| 2 | Genome-wide analysis of odorant-binding proteins and chemosensory proteins in the sweet potato whitefly, Bemisia tabaci显示文摘Odorant-binding proteins (OBPs) and chemosensory proteins (CSPs) of insects are thought to play roles in olfactory recognition affecting host choice, copulation, reproduction and other behaviors. Previous descriptions of OBPs and CSPs in the whitefly Bemisia tabaci often provided no or incomplete genetic information. In this study, we present a genome-wide and transcriptome-wide investigation of the OBPs and CSPs in B. tabaci MEAMl (Middle East-Asia Minorl species). Eight OBP and 19 CSP genes were identified that covered all previous sequences. Phylogenetic analyses showed that the CSP genes had a lineage-specific expansion (BtabBCSP人 BtabBCSP3, BtabBCSPl3, BtabBCSPl 7, BtabBCSP18 and BtabBCSP 19). Expression profiling of OBPs and CSPs by transcriptome sequencing and quantitative real-time polymerase chain reaction (qPCR) revealed that expression patterns differed among developmental stages of B. tabaci MEAM 1. Five OBP genes and 11 CSP genes significantly differed between males and females;four of the 19 CSP genes were highly expressed in adults, while two were highly expressed in nymphs. The expression profiles of the OBP and CSP genes in different tissues of B. tabaci MEAM1 adults were analyzed by qPCR. Four OBP genes found in B. tabaci MEAM 1 were highly expressed in the head. Conversely, only two CSPs were enriched in the head, while the other six CSPs were specifically expressed in other tissues. | 3 | Sequence Analysis of mtDNA CO/Gene and Molecular Phylogeny of Different Geographical Populations of Bemisia tabaci (Gennadius)显示文摘Bemisia tabaci(Gennadius) is a serious pest in many cropping systems worldwide and occurs in different biotypes. ThemtDNA COI gene of the 12 Bemisia tabaci (Gennadius) populations from different regions and countries were analyzed.Based on mtDNA COI sequences, their biotypes were characterized and phylogenetic relationships among thesepopulations were established with the method of UPGMA. The results indicated the genetic similarity between thosepopulations from Beijing, Zhengzhou, Zaozhuang, Nanjing, Shanghai, Haikou, and the B-biotype populations fromCalifornia, Texas, Arizona reached 99.8-100%, which meant the nation-wide infested populations of B.tabaci in China inrecent years were B-biotypes. Another population collected from Kunming of Yunnan Province showed very high similaritywith Q-biotype B.tabaci from Spain and Morocco, which meant the Kunming population was Q-biotype. | 4 | Transmission characteristics of Tomato chlorosis virus (ToCV) by Bemisia tabaci MED and its effects on host preference of vector whitefly显示文摘The epidemiology of Tomato chlorosis virus(ToCV)in China is closely associated with its vector whitefly,Bemisia tabaci(Gennadius)MED.However,the transmission characteristics of ToCV by B.tabaci MED remain poorly understood.In this study,we analyzed:1)the horizontal and vertical transmission of ToCV by B.tabaci MED whiteflies;2)the acquisition of ToCV by male and female B.tabaci MED whiteflies after different feeding durations;3)the transmission efficacy of viruliferous male and female B.tabaci MED whiteflies after different inoculation access periods(IAPs);4)the retention of ToCV by viruliferous male and female B.tabaci MED whiteflies after a 48 h acquisition access period(AAP);and 5)the effects of ToCV on host choice of healthy or ToCV-infected tomato plant of viruliferous and non-viruliferous B.tabaci MED at different time points.Our results showed that:1)viruliferous males could not transfer ToCV to non-viruliferous females,and vice versa,viruliferous females could not pass on ToCV to non-viruliferous males.ToCV could not be detected in the F1 generation adults;2)ToCV could be detected within 4.0%of females or males after a 20 min AAP;3)ToCV could be detected in 33.3%of tomato plants inoculated by 10 viruliferous males or females with IAPs of 20 or 30 min;4)the maximum retention time in females was 7 and 5 days in males;and 5)non-viruliferous B.tabaci MED did not show a preference for ToCV-infected tomato plants or healthy tomato plants.However,viruliferous B.tabaci MED whiteflies did prefer to settle on healthy tomato plants over ToCV-infected tomato plants.These findings will be helpful to better understand the epidemiology of the recently emerged plant virus,ToCV,in tomato fields in China. | WEI Ke-ke LI Jie DING Tian-bo LIU Tong-xian CHU Dong | 2019 | Journal of Integrative Agriculture2019,18,9: | 4 |
| 5 | Development and reproduction of Bemisia tabaci (Homoptera: Aleyrodidae) on three bean species显示文摘The development, survivorship and reproduction of Bemisia tabaci B-biotype on three bean species were studied at 26 ± 1 ℃ in the laboratory. The developmental periods from egg to adult varied from 27.80 days on garden beans to 18.20 days on soybeans. The survivorship from egg to adult on soybeans, cowpeas and garden beans was 77.14, 70.14 and 64.28%, respectively. The average longevity of female adults ranged from 12.30 days on soybeans to 9.80 days on garden beans, and the oviposition of B. tabaci varied from 160.85 eggs on soybeans to 98.00 eggs on garden beans. Life table parameters were calculated as biological attributes for Bemisia tabaci populations fed on three bean species. The results indicated that the intrinsic rate of increase (rm), the finite rate of increase ( λ ) and net reproductive rate (R0) were high for populations fed on soybeans, with values of 0.1857,1.2041 and 82.1576, respectively. The corresponding values were less for populations fed on garden beans, with values of 0.1097, 1.1159 and 31.2661, respectively. The parametric values for cowpeas were intermediate between soybeans and garden beans but no significant difference were observed for the rm values for soybeans and cowpeas. | 7 | Characterization of field-evolved resistance to cyantraniliprole in Bemisia tabaci MED from China显示文摘Cyantraniliprole is a novel anthranilic diamide insecticide with significant efficacy against Bemisia tabaci,an important pest insect worldwide.In this study,we conducted reversion and selection work and genetic analysis,and determined cross-resistance spectrum and synergism of cyantraniliprole resistance based on the reported population,SX population,of B.tabaci collected from Shanxi Province,China.Compared with a susceptible strain(MED-S),SX population,the field-evolved cyantraniliprole-resistant population exhibited 26.4-fold higher resistance to cyantraniliprole.In SX,a sharp decline of cyantraniliprole resistance was shown in the absence of selection.Another tested strain,SX-R,was established from SX population after successive selection with cyantraniliprole and recently developed 138.4-fold high resistance to cyantraniliprole.SX-R had no cross-resistance to abamectin,imidacloprid,thiamethoxam,sulfoxaflor,or bifenthrin.Genetic analysis illustrated that cyantraniliprole resistance in SX-R was autosomally inherited and incompletely dominant.Additionally,piperonyl butoxide(PBO)significantly inhibited cyantraniliprole resistance in the SX-R strain.In conclusion,the selection of SX with cyantraniliprole led to high resistance to cyantraniliprole which is incompletely dominant and no cross-resistance to several common types of insecticides.Enhanced oxidative metabolism is possibly involved in the resistance of SX-R,yet target-site resistance could not be excluded. | WANG Ran WANG Jin-da CHE Wu-nan SUN Yan LI Wen-xiang LUO Chen | 2019 | Journal of Integrative Agriculture2019,18,11: | 2 |
| 8 | Development and reproduction of ‘B' biotype Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) on four ornamentals显示文摘Effects of four commercial ornamentals on the development, survivorship and reproduction of the whitefly B biotype Bemisia tabaci (Gennadius) were studied in the laboratory (temperature 26 ±1℃; relative humidity 75%-90%; L: D 14:10). The total survivorship from egg to adult on hibiscus (Hibiscus rosa-sinensis L.), poinsettia (Euphorbia pulcherrima Will), cottonrose hibiscus (Hibiscus mutabilis L.) and variegated leafcroton (Codiaeum variegatum ‘Aucubaefolium') were 33.69%, 40.55%, 79.11%, and 29.39%,respectively. The developmental periods from egg to adult varied from 23.12 days on cottonrose hibiscus to 32.13 days on hibiscus. The average longevity of adult females ranged from 6.87 days on variegated leafcroton to 21.07 days on poinsettia. The average numbers of eggs laid per female were 9.20, 25.13, 54.45, and 26.79 on the above respective hosts. The intrinsic rates of natural increase (rm) for B biotype B. tabaci on cottonrose hibiscus was the highest.
