1Atorvastatin protects against cerebral ischemia/ reperfusion injury through anti-inflammatory and antioxidant effects显示文摘In addition to its lipid-lowering effect, atorvastatin exerts anti-inflammatory and antioxidant effects as well. In this study, we hypothesized that atorvastatin could protect against cerebral ischemia/reperfusion injury. The middle cerebral artery ischemia/reperfusion model was established, and atorvastatin, 6.5 mg/kg, was administered by gavage. We found that, after cerebral ischemia/ reperfusion injury, levels of the inflammation-related factors E-selectin and myeloperoxidase were upregulated, the oxidative stress-related marker malondialdehyde was increased, and superoxide dismutase activity was decreased in the ischemic cerebral cortex. Atorvastatin pretreatment significantly inhibited these changes. 2Protective effects of tumor necrosis factor α antibody and ulinastatin on liver ischemic reperfusion in rats显示文摘AIM: To study the protective effects of tumor necrosis factor α (TNFα) antibody and ulinastatin on liver ischemic reperfusion in rats.METHODS: One hundred and twenty male SD rats were randomly divided into four groups: normal control group, ischemic group, TNFα antibody group and TNFα antibody + ulinastatin group. The animals were killed at 0, 3, 6, 9, 12 h after ischemia for 60 min and followed by reperfusion. Serum alanine aminotransferase (ALT), malondialdehyde (MDA) and liver histopathology were observed.RESULTS: After ischemic reperfusion, the serum ALT and MDA were remarkably increased, and the hepatic congestion was obvious. 4胃癌并发急性大出血的临床分析及诊治探讨(附82例报告)(英文)显示文摘目的通过对82例胃癌并发大出血的病人进行临床分析,观察其临床特点,探讨改进诊治方法。方法分析82例胃癌并急性大出血病人的病史,临床表现,病理分型,治疗方法和治疗效果。结果胃癌并发急性大出血大部分病例发生在进展期胃癌BorrmannⅡ、Ⅲ型,少数病例发生于进展期胃癌BorrmannⅣ型及早期胃癌Ⅱc、Ⅲ型,而且大部分病灶位于胃体中上部小弯侧的前后壁。24 h出血量可达1000 ml以上。临床特点主要表现呕血,排柏油样大便或暗红色便,半数病例出现呕血+排柏油样大便并伴有休克征象。结论胃癌并发急性大出血病人,大部分经保守治疗可使病情稳定或止血。少数出血严重病人经保守治疗无效需直接手术治疗。钡餐及纤维胃镜检查仍是基层医院可靠的诊断手段,手术是首选的治疗方法。李大伟 林飞鹤 谢春发 周智群 2004中国现代医学杂志2004,14,19:11
5脓毒症疗程中的抗凝治疗显示文摘脓毒症的实质是失控的炎症反应和凝血紊乱,炎症反应可激活凝血系统,二者相互促进,相互影响,加速器官功能障碍的发生。在脓毒症的治疗过程中,抗炎和抗凝即成为关键的靶点。目前对抗炎药物的研究相对集中,但对患者的生存率改善并不明显。随着对脓毒症凝血功能紊乱发病机制的认识,一些潜在具有'炎症-凝血-纤溶'多重调节活性的生理性抗凝药物在脓毒症凝血功能障碍治疗中逐渐得到关注,包括活化蛋白C、抗凝血酶Ⅲ、重组血栓调节蛋白等和肝素等。本文就脓毒症的抗凝治疗做一简要回顾。李鑫 马晓春 2015中国实用内科杂志2015,35,11:10
6抗P-选择素功能域单抗对急性肾损伤防治及免疫防御功能的影响显示文摘目的 探讨抗P-选择素lectin-EGF功能域单抗(PsL-EGFmAb)对大鼠急性肾损伤防治及机体免疫防御功能影响。方法 建立大鼠肾缺血再灌注损伤模型,观察和比较经PsL-EGFmAb及其配体拮抗剂N-去硫酸肝素处理前后,大鼠肾组织P-选择素及其mRNA表达、组织病理学改变、肾功能变化以及免疫细胞功能状态。结果 肾缺血再灌注后,大鼠肾内以肾小管上皮细胞为主P-选择素表达增强,出现明显的组织病理学改变及肾功能损害;同时体内T、B细胞活性受抑,而NK细胞明显激活。经给予PsL-EGFmAb及其配体拮抗剂N-去硫酸肝素处理后,肾组织P-选择素及其mRNA表达受抑,病理学改变及肾功能损害减轻,尤以PsL-EGFmAb治疗效果更为明显;此外大鼠体内三种免疫细胞功能均受到不同程度的调节。结论 抗P-选择素功能域单抗对肾缺血再灌注损伤具有防治作用,其抗黏附靶向效果强于N-去硫酸肝素,并对大鼠免疫防御功能可能具有调节作用。周同 王锋 孙桂芝 张玉梅 张明均 张冬青 陈楠 2004上海第二医科大学学报2004,24,7:9

8辛夷挥发油对肾缺血再灌注损伤的保护作用显示文摘目的研究辛夷挥发油对大鼠肾缺血再灌注损伤的保护作用。方法建立肾缺血再灌注损伤大鼠模型,将实验动物随机分为4组:假手术组、生理盐水对照组、辛夷挥发油治疗组(0.05mL/kg和0.10mL/kg)。观察治疗前后肾组织病理学与肾功能改变、P-选择素(P-selectin)与细胞间黏附分子-1(ICAM-1)表达。结果肾缺血再灌注后,生理盐水对照组大鼠肾组织病理学明显改变,尿素氮(BUN)和血肌酐(Scr)明显升高(P<0.01),P-selectin和ICAM-1表达增高(P<0.01);辛夷挥发油治疗组(0.05mL/kg和0.10mL/kg)大鼠肾组织病理学轻微改变,BUN与Scr低于生理盐水对照组(P<0.01),P-selectin表达受抑制(P<0.05),ICAM-1表达受抑制(P<0.01)。结论辛夷挥发油能抑制P-selectin与ICAM-1的表达,对肾缺血再灌注损伤有保护作用。陈志东 王锋 汪年松 2009中国现代医学杂志2009,19,10:8

11辛夷抗炎作用研究初探显示文摘目的对中药辛夷的抗炎效应初步探讨。方法利用TNF—α诱导活化的内皮细胞与中性粒细胞的体外黏附实验与肾缺血再灌注损伤大鼠模型,观察辛夷挥发油对中性粒细胞黏附功能的影响以及肾损伤的保护作用。结果体外培养的人脐静脉内皮细胞在TNF—α刺激后,与中性粒细胞发生黏附,黏附量为248.6±.13.8;而辛夷挥发油处理后,黏附量明显降低,在0.5、1.0,2.0ul/ml浓度的黏附量分另4为191.6±8.2,158.2±9.0,155.2±9.7(均P〈0.05)。缺血再灌注后,肾组织出现明显炎症损伤改变.BUN、Ser水平明显升高,但辛夷治疗组肾脏组织病理改变与肾功能损害较轻。结论本实验条件下,辛夷具有一定抗炎效应。王锋 陈志东 汪年松 2007中国医师杂志2007,9,4:5

13抗黏附治疗对大鼠急性肾损伤中树突状细胞迁移聚集的影响显示文摘目的 探讨黏附分子P-选择素、ICAM-1及树突状细胞(DC)在大鼠肾缺血再灌注损伤中作用,以及抗P-选择素单抗的抗黏附抑制及其防治效果。方法 建立肾缺血再灌注损伤大鼠模型,随机分为P-选择素单抗治疗组(n=20)和非治疗组(n=20)。按不同再灌注时间(1,3,6和24h)再分为4组,另设假手术组(n=5)作为对照。采用免疫组化LSAB法和免疫双染与荧光图像分析法,分别观察大鼠肾组织中P-选择素、ICAM-1表达及CD1a^+CD80^+DC分布变化;同时观察丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性以及细胞凋亡等改变。结果 ①缺血再灌注1h起,P-选择素即以肾小管上皮细胞为主于肾内广泛高表达,至24h仍维持一定量水平;ICAM-1于缺血再灌注6h起持续以肾血管为主表达上调和明显增强。②CD1a^+CD80^+DC自再灌注1h起以肾小管间质为主分布数量逐渐增多,至24h达峰,并与大鼠血尿素氮和肌酐水平密切相关。③此外缺血再灌注后,大鼠肾组织MDA含量增加,SOD活性下降,出现明显的肾组织细胞凋亡。④经抗P-选择素单抗处理后,大鼠肾组织P-选择素和ICAM-1表达受到抑制,CD1a^+CD80^+DC分布及数量减少,MDA含量降低和SOD活性上升,细胞凋亡减少,以及肾组织病理损伤和肾功能也相应减轻和改善。结论 本研究提示。周同 孙桂芝 张玉梅 吴开胤 李晓 张冬青 陈玉英 胡庆沈 陈楠 2003中国微循环2003,7,5:2
14非抗凝N-去硫酸肝素对胃癌转移抑制作用的实验研究显示文摘目的研究N-去硫酸肝素对人胃癌组织原位移植非肥胖性糖尿病(non-obesity diabetes,NOD)重度联合免疫缺陷(SCID)小鼠转移模型的肿瘤转移抑制、血管生成和血管内皮生长因子(VEGF)表达的影响。方法建立人胃癌组织原位移植NOD SCID小鼠胃痛转移模型,20只小鼠均分成2组。移植后1周,分别静脉注射0.9%氯化钠溶液(0.9%氯化钠溶液组)与10 mg/kg N-去硫酸肝素(N-去硫酸肝素组),每周2次,共3周。第6周处死动物,观察肿瘤转移情况,免疫组化法检测肿瘤组织微血管密度、VEGF表达,荧光定量PCR检测肿瘤组织VEGF mRNA表达。结果0.9%氯化钠溶液组10只小鼠9只有肿瘤转移,N-去硫酸肝素组10只小鼠只有2只转移,两组间差异有统计学意义(P<0.05)。未发现出血等不良反应。0.9%氯化钠溶液组平均微血管密度为9.1±3.4,N-去硫酸肝素组为4.7±1.8,两组间差异有统计学意义(t=3.617,P<0.05)。0.9%氯化钠溶液组有9只VEGF阳性表达,明显高于N-去硫酸肝素组的2只(P<0.05)。荧光定量PCR测定显示,N-去硫酸肝素组VEGF mRNA(Ct:19.56±1.53)表达较0.9%氯化钠溶液组(Ct=16.38±1.68)低,两组间差异有统计学意义(t=4.425,P<0.05)。结论N-去硫酸肝素通过抑制肿瘤组织VEGF表达和血管生成,抑制肿瘤转移。N-去硫酸肝素无明显出血等不良反应。陈金联 陈维雄 朱金水 洪静 陆金来 陈明祥 陈尼维 陈国强 耿建国 2006中华消化杂志2006,26,12:2
15N-去硫酸肝素对SCID小鼠胃癌血管生成和VEGF表达的影响显示文摘目的:研究N-去硫酸肝素对人胃癌重度联合免疫缺陷(SCID) 小鼠转移模型肿瘤转移抑制、血管生成和血管内皮生长因子(VEGF)表达的影响.方法:建立人胃癌组织原位移植SCID小鼠转移模型,随机分成两组.移植1 wk,分别从静脉内注射生理盐水(生理盐水组) 与N-去硫酸肝素[10 mg/(kg·d)](N-去硫酸肝素组),2次/wk, 共3 wk.第6 wk处死动物,观察肿瘤转移情况,免疫组化方法检测肿瘤组织微血管密度(MVD)、VEGF的表达.结果:生理盐水组肿瘤转移率为80%,N-去硫酸肝素组转移率为20%,两组之间差异有统计学意义(P<0.05).未发现出血等副作用.生理盐水组平均微血管密度为8.0±3.1,N-去硫酸肝素治疗组平均微血管密度为4.3±1.8.经统计学处理,两组之间差别有显著意义(P<0.05).生理盐水组VEGF阳性表达率明显高于N-去硫酸肝素治疗组,分别为90%与20%(P<0.05).结论:N-去硫酸肝素通过抑制肿瘤组织VEGF表达和血管生成,从而抑制肿瘤的转移,并且无明显出血等不良反应.陈金联 陆金来 陈明祥 陈维雄 朱金水 陈尼维 陈国强 耿建国 2005世界华人消化杂志2005,13,22:2

17N-去硫酸肝素对胃癌血管生成及抑制作用的研究显示文摘目的:研究N-去硫酸肝素对人胃癌裸鼠模型肿瘤生长抑制、血管生成和细胞凋亡的影响。方法:建立人胃癌皮下移植裸鼠模 型,随机分成4组。移植1周后,分别从腹腔注射生理盐水(对照组),卡铂(5 mg·kg^(-1) ·d^(-1) ),N-去硫酸肝素(10 mg·kg^(-1) ·d^(-1) ), 及卡铂(5 mg·kg^(-1) ·d^(-1) )与 N-去硫酸肝素(10 mg·kg^(-1) ·d^(-1) ),每周3次,共3周。第7周处死动物,测量肿瘤大小与重量、抑瘤 率、微血管密度(MVD)、胃癌细胞凋亡指数(AI)。结果:对照组、卡铂组、N-去硫酸肝素组、卡铂与 N-去硫酸肝素联合组的肿瘤重量 (mg)分别为1750. 4±144. 2、 1655. 0±157. 4、 1118. 0±116. 2与725. 6±130. 0,抑瘤率分别为5. 6%、36. 1%和58. 5%。MVD 分别为 14. 8±4. 5、 13. 2±4. 1、 3. 8±1. 7与3. 4±1. 8。凋亡指数分别为1. 37±0. 19、 3. 85±0. 22、 11. 81±2. 9与18. 85±3. 24。与对照组、卡铂 组相比,N-去硫酸肝素组、卡铂与 N-去硫酸肝素联合组的胃癌生长、微血管密度明显抑制,细胞凋亡明显增高。结论:N-去硫酸肝 素通过抑制肿瘤血管生成,诱导肿瘤细胞凋亡,从而抑制肿瘤的生长。N-去硫酸肝素无明显出血等不良反应。陈金联 陈维雄 朱金水 陈尼维 罗小蓉 耿建国 2005中国临床医学2005,12,5:0
18LOW MOLECULAR WEIGHT HEPARIN DECREASES THE ALBUMINURIA AND RENAL PATHOLOGICAL CHANGES IN DIABETIC NEPHROPATHY RATS显示文摘Objective To investigate the renoprotection effect of low molecular weight heparin(LMWH)in diabetic rats.Methods Diabetic rats were induced by injection of steptozotocin(STZ).Rats were randomly divided into three groups:control group,diabetic model group,and LMWH treatment group.The levels of total cholesterol(TC),triglyceride(TG),prothrombin time(PT),kaolin partial thromboplastin time(KPTT),24-hour urinary albumin excretion rate(UAER),and serum soluble P-selectin were determined after 8 weeks.Glomerular morphology was observed by light microscopy.Results The levels of TG,UAER,and P-selectin in LWMH treatment group were lower than those in model group(all P<0.01).The levels of PT and KPTT in LWMH treatment group were shorter than those in model group.And LWMH improved the histological changes of diabetic rats.Conclusion LMWH has some renal protective effects in diabetic rats,partly through down-regulating the expression of P-selectin.王锋 邢涛 汪年松 晏春根 2010Medical Bulletin of Shanghai Jiaotong University2010,22,2:0

20ANTI-INFLAMMATORY EFFECTS OF MAGNOLIAE FARGESII VOLATILE OIL显示文摘Objective To explore the anti-inflammatory effects of magnoliae fargesii volatile oil.Methods Human umbilical vein endothelial cells (HUVECs) were stimulated by TNF-α to express the adhesion molecules. Then the anti-adhesion effects of magnoliae fargesii volatile oil between HUVECs and human peripheral neutrophils were observed. The ischemia-reperfusion animal models were established by 60min renal ischemia followed by 1, 3, 6 and 24h reperfusion. Rats were randomly divided into the following groups: the sham-operation controls, ischemic group only treated with normal saline, and treated group infused magnoliae fargesii volatile oil before reperfusion. Then the renal injury of rats was detected. Results High rate of cell adhesion between HUVECs and neutrophils was observed. Magnoliae fargesii volatile oil could inhibit the adhesion process at the concentration of 0.5μL/mL (191.6±8.6), 1.0μL/mL (158.2±9.0) and 2.0μL/mL (155.2±9.7) (P<0.05). The anti-adhesion effects were strengthened with the increase of volatile oil concentration. Blood urea nitrogen and creatinine levels of the animal models were significantly increased after 24h reperfusion while the increase was remarkably attenuated by the treatment with magnoliae fargesii volatile oil. The renal injury was severe after 1h reperfusion, which was significantly attenuated by the treatment of magnoliae fargesii volatile oil.
