1丁酸钠抑制氟中毒可诱导小胶质细胞活化及炎症因子表达增多显示文摘背景:在中枢神经系统疾病中,致病因素刺激小胶质细胞过度活化分泌炎性因子继而导致神经元进一步损伤,去乙酰化酶抑制剂丁酸钠能否降低氟中毒引起小胶质细胞的过度活化以及引起的炎性作用,值得深入研究。目的:丁酸钠对氟诱导的小胶质细胞过度活化以及炎性因子白细胞介素1β、白细胞介素6、肿瘤坏死因子α表达的影响。方法:用含体积分数为10%胎牛血清的DMEM/F12培养基常规培养BV-2小胶质细胞,取对数生长期细胞用CCK-8法观察不同浓度氟化钠、丁酸钠及二者联合使用对BV-2细胞活力的影响。通过倒置显微镜、RT-qPCR和Western blot法观察氟化钠、丁酸钠及二者联合应用后的BV-2细胞形态以及炎症因子白细胞介素1β、白细胞介素6、肿瘤坏死因子αmRNA和蛋白表达。结果与结论:①CCK-8结果:质量分数25×10^-6氟化钠能降低BV-2细胞活力,0.25 mmol/L丁酸钠能升高BV-2细胞活力;0.25 mmol/L丁酸钠能逆转25×10^-6氟化钠对BV-2细胞活力的降低作用;②RT-qPCR和western blot结果:0.25 mmol/L丁酸钠能够拮抗25×10^-6氟化钠引起的白细胞介素1β、白细胞介素6、肿瘤坏死因子α过表达作用;③结果说明在一定的剂量范围内,丁酸钠可减轻氟致小胶质细胞介导的炎症反应。王正东 黄娜 陈婧娴 郑作兵 胡鑫宇 李梅 苏晓 苏学森 颜南 2021中国组织工程研究2021,25,7:6
2Proprotein convertase 1/3-mediated down-regulation of brain-derived neurotrophic factor in cortical neurons induced by oxygen-glucose deprivation显示文摘Brain-derived neurotrophic factor(BDNF)has robust effects on synaptogenesis,neuronal differentiation and synaptic transmission and plasticity.The maturation of BDNF is a complex process.Proprotein convertase 1/3(PC1/3)has a key role in the cleavage of protein precursors that are directed to regulated secretory pathways;however,it is not clear whether PC1/3 mediates the change in BDNF levels caused by ischemia.To clarify the role of PC1/3 in BDNF maturation in ischemic cortical neurons,primary cortical neurons from fetal rats were cultured in a humidified environment of 95%N_2 and 5%CO_2 in a glucose-free Dulbecco's modified Eagle's medium at 37℃for3 hours.Enzyme-linked immunosorbent assays and western blotting showed that after oxygen-glucose deprivation,the secreted and intracellular levels of BDNF were significantly reduced and the intracellular level of PC1/3 was decreased.Transient transfection of cortical neurons with a PC1/3 overexpression plasmid followed by oxygen-glucose deprivation resulted in increased PC1/3 levels and increased BDNF levels.When levels of the BDNF precursor protein were reduced,the concentration of BDNF in the culture medium was increased.These results indicate that PC 1/3 cleavage of BDNF is critical for the conversion of pro-BDNF in rat cortical neurons during ischemia.The study was approved by the Animal Ethics Committee of Wuhan University School of Basic Medical Sciences.Xiang-Yang Zhang Feng Liu Yan Chen Wei-Chun Guo Zhao-Hui Zhang 2020Neural Regeneration Research2020,15,6:3

4Fingolimod(FTY720) improves postoperative cognitive dysfunction in mice subjected to D-galactose-induced aging显示文摘Neurocognitive dysfunction is a common postoperative complication,especially in older adult patients.Fingolimod(FTY720)is a sphingosine-1-phosphate receptor modulator that has been found to be neuroprotective in several animal models of central nervous system disease.However,few reports have examined whether FTY720 could mitigate postoperative cognitive dysfunction.In this study,we investigated whether FTY720 could prevent postoperative neurocognitive impairment in mice subjected to D-galactose-induced aging.We induced an accelerated model of aging by administering an intraperitoneal injection of D-galactose.Subsequently,we performed a partial hepatolobectomy under sevoflurane anesthesia.FTY720(1 mg/kg)was administered intraperitoneally 3 hours before and 24 hours after anesthesia and surgery.Our results indicated that anesthesia and surgery significantly impaired spatial memory in the Y-maze test 6 hours after surgery.We also found that problem solving ability and long-term memory in the puzzle box test on postoperative days 2–4 were significantly improved by FTY720 treatment.Immunohistochemical staining and western blot assay demonstrated that FTY720 significantly inhibited microglial activation in the hippocampal CA1 region of mice 6 hours and 3 days after anesthesia,and down-regulated the expression of synaptic-related proteins postsynaptic density protein 95 and GluR2 in the hippocampus.These results indicate that FTY720 improved postoperative neurocognitive dysfunction in mice subjected to D-galactose-induced aging.This study was approved by the Experimental Animal Ethics Committee of the Third Xiangya Hospital of Central South University of China(approval No.LLSC(LA)2016-025)on September 27,2016.Jie Zhang Bin Xiao Chen-Xu Li Yi Wang 2020Neural Regeneration Research2020,15,7:2

6“百会”“大椎”穴电针预处理减轻反复丙泊酚麻醉诱导的大鼠学习记忆障碍显示文摘目的:观察“百会”“大椎”穴电针预处理减轻反复丙泊酚麻醉诱发大鼠学习记忆障碍的作用。方法:通过给予连续5 d丙泊酚麻醉诱导学习记忆障碍模型,在丙泊酚给药前给予电针预处理连续5 d完成电针干预。Morris水迷宫检测大鼠学习记忆能力;HE染色用于观察海马神经细胞数量及形态改变;免疫荧光染色检测海马Dentate Gyrus区磷酸化环磷腺苷效应元件结合蛋白(pCREB)蛋白表达;用Western Blotting测定B细胞淋巴瘤-2(Bcl-2)、半胱氨酸蛋白酶-3(Caspase-3)、B细胞淋巴瘤-2相关X蛋白(Bax)和CREB蛋白的表达情况。结果:Morris水迷宫结果:与对照组比较,丙泊酚组大鼠的穿台次数、目标象限游泳时长及路径占比明显减少,差异具有统计学意义(P<0.05);电针组大鼠的穿台次数、目标象限游泳时长及路径占比均明显高于丙泊酚组,差异具有统计学意义(P<0.05)。HE结果:丙泊酚组海马DG区细胞层数减少,细胞密集度降低;电针组海马DG区神经元细胞层数显著恢复,细胞密集度明显升高。免疫荧光结果:与对照组比较,丙泊酚组海马区pCREB蛋白累积荧光强度显著降低,差异具有统计学意义(P<0.05);与丙泊酚组比较,电针组海马区pCREB蛋白累积荧光强度显著升高,差异具有统计学意义(P<0.05)。Western blot结果:与对照组比较,Bax、Caspase-3在丙泊酚组显著表达,差异具有统计学意义(P<0.05),而Bcl-2、CREB表达明显减少,差异具有统计学意义(P<0.05);与丙泊酚组比较,Bax、Caspase-3在电针组表达明显减少,差异具有统计学意义(P<0.05),相反,Bcl-2、CREB在电针组显著表达,差异具有统计学意义(P<0.05)。结论:“百会”“大椎”穴电针预处理可以有效减轻反复丙泊酚麻醉诱发的大鼠学习记忆障碍,其机制可能与上调pCREB、CREB蛋白以及调控凋亡相关因子的表达,进而抑制神经元凋亡,改善学习记忆,发挥神经保护作用。范舜钦 王喜军 韦松里 陆思施 郭永榕 2023针灸临床杂志2023,39,8:0
7影响认知功能的药物研发进展显示文摘神经系统药物在整个药物研发中处于重要地位,其中调节认知功能的药物更是成为社会发展的迫切需求,但认知类药物临床供应严重不足且研发面临受挫。本文结合认知药理学的研究现状,对两大类认知药物进行简要概述,分别为影响正常认知功能的药物及改善认知功能障碍的药物。目前的新药研发集中在调节神经递质、靶向Aβ和Tau蛋白、神经保护及疏通血管等方面,仍需要新的研究方法及思路。本文将结合临床及在研药物,以阿尔茨海默病为主,提出并分析认知药理学发展的任务和挑战,总结认知药物研发策略,指出认知药理学的发展机遇,以期为广大科研人员提供新思路,共同促进认知药物的研发与发展。于子茹 杜冠华 2020药学学报2020,55,5:0
