656 single-copy orthologs,a phylogenetic tree composed of 10 species was constructed.The present C.serrulata species diverged from Prunus yedoensis~17.34 million years ago(Mya),while the divergence of C.serrulata and C.avium was estimated to have occurred∼21.44 Mya.In addition,a total of 148 MADS-box family gene members were identified in C.serrulata,accompanying the loss of the AGL32 subfamily and the expansion of the SVP subfamily.The MYB and WRKY gene families comprising 372 and 66 genes could be divided into seven and eight subfamilies in C.serrulata,respectively,based on clustering analysis.Nine hundred forty-one plant disease-resistance genes(R-genes)were detected by searching C.serrulata within the PRGdb.This research provides high-quality genomic information about C.serrulata as well as insights into the evolutionary history of Cerasus species.Xian-Gui Yi Xia-Qing Yu Jie Chen Min Zhang Shao-Wei Liu Hong Zhu Meng Li Yi-Fan Duan Lin Chen Lei Wu Shun Zhu Zhong-Shuai Sun Xin-Hong Liu Xian-Rong Wang 2020Horticulture Research2020,7,1:3
17Porous-hollow nanorods constructed from alternate intercalation of carbon and MoS2 monolayers for lithium and sodium storage显示文摘Weak ion diffusion and electron transport due to limited interlayer spacing and poor electrical conductivity have been identified as critical roadbacks for fast and abundant energy storage of both MoS2-based lithium ion batteries (LIBs) and sodium ion batteries (SIBs). In this work, MoS2 porous-hollow nanorods (MoS2/m-C800) have been designed and synthesized via an annealing-followed chemistry-intercalated strategy to solve the two issues. They are uniformly assembled from ultrathin MoS2 nanosheets, deviated to the rod-axis direction, with expanded interlayer spacing due to alternate intercalation of N-doped carbon monolayers between the adjacent MoS2 monolayers. Electrochemical studies of the MoS2/m-C800 sample, as an anode of LIBs, demonstrate that the superstructure can deliver a reversible discharge capacity of 1,170 mAh·g^-1 after 100 cycles at 0.2 A·g^-1 and maintain a reversible capacity of 951 mAh·g^-1 at 1.25 A·g^-1 after 350 cycles. 20Development and Test of Hydrostatic Extraction Scheme in Spectral Model显示文摘The introduction of 'hydrostatic extraction' scheme, or 'standard stratification approximation', into spectral model gained some advantages compared with commonly used schemes. However, computational instability may occur for high vertical resolution versions if the stratification parameter C0 taken as a constant. In this paper, the possible cause leading to the instability is discussed and an improved scheme presented where C0 is generalized to be a function of both height and latitudes. Hence the reference atmosphere gets closer to the real atmosphere and the temperature deviation field to be expanded becomes smoother everywhere.
