1Bioinformatic analysis of cytokine expression in the proximal and distal nerve stumps after peripheral nerve injury显示文摘In our previous study,we investigated the dynamic expression of cytokines in the distal nerve stumps after peripheral nerve injury using microarray analysis,which can characterize the dynamic expression of proteins.In the present study,we used a rat model of right sciatic nerve transection to examine changes in the expression of cytokines at 1,7,14 and 28 days after injury using protein microarray analysis.Interleukins were increased in the distal nerve stumps at 1–14 days post nerve transection.However,growth factors and growth factor-related proteins were mainly upregulated in the proximal nerve stumps.The P-values of the inflammatory response,apoptotic response and cell-cell adhesion in the distal stumps were higher than those in the proximal nerve stumps,but the opposite was observed for angiogenesis.The number of cytokines related to axons in the distal stumps was greater than that in the proximal stumps,while the percentage of cytokines related to axons in the distal stumps was lower than that in the proximal nerve stumps.Visualization of the results revealed the specific expression patterns and differences in cytokines in and between the proximal and distal nerve stumps.Our findings offer potential therapeutic targets and should help advance the development of clinical treatments for peripheral nerve injury.Approval for animal use in this study was obtained from the Animal Ethics Committee of the Chinese PLA General Hospital on September 7,2016(approval No.2016-x9-07).Xiao-Qing Cheng Wen-Jing Xu Xiao Ding Gong-Hai Han Shuai Wei Ping Liu Hao-Ye Meng Ai-Jia Shang Yu Wang Ai-Yuan Wang 2021Neural Regeneration Research2021,16,5:0
22,5-己二酮染毒对大鼠坐骨神经和大脑皮质神经细胞粘附因子表达的影响显示文摘目的初步观察2,5-己二酮(2,5-hexanetione,HD)染毒大鼠中毒性周围神经病变时坐骨神经和大脑皮质中神经细胞粘附因子(neural cell adhesion molecule,NCAM)蛋白的表达变化。方法9周龄(体重280~310 g)SPF级Wistar雄性大鼠分为3组,每组23只,每日经腹腔注射染毒给予HD(低剂量组100 mg/kg,高剂量组300 mg/kg),连续8周,每周5次,对照组给予等体积生理盐水。处死各组10只动物并分离、冻存坐骨神经和大脑皮质运动代表区,采用免疫组化方法检测NCAM的蛋白表达。另取3只分离坐骨神经用于电镜观察。剩余动物停止染毒、自然恢复8周后取材。结果染毒后高剂量组动物第4周起动物后肢热痛觉阈值升高,染毒8周后高剂量组动物后肢全部瘫痪,低剂量组动物后肢肌力下降,步态异常。电镜观察染毒组动物坐骨神经髓鞘形态异常。对于坐骨神经,染毒8周时低、高剂量组中NCAM表达量分别为对照组的144%和85%,染毒终止、恢复8周后为166%(P<0.05)和145%。对于大脑皮质,染毒8周时与染毒终止、恢复8周时各剂量组蛋白表达与对照组差异无统计学意义(P>0.05)。结论HD染毒大鼠中毒性周围神经病变时坐骨神经NCAM蛋白表达发生变化,NCAM参与了中毒性周围神经病变发生。周雯 姚文环 程东 李慧 范轶欧 张静 唐慧 张天亮 2020卫生研究2020,49,4:0
