| 1 | Role of macrophages in peripheral nerve injury and repair显示文摘Resident and inflammatory macrophages are essential effectors of the innate immune system. These cells provide innate immune defenses and regulate tissue and organ homeostasis. In addition to their roles in diseases such as cancer, obesity and osteoarthritis, they play vital roles in tissue repair and disease rehabilitation. Macrophages and other inflammatory cells are recruited to tissue injury sites where they promote changes in the microenvironment. Among the inflammatory cell types, only macrophages have both pro-inflammatory (Ml) and anti-inflammatory (M2) actions, and M2 macrophages have four subtypes. The co-action of Ml and M2 subtypes can create a favorable microenvironment, releasing cytokines for damaged tissue repair. In this review, we discuss the activation of macrophages and their roles in severe peripheral nerve injury. | 2 | 大黄酚与转化生长因子-β1对体外培养人成纤维细胞胶原及盘状结构域受体2的调控作用及机制显示文摘目的探讨中药大黄提取物大黄酚和转化生长因子(TGF)-β1对体外培养人成纤维细胞Ⅰ型(Col-Ⅰ)、Ⅲ型胶原(Col-Ⅲ)、盘状结构域受体(DDR)2的调控作用及机制。方法体外培养人成纤维细胞,用大黄酚干扰24 h,观察细胞生长特征,并用实时定量聚合酶链反应(qRT-PCR)、Western印迹检测Col-Ⅰ、Col-Ⅲ、DDR2表达,流式细胞技术检测成纤维细胞细胞周期。细胞分为对照组、TGF组和大黄酚组。结果TGF-β1、大黄酚上调DDR2表达,与对照组有显著性差异(P<0.05)。TGF-β1组Col-Ⅰ表达上调,与对照组有显著性差异(P<0.05);大黄酚组Col-Ⅰ表达下调,与对照组有显著性差异(P<0.05)。TGF-β1组Col-Ⅲ表达与对照组无显著性差异(P>0.05);大黄酚组Col-Ⅲ表达上调,与对照组有显著性差异(P<0.05)。G1/G0期细胞比例,TGF-β1组与对照组无显著性差异(P>0.05),大黄酚组低于对照组,有显著性差异(P<0.05);S期细胞比例,TGF-β1组与对照组无显著性差异(P>0.05),大黄酚组高于对照组,有显著性差异(P<0.05);G2期细胞比例,各组间无明显差异(P>0.05)。结论大黄酚和TGF-β1对培养成纤维细胞的DDR2、Ⅰ型和Ⅲ型胶原蛋白表达具有调控作用,并能影响其细胞周期。TGF-β1作用是通过上调Col-Ⅰ表达促进组织修复,而大黄酚作用的机制可能是降低了G1/G0期细胞比例而增加细胞增殖活性来促进组织修复,同时上调DDR2,增加Col-Ⅰ降解,减少瘢痕形成。 | 杨成华 巫岳龙 李敏 程基焱 颜丽萍 | 2020 | 中国老年学杂志2020，40，18： | 4 |
