| 1 | 电针对骶上脊髓损伤后神经源性膀胱大鼠尿流动力学及髓内凋亡,神经营养因子双向调控作用的影响显示文摘Objective:To observe the effects of electroacupuncture on urodynamics and intramedullary apoptosis related factor B cell lymphoma gene-2(Bcl-2),Bcl-2 related protein X(Bax)and brain-derived neurotrophic factor(BDNF)in neurogenic bladder rats after supersacral spinal cord injury.Methods:A total of 60 female SD rats,SPF grade,were randomly selected,12 rats in the blank group and 12 rats in the sham operation group,and the remaining 36 rats were made models.Of these 36 rats,24 rats meeting the model standard were randomized into the model group and electroacupuncture group,with 12 rats in each group.The neurogenic bladder models after supracacral spinal cord injury were prepared by the Hassan Shaker spinal cord transection method,via combining the previous model study experience.At the end of spinal shock stage,'Dazhui(大椎GV14)''Zhongji(中极CV 3)''Ciliao(次髎BL 32)',and'Sanyinjiao(三阴交SP 6)'were selected for electroacupuncture.Observation on the histomorphological changes of bladder detrusor and spinal cord in rats was conducted by HE staining.The apoptosis rate of spinal cord cells was measured by TUNEL assay.The maximum bladder volume and compliance were measured by urodynamics test.The protein expressions of Bcl-2,Bax,and BDNF in spinal cord tissues were measured by Western blot assay.The expressions of Bcl-2,Bax,and BDNF mRNA were measured by RT-PCR assay.Results:(1)The results of HE staining showed that compared with the blank group and sham operation group,in the model group the bladder epithelial cell structure was destroyed,the detrusor muscle fibers proliferated,and there were severe bleeding changes,the spinal cord tissue was disordered,the structure was destroyed,the cell space was enlarged,and accompanied by a large number of inflammatory cell infiltration and cystic cavity formation.Compared with the model group,in the electroacupuncture group,the intact bladder epithelial cells increased,the detrusor muscle fiber proliferation decreased,the bleeding changes decreased,the complete structural area of spinal cord tissues increased,the cell space decreased,the levels were clearer,and the cystic cavity decreased.(2)The results of TUNEL assay indicated that the TUNEL positive rate of spinal cord cells in the model group was significantly higher than that in the blank group and sham group(both P<0.05).Compared with the model group,the TUNEL positive rate of spinal cord cells in the electroacupuncture group was significantly reduced(P<0.05).(3)The results of urodynamics test suggested that compared with the blank group and sham operation group,the maximum bladder volume and compliance of the model group were significantly reduced(all P<0.05).Compared with the model group,the maximum bladder volume and compliance were significantly increased in the electroacupuncture group(both P<0.05).(4)The results of Western blot assay indicated that,compared with the blank group and sham operation group,in the model group the protein contents of Bcl-2 and BDNF were significantly reduced(all P<0.05),while the protein content of Bax in the model group was significantly increased(both P<0.05).(5)The results of RT-PCR assay indicated that,compared with blank group and sham operation group,in the model group the intramedullary Bcl-2 and BDNF mRNA were significantly reduced(all P<0.05),and Bax mRNA was significantly increased(both P<0.05).Conclusion:Electroacupuncture at'GV14''CV3''BL32',and'SP6'can promote the repair of bladder detrusor,improve the maximum bladder volume and compliance of bladder,promote morphological repair of bladder detrusor,reduce spinal cord cell apoptosis rate,thus to restore the function of bladder.This is maybe related with the electroacupuncture’s two-way regulatory mechanism of increased expression of protein and gene of Bcl-2 and BDNF and decreased expression of protein and gene of Bax in spinal cord tissues. | 刘笑萌 许明 张泓 艾坤 邓石峰 余雨荷 | 2021 | World Journal of Acupuncture-Moxibustion2021,31,2: | 8 |
| 2 | BDNF在脊髓损伤修复中的双重作用及其表观遗传调节因素显示文摘脑源性神经营养因子(brain—derived neurotrophic factor,BDNF)是中枢内神经营养因子家族的重要一员,与神经生长因子(nerve growth factor,NGF)和神经营养素(neurotrophin-3/4,NT3/4)等一起发挥神经元保护作用,不仅作用于发育中神经元分化、轴突生长和突触形成,而且参与成体神经元突起修剪、损伤轴突再生和突触可塑性活动。成体中枢神经系统内BDNF等神经营养因子含量显著减少,神经保护功能降低,给予外源性神经营养因子有助于改善损伤后神经再生环境。因此,BDNF的合成和降解及其关键调节机制,对于维持其正常含量和保护功能至关重要。 | 姚欣怡 王玉玲 郭彦谷 陈孛玉 卞干兰 陈良为 | 2018 | 神经解剖学杂志2018,34,6: | 6 |
| 3 | 负载脑源性神经营养因子胶原/肝素硫酸支架促进颅脑创伤大鼠神经运动功能的恢复显示文摘背景:研究表明胶原和硫酸肝素交联后可有效固定生长因子,显著改善脊髓损伤大鼠的神经运动功能恢复。目的:观察负载脑源性生长因子胶原/硫酸肝素支架移植修复颅脑创伤的效果。方法:制作胶原支架、胶原/硫酸肝素支架与负载脑源性神经营养因子的胶原/硫酸肝素支架,检测胶原支架、胶原/硫酸肝素支架的孔隙率、吸水率、压缩模量和压缩应力;检测载脑源性生长因子胶原/硫酸肝素支架的体外缓释性能;检测胶原/硫酸肝素支架与负载脑源性神经营养因子的胶原/硫酸肝素支架的细胞毒性与细胞相容性。将48只SD大鼠随机分4组干预:对照组开骨窗后缝合,空腔组仅制作颅脑创伤空腔模型,支架组、生长因子支架组颅脑创伤后空腔处分别植入胶原/硫酸肝素支架、负载脑源性神经营养因子的胶原/硫酸肝素支架,术后分别进行改良神经功能缺损评分、水迷宫测试与脑损伤形态学观察。动物实验获得武警特色医学中心伦理委员会批准。结果与结论:①胶原/硫酸肝素支架的孔隙率、压缩模量和压缩应力高于胶原支架(P<0.05),吸水率低于胶原支架(P<0.05);②胶原/硫酸肝素支架无细胞毒性,负载脑源性神经营养因子后更加有利于脑微血管内皮细胞的增殖;脑微血管内皮细胞在负载脑源性神经营养因子支架微孔内生长良好,分布均匀;③形态学观察显示,两支架组损伤灶处可见大量新生神经细胞及神经纤维,其中以生长因子支架组的新生神经细胞数量及神经纤维密度更高;④生长因子支架组大鼠的逃逸潜伏期短于空腔组、支架组(P<0.01,P<0.05),象限停留时间和平台穿越次数高于支架组、空腔组(P<0.01,P<0.05);⑤生长因子支架组术后3-7周的改良神经功能缺损评分低于支架组、空腔组(P<0.05,P<0.01);⑥结果表明,负载脑源性神经营养因子胶原/硫酸肝素支架移植可以促进大鼠颅脑创伤后神经运动功能的恢复。 | 张健 陈淼 李伟鑫 叶益超 徐会友 马珂 陈旭义 孙洪涛 张赛 | 2020 | 中国组织工程研究2020,24,34: | 4 |
| 4 | MRI及生物标志物对创伤性颅脑损伤患者预后的评估显示文摘目的探讨MRI成像数据及生物标志物检测对创伤性颅脑损伤患者预后的评估价值。方法选取2018年4月~2019年4月我院确诊的76例创伤性颅脑损伤(TBI)患者为研究对象,根据MRI检查情况将患者分为3组,MRI阴性组(n=30),创伤性出血组(TH组,n=26)及创伤性微血管损伤组(TVI组,n=20)。采用酶联免疫吸附测定测量分别测量血浆中vWF、cFn、pNF-H水平,根据病变的数量、大小和位置对GRE、FLAIR、DWI和SWI阳性结果进行扫描评分,采用格拉斯哥预后评分(GOS)评估患者的临床预后。结果MRI阴性组GOS评分>3分患者显著多于TH组、TVI组患者(P<0.05)。损伤后≤48 h,TVI组患者vWF水平较MRI阴性组、TH组显著降低,pNF-H水平较MRI阴性组、TH组显著升高(均P<0.05)。损伤后>48 h,TH组患者vWF水平较MRI阴性组、TVI组显著升高(P<0.05);TVI组患者cFn、pNF-H水平较MRI阴性组、TH组显著升高(P<0.05)。Logistic回归Hosmer-Lemeshow拟合优度检验结果显示,当将损伤后<48 h测量的vWF、cFn和pNF-H水平添加到预测模型中时,预测预后良好的准确性从57%~60%增加到77%~81%。结论TBI患者损伤后血浆中vWF、cFn等水平变化显著,MRI成像数据及生物标志物检测对创伤性颅脑损伤患者预后具有较好的评估价值。 | 王静 李建蓉 杨染 刘国庆 温云 | 2020 | 西部医学2020,32,4: | 1 |
