1Prognostic significance of tumor immune microenvironment and immunotherapy:Novel insights and future perspectives in gastric cancer显示文摘Despite a decrease in gastric cancer incidence, the development of novel biologic agents and combined therapeutic strategies, the prognosis of gastric cancer remains poor. Recently, the introduction of modern immunotherapy, especially using immune checkpoint inhibitors, led to an improved prognosis in many cancers. The use of immunotherapy was also associated with manageable adverse event profiles and promising results in the treatment of patients with gastric cancer, especially in heavily pretreated patients. These data have led to an accelerated approval of some checkpoint inhibitors in this setting. Understanding the complex relationship between the host immune microenvironment and tumor and the immune escape phenomenon leading to cancer occurrence and progression will subsequently lead to the identification of prognostic immune markers. Furthermore, this understanding will result in the discovery of both new mechanisms for blocking tumor immunosuppressive signals and pathways to stimulate the local immune response by targeting and modulating different subsets of immune cells. Due to the molecular heterogeneity of gastric cancers associated with differentclinico-biologic parameters, immune markers expression and prognosis, novel immunotherapy algorithms should be personalized and addressed to selected subsets of gastric tumors, which have been proven to elicit the best clinical responses. 2Immunometabolism:A novel perspective of liver cancer microenvironment and its influence on tumor progression显示文摘The initiation and progression of liver cancer, including hepatocellular carcinoma and intrahepatic cholangiocarcinoma, are dependent on its tumor microenvironment. Immune cells are key players in the liver cancer microenvironment and show complicated crosstalk with cancer cells. Emerging evidence has shown that the functions of immune cells are closely related to cell metabolism. However, the effects of metabolic changes of immune cells on liver cancer progression are largely undefined. In this review, we summarize the recent findings of immunometabolism and relate these findings to liver cancer progression. 3Aiming to immune elimination of ovarian cancer stem cells显示文摘Ovarian cancer accounts for only 3% of all cancers in women, but it causes more deaths than any other gynecologic cancer. Treatment with chemotherapy and cytoreductive surgery shows a good response to the therapy. However, in a large proportion of the patients the tumor grows back within a few years. Cancer stem cells, that are less responsive to these treatments, are blamed for this recurrence of disease. Immune therapy either cellular or humoral is a novel concept to treat cancer. It is based on the notice that immune cells invade the tumor. However, the tumor invest heavily to escape from immune elimination by recruiting several immune suppressive mechanisms. These processes are normally in place to limit excessive immune activation and prevent autoimmune phenomena. Here, we discuss current knowledge about the immune(suppressive)status in ovarian cancer. 4Awakening immunity against cancer:a 2017 primer for clinicians显示文摘Cancer immunotherapy has finally joined the pillars of cancer treatment-surgery, radiation, chemotherapy, hormonal therapy, and targeted therapy-in improving cancer patient lives. In the last 5 years, the development of immune checkpoint inhibitor and T cell therapy, particularly chimeric antigen receptor(CAR) T-cell therapy, has been remarkable for the speed, scale, and number of drug approvals. Still, these treatments may also bring unusual adverse effects and clinical outcomes including unprecedented long-term survival. Interrogating the tumor microenvironment and identifying better biomarkers hold the key to improving cancer immunotherapies. CAR T-cell therapy has dramatic effect on leukemias and lymphomas with significant cure rates, but has yet to show comparable effect on solid tumors. Cutting-edge technology will improve both processing and clinical effect of such therapies. Asia has the largest, most rapidly aging population and the largest number of cancer patients in the world. 8Interrelations Between Psychological Stress and Cancer-Deciphering Molecular Signals in the Tumor Microenvironment显示文摘Current research has revealed some links between psychological stress and cellular mutation,neoplastic proliferation and metastasis in patients with cancer.In stressful situations,the stress-related neuroendocrine mediators(e.g.,catecholamines,and glucocorticoids(GSs))are being secreted,via stimulation of the sympathetic nervous system(SNS),and the hypothalamic-pituitaryadrenocortical(HPA)axis.Catecholamine may affect the malignant progression,since they can regulate various cellular signaling pathways,via adrenergic receptors(ARs)that are expressed by different types of neoplastic cells.The ARs increase the proliferation and invasive potential of such cells,and change their“behavior”in the tumor microenvironment.Similarly,cortisol and its glucocorticoid receptors(GRs)can promote stress-induced malignant growth and metastasis.Maladaptation to stressful situations,often relevant to the cancer diagnosis and treatment,may accelerate tumor growth and spread(e.g.,via inflammation,angiogenesis,and migration).Studies have shown that psychological interventions can be helpful for adaptation to adverse circumstances during the therapeutic process in patients with cancer.This mini-review will address some interrelations between psychological stress and cancer.It will discuss how the receptor-mediated signaling pathways may lead to cancer initiation,propagation,and spread.In addition,it will describe a supportive role of the stress reduction strategies,for example,in patients with breast cancer(BC).Katarzyna Anna Rygiel 2018Advances in Modern Oncology Research2018,4,5:0
