1Nobiletin and tangeretin(citrus polymethoxyflavones): an overview on their chemistry, pharmacology and cytotoxic activities against breast cancer显示文摘The fruit peel of Citrus species(Chenpi),particularly those of mandarin oranges,is a useful source of food and medicine in China.Flavonoids from the citrus fruit peel are mainly polymethoxyflavones(PMFs),of which nobiletin and tangeretin are the most abundant components.In the present review,we summarized the cytotoxic activities of these two PMFs to breast cancer cells.Studies have reported that these two compounds inhibit the growth of breast cancer cells by inducing apoptosis,cytostatic cell death and cell cycle arrest,or by inhibiting cell proliferation,metastasis and tumour angiogenesis,depending on the molecular subtypes of breast cancer.In vitro and in vivo cytotoxic activities involve different molecular targets and signalling pathways.Analyses on the structure-activity relationship(SAR)of nobiletin and tangeretin have shown that the presence of a methoxy group at C8 and a hydroxyl group at C3 or C5 are essential for anti-proliferative activity.Some future perspectives and research needs are suggested.Sources of information are from Pub Med,Pub Med Central,Science Direct,Google Scholar,J-Stage,Pub Chem and CNKI using keyword search.Eric Wei Chiang Chan Oi Yoon Michelle Soo Yong Hui Tan Siu Kuin Wong Hung Tuck Chan 2020Journal of Chinese Pharmaceutical Sciences2020,29,7:0
2Insulin receptor is implicated in triple-negative breast cancer by decreasing cell mobility显示文摘Triple-negative breast cancer(TNBC)has a poor prognosis and typically earlier onset of metastasis in comparison with other breast cancer subtypes.It has been reported that insulin receptor(INSR)is downregulated in TNBC,however,its clinical significance and functions in TNBC remain to be elucidated.In this study,we found that INSR expression was significantly downregulated in TNBC,and overexpression of INSR suppressed cell migration and invasion in TNBC.In addition,the survival rate of breast cancer patients with low INSR expression was lower than that of patients with high INSR expression.INSR expression was significantly correlated with lymph node metastasis,clinical tumor stages,ER status,PR status,and the proliferation index Ki-67 expression.In summary,our study suggests that INSR may serve as a biomarker for breast cancer prognosis and it may be a potential target for TNBC treatment.Ying Yang Xiang Chen Changyan Ma 2021The Journal of Biomedical Research2021,35,3:0
3α-ketoglutarate promotes the specialization of primordial germ cell-like cells through regulating epigenetic reprogramming显示文摘There is growing evidence that cellular metabolism can directly participate in epigenetic dynamics and consequently modulate gene expression.However,the role of metabolites in activating the key gene regulatory network for specialization of germ cell lineage remains largely unknown.Here,we identified some cellular metabolites with significant changes by untargeted metabolomics between mouse epiblast-like cells(EpiLCs)and primordial germ cell-like cells(PGCLCs).More importantly,we found that inhibition of glutaminolysis by bis-2-(5-phenylacetamido-1,3,4-thiadiazol-2-yl)ethyl sulfide(BPTES)impeded PGCLC specialization,but the impediment could be rescued by addition ofα-ketoglutarate(αKG),the intermediate metabolite of oxidative phosphorylation and glutaminolysis.Moreover,adding aKG alone to the PGCLC medium accelerated the PGCLC specialization through promoting H3 K27 me3 demethylation.Thus,our study reveals the importance of metabolite aKG in the germ cell fate determination and highlights the essential role of cellular metabolism in shaping the cell identities through epigenetic events.Ming Xing Na Wang Hanyi Zeng Jun Zhang 2021The Journal of Biomedical Research2021,35,1:0
