1基于氧化应激-炎症-衰老探讨当归四逆汤对亚急性衰老大鼠免疫功能的影响显示文摘目的 探讨当归四逆汤通过调节氧化应激反应对衰老机体免疫功能的影响。方法 通过注射D-半乳糖(D-glactose, D-gal)制备亚急性衰老大鼠模型,实验设正常组、模型组、V组、当归四逆汤组,模型制备完成后给予当归四逆汤组大鼠灌胃相应剂量的当归四逆汤,V组大鼠给予维生素E,其余2组大鼠灌胃等容积0.9%氯化钠溶液。Morris水迷宫实验观察大鼠的学习记忆能力,测量大鼠脾、肝、胸腺的脏器指数,比色法检测大鼠血清中超氧化物歧化酶(superoxide dismutase, SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)、丙二醛(malondialdehyde, MDA),铁离子还原法检测大鼠血清中总抗氧化能力(total anti-oxidation capability, T-AOC),酶联免疫吸附试验检测大鼠血清中白细胞介素(interleukin, IL)-2、IL-6及IL-10水平。结果 与正常组比较,模型组大鼠进入逃生平台范围次数明显减少(P<0.01)、逃避潜伏期明显延长(P<0.01),脾脏、肝脏、胸腺指数皆明显降低(P<0.01),血清中SOD、GSH-Px、T-AOC、IL-2、IL-10含量明显降低(P<0.01),MDA、IL-6含量明显升高(P<0.01);与模型组比较,V组和当归四逆汤组进入逃生平台范围次数明显增多(P<0.01),逃避潜伏期明显缩短(P<0.05),脾脏、肝脏、胸腺指数皆明显升高(P<0.05,P<0.01),血清中SOD、GSH-Px、T-AOC、IL-2、IL-10含量明显升高(P<0.01)、MDA、IL-6含量明显降低(P<0.01)。与V组比较,当归四逆汤组大鼠血清中IL-2、IL-10含量明显升高(P<0.01),IL-6含量明显降低(P<0.01)。结论 当归四逆汤能够通过减弱衰老机体的氧化应激反应,提高免疫系统功能。刘紫微 王振亮 王兵 2022中国中医基础医学杂志2022,28,11:3
2Efficacy of quercetin, oleanolic acid, icariin on apoptosis and mitogen-activated protein kinases signaling pathways in hippocampal neurons of Sprague-Dawley rats cultured with high glucose medium显示文摘OBJECTIVE:To investigate the effect of quercetin,oleanolic acid,icariin and their compatibility on the apoptosisofhippocampalneuronsof Sprague-Dawley(SD)rats cultured with high glucose medium and the possible mechanism.METHODS:The extracts were purchased from China Food and Drug Control Institute and Sellect.Hippocampus was obtained from newborn 24 h SD rats.After culturing the hippocampus in different medium for 72 h,flow cytometry was used to detect the apoptosis of hippocampal neurons,and Western blot was utilized to test the expressions of p-p38,p38,p-c-Jun N-terminal kinase(JNK)and JNK.RESULTS:Compared with the control group(CG),the neuronal apoptosis rate and the ratios of p-p38/p38 and p-JNK/JNK were significantly increased in the high glucose group(GG)(P<0.01);Compared with the GG,the apoptosis rate and the ratios ofp-p38/p38 and p-JNK/JNK were significantly decreased in other drug groups(P<0.01);Compared with the monomer groups respectively,the apoptosis rate and the ratios of p-p38/p38 and p-JNK/JNK in the two-drug groups and the three-drug group all decreased(P<0.01);Compared with the two-drug groups,the neuronal apoptosis rate and the ratio of p-JNK/JNK of the three-drug group decreased(P<0.05).CONCLUSION:Under the condition of high glucose,the quercetin,oleanolic acid and icariin can alleviate the apoptosis of hippocampus neurons,reduce the phosphorylation of p38 and JNK in p38 mitogen-activated protein kinases and JNK signaling pathway.And the efficacy of the three drugs in combination with each other can be strengthened.YAN Bin WANG Jingbo TIAN Guoqing 2021Journal of Traditional Chinese Medicine2021,41,5:2
