2肿瘤炎症微环境对小鼠结肠癌cell-in-cell的影响研究显示文摘目的免疫细胞与肿瘤细胞的cell-in-cell常见于结肠癌组织内,被认为是肿瘤免疫抵抗的一种特殊方式,研究表明IL-6可以促进该结构的形成,这里我们进一步利用动物模型的了解在体内炎症对结肠癌cell-in-cell的影响。方法我们建立了小鼠DSS结肠炎模型、小鼠AOM结肠癌模型以及小鼠AOM+DSS结肠炎性结肠癌模型,比较cell-in-cell的发生频率以及内部免疫细胞种类与命运。结果结肠炎性结肠癌组织内高水平的IL-6/TNF-α伴随高频率的cell-in-cell结构,进入肿瘤细胞内部的免疫细胞以CD8 T细胞为主,其余为CD4 T细胞和中性粒细胞为主;进入的免疫细胞发生缺氧,呈现cleaved caspase 3阳性的细胞死亡,并且AOM+DSS组的cell-in-cell内部免疫细胞死亡相对AOM组增加,这可能与该组结肠癌组织内高IL-6/TNF-α水平增加了肿瘤微环境缺氧有关。结论该研究证实了结肠癌组织的炎症微环境促进免疫细胞进入肿瘤细胞内并发生死亡,可能是肿瘤细胞反攻免疫系统的一种特殊方式,也是炎症微环境促进肿瘤免疫逃逸的新证据。王珊 周亚光 陶爱林 2019国际医药卫生导报2019,25,20:1
3Molecular mechanisms underlying cell-in-cell formation:core machineries and beyond显示文摘Although cell-in-cell structures(CICs),with one or more cells present inside another cell,had been identified for a century,it was not until recent years that scientists started to uncover their pivotal roles in multiple biological processes,primarily via mediating the death of internalized cells.Meanwhile,considerable progresses were made on deciphering the mechanisms underlying their formation based on different models.Entosis was one of the best investigated CIC models,where cell internalization was coordinately driven by adherens junction and contractile ac-tomyosin,the two spatially polarized and complementary core elements that were coupled by mechanical ring,a recently identified core element.Meanwhile,an expanding group of factors were found capable of regulating CIC formation by targeting these core elements.The elucidation of the molecular machinery controlling CIC formation enables synthetic engineering of cells used for clinical and research purposes。Zubiao Niu Meifang He Qiang Sun 2021Journal of Molecular Cell Biology2021,13,5:1
